Summary The immune response of mice to a transplacentally induced alveolar cell tumor was studied with the leukocyte adherence inhibition (LAI) assay. The lung tumor, designated 85, was induced in a C3HfB/HeN (C3Hf) mouse by l-ethyl-l-nitrosourea (ENU). While a dose of 105 cells of this tumor does not grow in syngeneic C3Hf mice, it does grow readily in (A×C3Hf)F1 hybrid mice. The tumor possesses a tumor associated transplantation antigen (TATA) which cross-reacts with a normal tissue alloantigen in strain A/HeN (A) mice. Normal mice, tumor-immunized C3Hf mice, and tumor-bearing (A×C3Hf)F1 mice possessed peritoneal cells, the majority of which adhered rapidly to glass and resisted gentle washing. When incubated with an extract of the 85 tumor, peritoneal cells from tumor-immunized mice demonstrated marked inhibition of adherence (62.4%) compared to similarly incubated peritoneal cells of either normal mice (30.3%) or tumor bearing mice (37.1%). Specificity of the reactivity in the LAI assay was demonstrated with a neuroblastoma extract and peritoneal cells from neuroblastoma-immunized C3Hf mice. Peritoneal cells from lung tumor-immunized mice, but not tumor-bearing mice, responded to a lung extract from strain A mice. In contrast to the microcytotoxicity assay, the LAI assay is capable of distinguishing the effective anti-tumor response of tumor-immunized C3Hf mice from the ineffective immune response of tumor-bearing (A×C3Hf)F1 mice. 相似文献

Sequence, organization, and evolution of the A+T region of Drosophila melanogaster mitochondrial DNA 总被引:2,自引:0,他引:2
The long (4.6-kb) A+T region of Drosophila melanogaster mitochondrial DNA
has been cloned and sequenced. The A+T region is organized in two large
arrays of tandemly repeated DNA sequence elements, with nonrepetitive
intervening and flanking sequences comprising only 22% of its length. The
first repeat array consists of five repeats of 338-373 bp. The second
consists of four intact 464-bp repeats and a fifth partial repeat of 137
bp. Three DNA sequence elements are found to be highly conserved in D.
melanogaster and in several Drosophila species with short A+T regions.
These include a 300-bp DNA sequence element that overlaps the DNA
replication origin and two thymidylate stretches identified on opposite DNA
strands. We conclude that the length heterogeneity observed in the A+T
regulatory region in mitochondrial DNAs from the genus Drosophila results
from the expansion (and contraction) of the number of repeated DNA sequence
elements. We also propose that the 300-bp conserved DNA sequence element,
in conjunction with another primary sequence determinant, perhaps the
adjacent thymidylate stretch, functions in the regulation of mitochondrial
DNA replication.

Grassland ecosystems evolved with natural disturbance events on multiple spatial scales in which focal, fine-scale soil disturbance
by animals often was imbedded within large-scale grazing disturbance. The resulting plant communities adapted to both broad-scale
and fine-scale disturbance that resulted in species-rich plant communities. These natural disturbance regimes have been largely
replaced by anthropogenic disturbance. While we generally understand grassland response to modern grazing practices, we know
much less about plant community response to soil disturbance imbedded within non-focal grazing. Therefore, we used a tracked
vehicle to focally disturb soil in a North American mesic mixed prairie that was either undisturbed prairie or prairie with
a recent history of disturbance from either grazing or haying. Successional trajectory and recovery time following focal soil
disturbance was similar between grazed and hayed plant communities. Species composition did not differ (P < 0.05) between grazed or hayed prairie and the respective undisturbed prairie. Plant species richness and bare ground increased
(P < 0.05) following focal soil disturbance in both grazed and hayed communities, but focal soil disturbance combined with either
grazing or haying did not change either plant species richness or bare ground more than (P > 0.05) focal soil disturbance alone. Also, the effect of focal soil disturbance was shortlived with recovery in two growing
seasons. Our results suggest that anthropogenic focal soil disturbance is a reasonable mechanism to restore soil disturbance

Sign surveys are commonly used to study and monitor wildlife species but may be flawed when surveys are conducted only once and cover short distances, which can lead to a lack of accountability for false absences. Multiple observers surveyed for river otter (Lontra canadensis) scat and tracks along stream and reservoir shorelines at 110 randomly selected sites in eastern Kansas from January to April 2008 and 2009 to determine if detection probability differed among substrates, sign types, observers, survey lengths, and near access points. We estimated detection probabilities (p) of river otters using occupancy models in Program PRESENCE. Mean detection probability for a 400-m survey was highest in mud substrates (p = 0.60) and lowest in snow (p = 0.18) and leaf litter substrates (p = 0.27). Scat had a higher detection probability (p = 0.53) than tracks (p = 0.18), and experienced observers had higher detection probabilities (p > 0.71) than novice observers (p < 0.55). Detection probabilities increased almost 3-fold as survey length increased from 200 m to 1,000 m, and otter sign was not concentrated near access points. After accounting for imperfect detection, our estimates of otter site occupancy based on a 400-m survey increased >3-fold, providing further evidence of the potential negative bias that can occur in estimates from sign surveys when imperfect detection is not addressed. Shugoshin is a protein conserved in eukaryotes and protects sister chromatid cohesion at centromeres in meiosis. In our study, we identified the homologs of SGO1 and SGO2 in Arabidopsis thaliana. We show that AtSGO1 is necessary for the maintenance of centromere cohesion in meiosis I since atsgo1 mutants display premature separation of sister chromatids starting from anaphase I. Furthermore, we show that the localization of the specific centromeric cohesin AtSYN1 is not affected in atsgo1, suggesting that SGO1 centromere cohesion maintenance is not mediated by protection of SYN1 from cleavage. Finally, we show that AtSGO2 is dispensable for both meiotic and mitotic cell progression in Arabidopsis. 相似文献
