A method of estimating the population density of roach in Tjeukemeer (21.3 km2) using 20 529 introduced fin-clipped fish is described. Fyke nets proved to be an effective method of sampling the population for marked fish during the spawning season. A total of 20 277 roach were processed during the recapturing period. The population density of roach (⋝ 14 cm) was estimated to be 1 246 458. The growth rate of roach in the lake although relatively poor (von Bertalanffy's L ∞ for males and females, 22 and 26 cm respectively) for the species was similar to that recorded in three other Friesian lakes. The mortality rate of males was higher than that of females. Logarithmic length-weight regression analyses showed that the value of the coefficient varied both within and between the sexes, that of females being higher (range 3.03–3.375) than that of males (range 2.76–3.254). Seasonal changes in the size of the coefficient were due to a disproportionate loss of gonad weight in larger fish. The fecundity of the population was comparatively high for the species.
The total production of the population was estimated to be 95 hg ha−1 of which 39 kg ha−1 was contributed by fry. In older (⋝ IV) fish the production of females (12 kg ha−1) exceeded that (2 kg ha−1) of males, due to differences in their growth and mortality rates. The relatively poor performance of roach in Tjeukemeer, in terms of biomass and production was due to a scarcity of zoobenthos and competition from other species offish. There is no evidence either from this study or the literature that the productive potential of roach in lakes is high, even though macrophytes and detritus can be consumed in significant amounts.  相似文献

Myofibrillar ATPase activity was measured in the epaxial musculature of rainbow trout acclimated to 5 and 21.°C, and goldfish acclimated to 5 and 30°C. The acclimated goldfish showed considerable differences in the specific activity and thermostability of this enzyme. However, such alterations were not apparent in the acclimated trout. Hypoxia-inducible factor-1 alpha (HIF-1 alpha) plays a role in a number of cell-protective pathways after ischemia. There are clear sex-related differences in the remodeling process, and hearts from males tend to dilate in response to pathological loads and ischemia to a greater degree than do hearts from females. Thus we hypothesized that there would be a sex-related dimorphic response of HIF-1 alpha to an ischemic event. Male and female rats were euthanized 5 and 24 h after coronary ligation (M-MI and F-MI; MI, myocardial ischemia), and HIF-1 alpha expression was determined by immunohistochemistry, Western blot, and quantitative RT-PCR. Sham-operated male and female animals served as controls (M-SH and F-SH). In the ischemic area, histochemical analysis at 5 h showed that HIF was expressed in 33% of cell nuclei in M-MI and in 55% in F-MI. At 24 h, HIF expression increased to 49% in M-MI and to 82% in F-MI (P < 0.05 vs. SH and also M-MI vs. F-MI). This difference was not only statistically significant between the two sexes at 24 h but also within each sex at 5 and 24 h after ligation. Western blots confirmed that, at 24 h after ischemia, HIF protein increased significantly in both male and female hearts relative to sham-operated animals but that the increase in females was 60% greater than that seen in males. mRNA expression of HIF was significantly increased at 24 h in F-MI versus M-MI and sham-operated animals. Expression of downstream HIF target genes (heme oxygenase and brain natriuretic peptide) was increased in proportion to the levels of HIF expression. These data suggest a novel cellular mechanism to explain the sex-related dimorphic response to ischemia and also the possibility that exogenous modulation of HIF might represent a new therapeutic approach to preventing left ventricular remodeling.  相似文献

The effect of different temperatures (4 degrees C and 12 degrees C) on myogenic regulatory factors (MyoD and myogenin) and myosin heavy chain (MyHC) expression was investigated in rainbow trout (Onchrhynchus mykiss) during early development. MyoD is first switched on at stage 14 [about 5 somites are formed (1/2 epiboly)] while myogenin mRNA is expressed at stage 15 [around 15 somites are visible (2/3 epiboly)] at both temperatures. Subsequently (up to at least stage 20), the most caudal somites exhibit less myogenin mRNA at 4 degrees C compared to 12 degrees C. At the eyed stage (stage 23-24), both myogenin mRNA and protein are present in greater amounts throughout all myotomes at the lower temperature, with mRNA levels in warmer (12 degrees C) embryos at 83% for MyoD and 72% for myogenin of the levels seen in 4 degrees C embryos. Conversely, however, at this same stage, fast-MyHC mRNA and protein are more abundant in 12 degrees C than in 4 degrees C embryos. This indicates relatively advanced muscle differentiation at the warmer temperature. At hatching, myogenin-positive cells are concentrated within the myosepta at both temperatures and they are also sparsely distributed in the myotome at 4 degrees C, but not at 12 degrees C. MyoD, myogenin, and MyHC levels provide an indication of differentiation of muscle cells. These findings suggest that myogenic regulatory factor expression is delayed but prolonged by the lowering of temperature.  相似文献

Background

Diagnostic accuracy of lymphoma, a heterogeneous cancer, is essential for patient management. Several ancillary tests including immunophenotyping, and sometimes cytogenetics and PCR are required to aid histological diagnosis. In this proof of principle study, gene expression microarray was evaluated as a single platform test in the differential diagnosis of common lymphoma subtypes and reactive lymphadenopathy (RL) in lymph node biopsies.

Methods

116 lymph node biopsies diagnosed as RL, classical Hodgkin lymphoma (cHL), diffuse large B cell lymphoma (DLBCL) or follicular lymphoma (FL) were assayed by mRNA microarray. Three supervised classification strategies (global multi-class, local binary-class and global binary-class classifications) using diagonal linear discriminant analysis was performed on training sets of array data and the classification error rates calculated by leave one out cross-validation. The independent error rate was then evaluated by testing the identified gene classifiers on an independent (test) set of array data.

Results

The binary classifications provided prediction accuracies, between a subtype of interest and the remaining samples, of 88.5%, 82.8%, 82.8% and 80.0% for FL, cHL, DLBCL, and RL respectively. Identified gene classifiers include LIM domain only-2 (LMO2), Chemokine (C-C motif) ligand 22 (CCL22) and Cyclin-dependent kinase inhibitor-3 (CDK3) specifically for FL, cHL and DLBCL subtypes respectively.

Conclusions

This study highlights the ability of gene expression profiling to distinguish lymphoma from reactive conditions and classify the major subtypes of lymphoma in a diagnostic setting. A cost-effective single platform "mini-chip" assay could, in principle, be developed to aid the quick diagnosis of lymph node biopsies with the potential to incorporate other pathological entities into such an assay.

Myocardial physiology in the aftermath of myocardial infarction (MI) before remodeling is an under-explored area of investigation. Here, we describe the effects of MI on the cardiac sarcomere with focus on the possible contributions of reactive oxygen species. We surgically induced MI in 6-7-month-old female CD1 mice by ligation of the left anterior descending coronary artery. Data were collected 3-4 days after MI or sham (SH) surgery. MI hearts demonstrated ventricular dilatation and systolic dysfunction upon echo cardiographic analysis. Sub-maximum Ca-activated tension in detergent-extracted fiber bundles from papillary muscles increased significantly in the preparations from MI hearts. Ca(2+) sensitivity increased after MI, whereas cooperativity of activation decreased. To assess myosin enzymatic integrity we measured splitting of Ca-ATP in myofibrillar preparations, which demonstrated a decline in Ca-ATPase activity of myofilament myosin. Biochemical analysis demonstrated post-translational modification of sarcomeric proteins. Phosphorylation of cardiac troponin I and myosin light chain 2 was reduced after MI in papillary samples, as measured using a phospho-specific stain. Tropomyosin was oxidized after MI, forming disulfide products detectable by diagonal non-reducing-reducing SDS-PAGE. Our analysis of myocardial protein oxidation post-MI also demonstrated increased S-glutathionylation. We functionally linked protein oxidation with sarcomere function by treating skinned fibers with the sulfhydryl reducing agent dithiothreitol, which reduced Ca(2+) sensitivity in MI, but not SH, samples. Our data indicate important structural and functional alterations to the cardiac sarcomere after MI, and the contribution of protein oxidation to this process.  相似文献
