通过盆栽试验,模拟蓝桉(Eucalyptus globulus)凋落叶在土壤中分解对受体植物菠菜(Spinacia oleracea)生长及光合特性的影响,设置L30(30 g/盆)、L60(60 g/盆)、L90(90 g/盆)和L120(120 g/盆)4个凋落叶施用水平,对照(CK)不加凋落叶。结果表明:(1)在分解30 d时,低量的凋落叶(L30)未抑制菠菜生物量的积累,而中量和高量处理(L60、L90和L120)下其生物量显著降低(P0.05);在凋落叶分解50 d后,蓝桉凋落叶的化感抑制效应已经减弱,仅L120仍然抑制菠菜生物量的积累;(2)一定量的凋落叶(L30和L60)对菠菜叶绿素的合成有促进作用,而凋落叶超过一定量时(L120)转为抑制作用,在凋落叶分解28 d时相对不明显,40 d后逐渐显现;(3)经凋落叶处理的菠菜叶片胞间CO2浓度(Ci)、气孔导度(Gs)及蒸腾速率(Tr)均显著高于对照(CK)(P0.05),而净光合速率(Pn)在L30处理下最高,L60和L90处理与CK差异不显著,仅L120处理显著低于CK(P0.05);(4)光响应与CO2响应曲线的特征参数表观量子效率(AQY)、最大净光合速率(Pmax)、光饱和点(Lsp)、光补偿点(Lcp)、暗呼吸速率(Rd)、Ru BP表观羧化效率(CE)和光呼吸速率(Rp)在L30处理下高于CK,其余凋落叶处理均低于CK,而CO2饱和点(Csp)、CO2补偿点(Ccp)随凋落叶量的增加而升高。综合各指标可见,少量的蓝桉凋落叶分解(L30)可通过增加叶绿素含量,提高对光和CO2的利用能力促进菠菜的光合作用,进而促进其生长,而凋落叶超过一定量时(L90—L120)则起到相反的作用。  相似文献

Soil salinity acts as a critical environmental filter on microbial communities, but the consequences for microbial diversity and biogeochemical processes are poorly understood. Here, we characterized soil bacterial communities and microbial functional genes in a coastal estuarine wetland ecosystem across a gradient (~5 km) ranging from oligohaline to hypersaline habitats by applying the PCR-amplified 16S rRNA (rRNA) genes sequencing and microarray-based GeoChip 5.0 respectively. Results showed that saline soils in marine intertidal and supratidal zone exhibited higher bacterial richness and Faith's phylogenetic diversity than that in the freshwater-affected habitats. The relative abundance of taxa assigned to Gammaproteobacteria, Bacteroidetes and Firmicutes was higher with increasing salinity, while those affiliated with Acidobacteria, Chloroflexi and Cyanobacteria were more prevalent in wetland soils with low salinity. The phylogenetic inferences demonstrated the deterministic role of salinity filtering on the bacterial community assembly processes. The abundance of most functional genes involved in carbon degradation and nitrogen cycling correlated negatively with salinity, except for the hzo gene, suggesting a critical role of the anammox process in tidal affected zones. Overall, the salinity filtering effect shapes the soil bacterial community composition, and soil salinity act as a critical inhibitor in the soil biogeochemical processes in estuary ecosystems.  相似文献

Tissue vascularization is critical to enable oxygen and nutrient supply. Therefore, establishing expedient vasculature is necessary for the survival of tissue after transplantation. The use of biomechanical forces, such as cell-induced traction forces, may be a promising method to encourage growth of the vascular network. Three-dimensional (3D) bioprinting, which offers unprecedented versatility through precise control over spatial distribution and structure of tissue constructs, can be used to generate capillary-like structures in vitro that would mimic microvessels. This study aimed to develop an in vitro, 3D bioprinted tissue model to study the effect of cellular forces on the spatial organization of vascular structures and tissue maturation. The developed in vitro model consists of a 3D bioprinted polycaprolactone (PCL) frame with a gelatin spacer hydrogel layer and a gelatin–fibrin–hyaluronic acid hydrogel layer containing normal human dermal fibroblasts and human umbilical vein endothelial cells printed as vessel lines on top. The formation of vessel-like networks and vessel lumens in the 3D bioprinted in vitro model was assessed at different fibrinogen concentrations with and without inhibitors of cell-mediated traction forces. Constructs containing 5 mg/ml fibrinogen had longer vessels compared to the other concentrations of fibrinogen used. Also, for all concentrations of fibrinogen used, most of the vessel-like structures grew parallel to the direction the PCL frame-mediated tensile forces, with very few branching structures observed. Treatment of the 3D bioprinted constructs with traction inhibitors resulted in a significant reduction in length of vessel-like networks. The 3D bioprinted constructs also had better lumen formation, increased collagen deposition, more elaborate actin networks, and well-aligned matrix fibers due to the increased cell-mediated traction forces present compared to the non-anchored, floating control constructs. This study showed that cell traction forces from the actomyosin complex are critical for vascular network assembly in 3D bioprinted tissue. Strategies involving the use of cell-mediated traction forces may be promising for the development of bioprinting approaches for fabrication of vascularized tissue constructs.  相似文献

红外相机技术在野生动物调查研究中得到广泛应用, 其发展和普及为中国自然保护地生物多样性保护带来了诸多机会。为进一步推广该技术在我国自然保护地野生动物监测中的应用, 中国林业科学研究院森林生态环境与保护研究所在自然保护区生物标本资源共享子平台增设野生动物红外相机数据库专栏, 并通过门户网站“中国自然保护区生物标本资源共享平台” (http://www.papc.cn/)对外发布, 向公众开放, 可以随时查阅、下载信息。本文重点介绍了该平台的野生动物红外相机数据库建设进展, 包括所监测的自然保护区、监测方案、数据规范存储、标准化分析、成果介绍、工作计划等内容。2010-2019年间, 该平台使用红外相机调查技术在全国13个不同类型的保护地开展兽类和鸟类资源调查, 累计27.25万个有效工作日, 共拍摄到8.41万张独立图像, 经鉴定有80种野生兽类和200种野生鸟类, 并利用这些数据对野生动物行为、稀有物种探测、人为干扰, 以及气候变化影响等方面进行研究, 取得了重要的阶段性成果。下一步, 子平台牵头单位将筹建自然保护地红外相机数据共享平台, 制定红外相机调查设计、监测技术、数据格式等统一标准化方案, 逐步完善我国自然保护地野生动物红外相机监测网络。  相似文献

Acute Respiratory Distress Syndrome (ARDS) is a clinical syndrome characterized by diffuse alveolar damage usually secondary to an intense host inflammatory response of the lung to a pulmonary or extrapulmonary infectious or non-infectious insult often leading to the development of intra-alveolar and interstitial fibrosis. Curcumin, the principal curcumoid of the popular Indian spice turmeric, has been demonstrated as an anti-oxidant and anti-inflammatory agent in a broad spectrum of diseases. Using our well-established model of reovirus 1/L-induced acute viral pneumonia, which displays many of the characteristics of the human ALI/ARDS, we evaluated the anti-inflammatory and anti-fibrotic effects of curcumin. Female CBA/J mice were treated with curcumin (50 mg/kg) 5 days prior to intranasal inoculation with 107 pfu reovirus 1/L and daily, thereafter. Mice were evaluated for key features associated with ALI/ARDS. Administration of curcumin significantly modulated inflammation and fibrosis, as revealed by histological and biochemical analysis. The expression of IL-6, IL-10, IFNγ, and MCP-1, key chemokines/cytokines implicated in the development of ALI/ARDS, from both the inflammatory infiltrate and whole lung tissue were modulated by curcumin potentially through a reduction in the phosphorylated form of NFκB p65. While the expression of TGFß1 was not modulated by curcumin, TGFß Receptor II, which is required for TGFß signaling, was significantly reduced. In addition, curcumin also significantly inhibited the expression of α-smooth muscle actin and Tenascin-C, key markers of myofibroblast activation. This data strongly supports a role for curcumin in modulating the pathogenesis of viral-induced ALI/ARDS in a pre-clinical model potentially manifested through the alteration of inflammation and myofibroblast differentiation.  相似文献

Background

To overcome the limitations of conventional diffusion tensor magnetic resonance imaging resulting from the assumption of a Gaussian diffusion model for characterizing voxels containing multiple axonal orientations, Shannon''s entropy was employed to evaluate white matter structure in human brain and in brain remodeling after traumatic brain injury (TBI) in a rat.

Methods

Thirteen healthy subjects were investigated using a Q-ball based DTI data sampling scheme. FA and entropy values were measured in white matter bundles, white matter fiber crossing areas, different gray matter (GM) regions and cerebrospinal fluid (CSF). Axonal densities'' from the same regions of interest (ROIs) were evaluated in Bielschowsky and Luxol fast blue stained autopsy (n = 30) brain sections by light microscopy. As a case demonstration, a Wistar rat subjected to TBI and treated with bone marrow stromal cells (MSC) 1 week after TBI was employed to illustrate the superior ability of entropy over FA in detecting reorganized crossing axonal bundles as confirmed by histological analysis with Bielschowsky and Luxol fast blue staining.

Results

Unlike FA, entropy was less affected by axonal orientation and more affected by axonal density. A significant agreement (r = 0.91) was detected between entropy values from in vivo human brain and histologically measured axonal density from post mortum from the same brain structures. The MSC treated TBI rat demonstrated that the entropy approach is superior to FA in detecting axonal remodeling after injury. Compared with FA, entropy detected new axonal remodeling regions with crossing axons, confirmed with immunohistological staining.

Conclusions

Entropy measurement is more effective in distinguishing axonal remodeling after injury, when compared with FA. Entropy is also more sensitive to axonal density than axonal orientation, and thus may provide a more accurate reflection of axonal changes that occur in neurological injury and disease.  相似文献

Background

Though multiple single nucleotide polymorphisms (SNPs) associated with type 2 diabetes have been identified, the genetic bases of isolated fasting hyperglycaemia (IFH) and isolated postprandial hyperglycaemia (IPH) were still unclear. In present study, we aimed to investigate the association of genome-wide association study-validated genetic variants and IFH or IPH in Han Chinese.

Methods/Principal Findings

We genotyped 27 validated SNPs in 6,663 unrelated individuals comprising 341 IFH, 865 IPH, 1,203 combined fasting hyperglycaemia and postprandial hyperglycaemia, and 4,254 normal glycaemic subjects of Han ancestry. The distributions of genotype frequencies of FTO, CDKAL1 and GCKR were significant different between individuals with IFH and those with IPH (SNP(ptrend): rs8050136(0.0024), rs9939609(0.0049), rs7756992(0.0122), rs780094(0.0037)). Risk allele of FTO specifically increased the risk of IFH (rs8050136: OR 1.403 [95% CI 1.125–1.750], p = 0.0027; rs9939609: 1.398 [1.120–1.744], p = 0.0030). G allele of CDKAL1 specifically increased the risk of IPH (1.217 [1.092–1.355], p = 0.0004). G allele of GCKR increased the risk of IFH (1.167 [0.999–1.362], p = 0.0513), but decreased the risk of IPH (0.891 [0.801–0.991], p = 0.0331). In addition, TCF7L2 and KCNQ1 increased the risk of both IFH and IPH. When combined, each additional risk allele associated with IFH increased the risk for IFH by 1.246-fold (p<0.0001), while each additional risk allele associated with IPH increased the risk for IPH by 1.190-fold (p<0.0001).

Conclusion/Significance

Our results indicate that genotype distributions of variants from FTO, GCKR, CDKAL1 were different between IPH and IFH in Han Chinese. Variants of genes modulating insulin sensitivity (FTO, GCKR) contributed to the risk of IFH, while variants of genes related to beta cell function (CDKAL1) increase the risk of IPH.  相似文献

The vertebrate body plan features a consistent left-right (LR) asymmetry of internal organs. In several vertebrate embryos, motile cilia generate an asymmetric fluid flow that is necessary for normal LR development. However, the mechanisms involved in orienting LR asymmetric flow with previously established anteroposterior (AP) and dorsoventral (DV) axes remain poorly understood. In zebrafish, asymmetric flow is generated in Kupffer's vesicle (KV). The cellular architecture of KV is asymmetric along the AP axis, with more ciliated cells densely packed into the anterior region. Here, we identify a Rho kinase gene, rock2b, which is required for normal AP patterning of KV and subsequent LR development in the embryo. Antisense depletion of rock2b in the whole embryo or specifically in the KV cell lineage perturbed asymmetric gene expression in lateral plate mesoderm and disrupted organ LR asymmetries. Analyses of KV architecture demonstrated that rock2b knockdown altered the AP placement of ciliated cells without affecting cilia number or length. In control embryos, leftward flow across the anterior pole of KV was stronger than rightward flow at the posterior end, correlating with the normal AP asymmetric distribution of ciliated cells. By contrast, rock2b knockdown embryos with AP patterning defects in KV exhibited randomized flow direction and equal flow velocities in the anterior and posterior regions. Live imaging of Tg(dusp6:memGFP)(pt19) transgenic embryos that express GFP in KV cells revealed that rock2b regulates KV cell morphology. Our results suggest a link between AP patterning of the ciliated Kupffer's vesicle and LR patterning of the zebrafish embryo.  相似文献

香樟凋落叶分解物对辣椒生长发育的影响   总被引:1,自引:0,他引:1  
采用盆栽试验,研究了香樟(Cinnamomum camphora)凋落叶(0、25、50、75、100g凋落叶分别与10kg土壤混合)分解过程中对辣椒(Capsicum annuum)及其子代生长发育的影响,并采用气质联用(GC-MS)技术对凋落叶分解前后的萜类物质进行了检测和对比。结果显示:(1)香樟凋落叶分解物显著地抑制了辣椒的叶片数量、整株叶面积以及株高和基径生长,抑制效应随凋落叶剂量的增大而增强,而分解过的凋落叶和塑料片替代凋落叶原样的试验并不抑制辣椒的生长。(2)观察期内(处理45~76d),辣椒的现蕾数、开花数和结实数在各凋落叶处理下均明显低于同期对照;采用指数方程拟合的现蕾、开花和结实动态显示,25g凋落叶处理使辣椒的始蕾期、始花期和始果期分别推迟了0.81、0.17和1.35d,50g处理使这3项参数分别推迟了4.69、5.78和6.27d,而75g和100g处理均使这3项参数推迟10d以上。(3)辣椒的单株果实产量在凋落叶处理下降低29.44%~61.29%,而单果重受到的影响相对较小。(4)辣椒产出的种子千粒重和子代的生长状况并未受到凋落叶分解物的影响。(5)凋落叶的正己烷提取物中萜类占78.05%,而分解后其相对总量下降至40.76%,峰面积下降至原样的约1%。研究认为:香樟凋落叶添加处理对辣椒营养生长和生殖生长产生的明显抑制作用,不是由凋落叶添加对土壤通气透水性或对受体生长造成机械阻隔引起的,其在分解过程中释放樟脑(Camphor)、1,8-桉叶油醇(1,8-cineole)等萜类物质引起化感作用可能才是关键原因。  相似文献
