In this study, we investigated the microbially mediated transformation of labile Synechococcus-derived DOM to RDOM using a 60-day experimental incubation system. Three phases of TOC degradation activity (I, II and III) were observed following the addition of Synechococcus-derived DOM. The phases were characterized by organic carbon consumption rates of 8.77, 1.26 and 0.16 μmol L−1 day−1, respectively. Excitation emission matrix analysis revealed the presence of three FDOM components including tyrosine-like, fulvic acid-like, and humic-like molecules. The three components also exhibited differing biological availabilities that could be considered as labile DOM (LDOM), semi-labile DOM (SLDOM) and RDOM, respectively. DOM molecular composition was also evaluated using FT-ICR MS. Based on differing biological turnover rates and normalized intensity values, a total of 1704 formulas were identified as candidate LDOM, SLDOM and RDOM molecules. Microbial transformation of LDOM to RDOM tended to proceed from high to low molecular weight, as well as from molecules with high to low double bond equivalent (DBE) values. Relatively higher aromaticity was observed in the formulas of RDOM molecules relative to those of LDOM molecules. FDOM components provide valuable proxy information to investigate variation in the bioavailability of DOM. These results suggest that coordinating fluorescence spectroscopy and FT-ICR MS of DOM, as conducted here, is an effective strategy to identify and characterize LDOM, SLDOM and RDOM molecules in incubation experiments emulating natural systems. The results described here provide greater insight into the metabolism of phytoplankton photosynthate by heterotrophic bacteria in marine environments.  相似文献

Brucella melitensis infection causes acute necrotizing inflammation in pregnant animals; however, the pathophysiological mechanisms leading to placentitis are unknown. Here, we demonstrate that high‐mobility group box 1 (HMGB1) acts as a mediator of placenta inflammation in Bmelitensis‐infected pregnant mice model. HMGB1 levels were increased in trophoblasts or placental explant during B. melitensis infection. Inhibition of HMGB1 activity with neutralising antibody significantly reduced the secretion of inflammatory cytokines in B. melitensis‐infected trophoblasts or placenta, whereas administration of recombinant HMGB1 (rHMGB1) increased the inflammatory response. Mechanistically, this decreased inflammatory response results from inhibition of HMGB1 activity, which cause the suppression of both mitogen‐activated protein kinases and nuclear factor kappa‐light‐chain‐enhancer of activated B cells (NF‐κB) activation. Moreover, neutralising antibody to HMGB1 prevented B. melitensis infection‐induced activation of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase in trophoblasts. In contrast, in vitro stimulation of trophoblasts with rHMGB1 caused activation of NADPH oxidase and increased the production of ROS, which contributes to high bacterial burden within trophoblasts or placenta. In vivo, treatment with anti‐HMGB1 antibody increases the number of Brucella survival within placenta in B. melitensis‐infected pregnant mice but successfully reduced the severity of placentitis and abortion.  相似文献

The soil-borne vascular pathogen Verticillium dahliae infects many dicotyledonous plants to cause devastating wilt diseases. During colonization, V. dahliae spores develop hyphae surrounding the roots. Only a few hyphae that adhere tightly to the root surface form hyphopodia at the infection site, which further differentiate into penetration pegs to facilitate infection. The molecular mechanisms controlling hyphopodium formation in V. dahliae remain unclear. Here, we uncovered a cellophane surface-induced gene (VdCSIN1) as a regulator of V. dahliae hyphopodium formation and pathogenesis. Deletion of VdCSIN1 compromises hyphopodium formation, hyphal development and pathogenesis. Exogenous application of cyclic adenosine monophosphate (cAMP) degradation inhibitor or disruption of the cAMP phosphodiesterase gene (VdPDEH) partially restores hyphopodium formation in the VdΔcsin1 mutant. Moreover, deletion of VdPDEH partially restores the pathogenesis of the VdΔcsin1 mutant. These findings indicate that VdCSIN1 regulates hyphopodium formation via cAMP-mediated signalling to promote host colonization by V. dahliae.  相似文献

Xanthomonas campestris pv. campestris (Xcc), the causal agent of black rot in crucifers, produces a membrane-bound yellow pigment called xanthomonadin to protect against photobiological and peroxidative damage, and uses a quorum-sensing mechanism mediated by the diffusible signal factor (DSF) family signals to regulate virulence factors production. The Xcc gene XCC4003, annotated as Xcc fabG3, is located in the pig cluster, which may be responsible for xanthomonadin synthesis. We report that fabG3 expression restored the growth of the Escherichia coli fabG temperature-sensitive mutant CL104 under non-permissive conditions. In vitro assays demonstrated that FabG3 catalyses the reduction of 3-oxoacyl-acyl carrier protein (ACP) intermediates in fatty acid synthetic reactions, although FabG3 had a lower activity than FabG1. Moreover, the fabG3 deletion did not affect growth or fatty acid composition. These results indicate that Xcc fabG3 encodes a 3-oxoacyl-ACP reductase, but is not essential for growth or fatty acid synthesis. However, the Xcc fabG3 knock-out mutant abolished xanthomonadin production, which could be only restored by wild-type fabG3, but not by other 3-oxoacyl-ACP reductase-encoding genes, indicating that Xcc FabG3 is specifically involved in xanthomonadin biosynthesis. Additionally, our study also shows that the Xcc fabG3-disrupted mutant affects Xcc virulence in host plants.  相似文献

甘肃河西走廊经过60多年的防沙治沙,在绿洲边缘形成了一条积沙带。其中,民勤绿洲边缘积沙带相对最典型、最完整。积沙带上的植被不仅是积沙带的形成因素,更是维持积沙带稳定的重要因素。从一定意义上说,积沙带上植被的健康状况,决定着积沙带的稳定性。为了分析积沙带上的植被状况,在民勤绿洲边缘积沙带上等间距设定了11条样线,通过60多个样方的植被调查,对民勤绿洲边缘积沙带上植物群落进行了健康序列分析。结果表明:民勤绿洲边缘积沙带上的植被普遍是不健康的,民勤绿洲边缘人工固沙林积沙带上的植被健康状况相对好于天然灌丛积沙带上的植被健康状况,乔灌混交林积沙带上的植被健康状况最差。民勤绿洲下游积沙带上的优势种植物柽柳严重枯死,健康状况最差,积沙带潜在风蚀活化的危险。农田弃耕后,沙区农田边缘防护林及其积沙带的保护是一个新的值得考虑的问题。  相似文献

以新疆阿克苏河流域为研究区,以GF-1 WFV和Landsat8 OLI两种高分辨率遥感影像为数据源,结合102个不同深度层的土壤湿度实测数据,选择垂直干旱指数(PDI)、改进型垂直干旱指数(MPDI)和植被调整垂直干旱指数(VAPDI),对土壤湿度指数反演的效果进行比较和验证。结果表明,两种数据源下的PDI、MPDI、VAPDI与土壤湿度实测含水量的决定系数较高,尤其是0—10 cm的相关性最强,平均决定系数达到0.68,说明基于光学遥感影像近红外和红光波段反射率构建的反演指数对近地表层土壤湿度信息更敏感,但对地下较深层次的土壤湿度反演精度略低;MPDI和VAPDI能够在一定程度上克服混合像元对土壤湿度光谱信息的影响,反演的精度要比PDI高,尤其是高植被覆盖度区,采用垂直植被指数(PVI)修正的VAPDI反演精度最佳;基于两种遥感数据源的土壤湿度空间异质性基本一致,但空间分辨率较高的GF-1 WFV模拟的土壤湿度空间分异更加精细和明显。研究结果可为"一带一路"背景下干旱半干旱地区大范围和动态监测土壤湿度、开展定量节水灌溉等提供理论基础和实践参考。  相似文献

气候变化和人类活动是对陆地生态系统碳循环产生重要影响的两个因素,定量评估气候变化与人类活动对植被净初级生产力(NPP)的相对影响,对深入理解其驱动机制和控制荒漠化发展具有重要意义。以疏勒河流域为研究区,利用遥感和气象数据计算潜在NPP(PNPP)及其与实际NPP(ANPP)之间的差值,分别衡量了气候变化和人类活动对流域NPP的相对影响。研究结果表明:(1)2001—2015年疏勒河流域年ANPP整体呈缓慢增加趋势,与全国和西北地区相比,普遍较低,流域植被整体生产力水平不高。流域年ANPP空间分布呈现上游祁连山区和中下游绿洲区ANPP较高,而中下游荒漠戈壁区ANPP较低的分布格局。(2)2001—2015年流域年PNPP的变化趋势表明,降水量的变化是导致疏勒河流域植被退化加剧或缓解的主要气候驱动因素,但气温的变化对植被的影响较为复杂。(3)2001—2015年流域大部分地区植被退化系人类活动造成的,但人类活动的负向影响力在减弱。(4)气候变化和人类活动对植被NPP的相对影响均表现出明显的空间异质性,其中人类活动是疏勒河流域植被变化的主要驱动因素。  相似文献

场镇发展是西南山区城镇发展的重要模式,且大部分场镇沿河分布,快速城镇发展给河流水环境及生物地化过程带来了一系列影响,然而其对河流温室气体排放时空格局的影响及机制尚不清楚。选择流域场镇发展特征明显的黑水滩河为研究对象,于2014年9月、12月、2015年3月、6月,对流域内干、支流水体温室气体浓度及扩散通量进行分析,旨在阐明流域场镇式发展下河流温室气体排放时空特征及关键驱动因素。研究结果表明,黑水滩河干、支流水体年均二氧化碳分压(pCO_2)及甲烷(CH_4)、一氧化二氮(N_2O)浓度均处于过饱和状态,是大气温室气体的净排放源;流域内干、支流水体流经不同场镇区前后水体碳、氮、磷及叶绿素a含量均不同程度增加,从上游向下游呈现明显的污染累积;水体溶存pCO_2\\CH_4\\N_2O浓度及扩散通量在不同场镇前后也呈现显著增加的趋势,三种温室气体扩散通量平均增幅分别为25.88%、55.22%、99.64%;河流水体pCO_2与N_2O浓度及通量秋季高于其他季节,CH_4浓度及扩散通量春季最高,秋季次之,夏、冬季最低,温室气体浓度及排放的季节变化主要受温度和降雨格局共同影响。相关分析表明,pCO_2与水温和pH关系密切,而水体CH_4和N_2O浓度与水体碳、氮、磷等生源要素均呈显著的正相关关系,水体CH_4与N_2O浓度对生源要素输入极为敏感,流域场镇发展带来的河流污染负荷的增加可能对水体CH_4与N_2O排放产生明显的激发效应。本研究认为,山区河流流域内沿河串珠状场镇分布对河流水体生源要素及其他理化性质产生累积影响,进而改变了水体温室气体的产生与排放时空格局。  相似文献

PAEs have been proved to be one of the major organic pollutants. The present study determined the level of PAEs using surface water samples from the upper reaches of the Yangtze River. The results showed that DEHP contributed the most to PAEs pollution, followed by DBP. Risk Quotients of PAEs were used for preliminary screening, and DEHP and BBP were identified as potential risk factors with RQ of 310 and 70.7. The MOS10 of DEHP and BBP were 1.40 and 1.32×105, respectively, indicating that BBP may pose little risk to aquatic organisms. The potential risk of DEHP was further analyzed with joint probabilistic curves. Among different biological groups, fish and zoobenthos were two major groups sensitive to DEHP-induced damage under current concentrations with 99.4% and 98.3% for 5% species of each group. While among the toxicity endpoints, reproduction may be more sensitive than others. The 5% and 10% aquatic species were suffered from reproductive damages by 100% and 97.8%, respectively. Overall, our results indicated that DEHP in the surface water of the upper Yangtze River may pose potential risk to aquatic organisms, especially on their reproduction. We generally reviewed the distributions and potential risks of heavy metals, polycyclic aromatic hydrocarbons (PAHs) and phthalic acid esters (PAEs) in the upper reaches of the Yangtze River based on recent studies. The distribution of heavy metals varied significantly with locations and types of mediums, and contents of some metals exceeded the limits in food. The contents of PAHs in the upper reaches of the Yangtze River were in line with those in the lower reaches. Low molecular weight PAHs were predominant in surface water, while high molecular weight PAHs were predominant in sediments, indicating low molecular PAHs may pose greater risks to aquatic ecosystems. DEHP and DBP were the predominant monomers of PAEs in surface water and sediments, and BBP and DBP ranked first and second respectively in fish bodies. The results of risk assessment also indicated that monomers such as DBP and DnBP had greater risk than DEHP. Over all, heavy metals and organic pollutants were widely distributed in the aquatic environment in the upper reaches of the Yangtze River, which may pose potential risks to the ecosystem. © 2019, Institute of Hydrobiology, Chinese Academy of Sciences.
