Xylem sap composition of spruce is influenced by several factors, such as the sampled organ, the sampling period, the availability of soil nutrients, and the soil water potential. Based on literature data and ongoing investigations carried out with adult trees, we present an overview on the main factors influencing xylem sap concentrations of Norway spruce. Direct measurements of nutrient fluxes in the xylem sap are then used to suggest a general scheme of mineral element cycling within adult trees. In Norway spruce (Picea abies Karst.), nutrient concentration in the xylem sap was higher in twigs and fine roots compared to the bottom of the trunk, the highest concentrations beeing observed in spring during the shoot elongation. Xylem sap concentrations were higher in spruce growing at nutrient rich sites than at poor sites. The combination of twig and trunk xylem sap analysis, together with xylem flow measurements in the trunk during the course of a vegetation period allowed the quantification of mineral fluxes via xylem sap flow in the trunk and twigs. These results were compared to gross mineral uptake measurements at the same site. Ca flux in the trunk xylem sap was lower than the gross uptake of Ca. Mg flux in trunk sap was approximately equivalent to Mg gross uptake whereas P and K fluxes in trunk sap were much higher than the gross uptake. Fluxes of Ca, Mg, K and P in the twig sap were much higher than that in trunk sap. Data suggest that internal cycling is responsible for a large part of the nutrient fluxes in the xylem sap of the crown. Xylem sap composition thus appears to be a tool which can complement other sources of information on mineral uptake and cycling in adult spruce  相似文献

A plastid vesicle preparation isolated from exocarpium of young Citrofortunella mitis (calamondin) fruits was able to synthesise monoterpene hydrocarbons when incubated with isopentenyl pyrophosphate. The electron-microscope comparison between this organelle fraction and the various plastid classes present in the peel tissues has shown the structural identity between these plastid vesicles and the leucoplasts of the epithelial cells lining the secretory pockets. The monoterpene biosynthesis required the presence of dimethylallyl pyrophosphate, Mn2+ or Mg2+ and was increased by addition of 2-mercaptoethanol. Evidence is provided that the leucoplast vesicles act as a complete system in which occur all the successive steps involved in monoterpene hydrocarbon elaboration from isopentenyl pyrophosphate.  相似文献

The biosynthesis of monoterpene hydrocarbons was studied in maritime pine needles by incorporation of 14CO2. It was shown that the acyclic terpenes β-myrcene and trans-β-ocimene, act as transitory compounds before the biosynthesis of cyclic monoterpenes such as α- and β-pinene. C17-S1-D-T984 (to be referred to as T984) is a myogenic clonal cell line isolated from a mouse teratocarcinoma. T984 exhibits phenotypic instability since it gives rise not only to myogenic but also to fibro-adipogenic and fibroblastic clones. A cell line of each clone type has been established and studied with respect to (1) phenotypic expression and stability; and (2) growth and differentiation in serum-free and serum-supplemented media. In both respects, marked differences between the three cell lines were observed. All three cell lines respond by increased growth in serum-free media to insulin, transferrin, fibroblast growth factor (FGF) and the serum-spreading factor of Holmes. The fibroblastic and the fibro-adipogenic cell lines can both be grown indefinitely in a serum-free medium which contains the above factors. The fibro-adipogenic cell line, which differentiates in serum-supplemented medium, exhibits very limited differentiation in the absence of serum; the serum factor(s) required for adipogenic differentiation is (are) probably proteins of molecular weight superior to 10 000. In direct contrast, the myogenic cell line exhibits limited growth in serum-free medium but readily differentiates under these conditions. Moreover, myogenic differentiation could be obtained in the defined medium at very low densities and was not influenced by the addition of medium conditioned by cells seeded at high densities. Thus, in this system, muscular differentiation is apparently independent of diffusible endogenous or exogenous factors and is probably triggered by the arrest of growth. While our results do not explain the reason why T984 exhibits phenotypic instability, they do indicate that this clonal cell line and its clonal derivatives could be used to identify the factors that influence the growth and the differentiation of cells of different mesenchymal phenotypes. The possible relationship of phenotypic instability to muscular dystrophies is also discussed.  相似文献

The efficiency of a cantilevered bridle net was tested in comparisonwith a Wisconsin net and a pumping system to sample zooplanktonorganisms in three water layers (epi-, meta- and hypolimnion)of three Canadian Shield lakes. Variations among samplers werecompared to variations due to within-lake vertical distributionof zooplankters and among lake variations. For each lake andwater layer, we also assessed the efficiency of the three methodsaccording to the catches of zooplanktonic taxa. The highestpercentages of variation were generally due to lake or water-layereffects; interaction between sampling gears and water layerswas above 50% for most taxa, except cladoceran. Sampling methodsexplained more variation than the lake effect for some zooplanktontaxa, indicating that using different sampling devices couldpotentially alter the among-lake variation interpretation ofzooplankton abundance. The pumping system captured higher densitiesof animals per taxa than the cantilever and the Wisconsin nets.The cantilever net generally captured mobile taxa more efficiently(Polyarthra vulgaris, copepods, Daphnia sp., Diaphanosoma brachyurumand chaoborids) than the Wisconsin net and the pumping system,but its efficiency varied among water layers.  相似文献

Control of Nitrification by Tree Species in a Common-Garden Experiment   总被引:1,自引:0,他引:1  
We studied the effect of tree species on nitrification in five young plantations and an old native beech coppice forest at the Breuil experimental site in central France. The potential net nitrification (PNN) of soil was high in beech, Corsican pine, and Douglas fir plantations (high nitrifying stands denoted H) and low in spruce and Nordmann fir plantations as well as in native forest stands (low nitrifying stands denoted L). We hypothesized that tree species would stimulate or inhibit nitrification in transplanted soil cores within a few years after the cores were transplanted between stands. We first initiated a transplant experiment where soil cores were exchanged between all stands. The PNN remained high in soil cores from H transferred to H and low in soil cores from L transferred to L. The PNN increased considerably after 16 months in soil cores transferred from L to H, whereas the transfer of soil cores from H to L decreased the PNN only slightly after 28 months. In a second transplant experiment, forest floor material was exchanged between the Douglas fir (H) and the native forest (L) stand. Six months later, the forest floor from the native forest had increased the PNN of the Douglas fir soil considerably, whereas the forest floor from Douglas fir did not affect the PNN of the soil in the native forest stand. It was concluded that beech, Corsican pine, and Douglas fir rapidly stimulate soil nitrification by either activation of suppressed nitrifier communities and/or colonization by new nitrifier communities. Conversely, the slow and irregular reduction of nitrification in spruce, Nordmann fir, and native forest was probably due to the low and heterogeneously distributed flux of inhibiting substances per volume of soil. Our experiments suggest that the inhibition of nitrification is not tightly connected to forest floor leachates, but that the forest floor both reflects and maintains the major ongoing processes. In the long term, humus build up and the production of inhibiting substances may completely block the nitrification activity.  相似文献

Holothurians (sea cucumbers) have been known from ancient times to have the capacity to regenerate their internal organs. In the species Holothuria glaberrima, intestinal regeneration involves the formation of thickenings along the free mesentery edge; these thickenings will later give rise to the regenerated organ. We have previously documented that a remodeling of the extracellular matrix and changes in the muscle layer occur during the formation of the intestinal primordium. In order to analyze these changes in depth, we have now used immunocytochemical techniques and transmission electron microscopy. Our results show a striking disorganization of the muscle layer together with myocyte dedifferentiation. This dedifferentiation involves nucleic activation, disruptions of intercellular junctions, and the disappearance of cell projections, but more prominently, the loss of the contractile apparatus by the formation and elimination of spindle-like structures. Muscle dedifferentiation can be seen as early as 2 days following evisceration and continues during the next 2 weeks of the regeneration process. Dedifferentiation of myocytes might result in cells that proliferate and give rise to new myocytes. Alternatively, dedifferentiating myocytes could give rise to cells with high nuclear-to-cytoplasmic ratios, with some being eliminated by apoptosis. Our results, together with those in other regeneration models, show that myocyte dedifferentiation is a common event in regeneration processes and that the dedifferentiated cells might play an important role in the formation of the new tissues or organs. Benzimidazole-based allosteric inhibitors of the hepatitis C virus (HCV) NS5B polymerase were diversified to a variety of topologically related scaffolds. Replacement of the polar benzimidazole core by lipophilic indoles led to inhibitors with improved potency in the cell-based subgenomic HCV replicon system. Transposing the indole scaffold into a previously described series of benzimidazole-tryptophan amides generated the most potent inhibitors of HCV RNA replication in cell culture reported to date in this series (EC(50) approximately 50 nM).  相似文献
