ABSTRACT The grasslands of southeastern South America (SESA), comprising one of the most extensive grassland ecosystems in the Neotropics, have been negatively impacted by the development of the livestock industry, arable agriculture, and forestry. SESA grasslands have a rich avifauna that includes 22 globally threatened and near‐threatened species, and many other species have suffered local population extinctions and range reductions. In addition to habitat loss and fragmentation, grassland birds in SESA are threatened by improper use of agrochemicals, unfavorable fire management regimes, pollution, and illegal capture and hunting. Studies to date have provided information about the distribution of grassland birds, the threats populations face, and the habitat requirements of some threatened species, but more information is needed concerning dispersal and migration patterns, genetics, and factors that influence habitat use and species survival in both natural and agricultural landscapes. There are few public protected areas in the region (1% of original grasslands), and many populations of threatened grassland birds are found on private lands. Therefore, efforts to preserve grassland habitat must reconcile the interests of land owners and conservationists. Current conservation efforts include establishment of public and private reserves, promotion of agricultural activities that reconcile production with biodiversity conservation, development of multilateral conservation projects across countries, and elaboration of action plans. Measures that result in significant losses to private land owners should include economic compensation, and use of economic incentives to promote agriculture and forestry in native grassland areas should be discouraged, especially in priority areas for grassland birds. Although more studies are needed, some actions, particularly habitat protection and improved management of public and private lands, should be taken immediately to improve the conservation status of grassland birds in SESA.  相似文献

The interaction of water molecules with copper in wild-type azurin and different site-directed mutants of the coordinated residues is studied by nuclear magnetic relaxation dispersion. Different degrees of solvent accessibility are found. The low relaxivity of wild-type azurin agrees with a solvent-protected copper site in solution, the closest water being found at a distance of more than 5?Å from the copper. This low relaxivity contrasts with the relatively large relaxivity of the His46Gly and His117Gly azurin mutants, which shows clear evidence of copper-coordinated water. The data on the latter mutants are best analyzed in terms of one and two water molecules coordinated to the copper in His46Gly and His117Gly, respectively. The Met121His azurin mutant shows an intermediate behavior. The data are analyzed in terms of an increased solvent accessibility with respect to the wild-type azurin, resulting in semi-coordination of water at low pH. These different modes of coordination lead to different geometries, ranging from the trigonal type 1 site of wild-type azurin to the tetragonal type 2 copper sites of the His117Gly and His46Gly azurin mutants through a so-called type 1.5 site of the Met121His mutant. A correlation is found between the relaxation time (τs) of the unpaired electron of copper(II) and the geometry of the metal site: as the tetragonal character decreases the relaxation becomes significantly faster. τs values of ≤1?ns are found for the tetrahedrally distorted type 1 and type 1.5 sites and of 5–15?ns for the tetragonal type 2 sites.  相似文献

Sarcoidosis is a granulomatous disease whose outcome varies from spontaneous remission to chronic refractory disease. Provided that steroids represent the gold standard as a first line treatment, many immune suppressants drugs are currently used in the disease management. However, refractory disease is still a great challenge. Rituximab is an anti-CD20 chimeric monoclonal antibody, currently used for the treatment of B cell malignancies and systemic autoimmune diseases. There are few case reports describing the successful use of Rituximab in refractory sarcoidosis with lung, eye, lymph nodes and skin involvement. In this paper we described three different case reports in which Rituximab has been used to treat refractory sarcoidosis and we reviewed the existing literature.  相似文献

The DNA polymerase gamma (Polg) is a nuclear-encoded enzyme involved in DNA replication in animal mitochondria. In humans, mutations in the POLG gene underlie a set of mitochondrial diseases characterized by mitochondrial DNA (mtDNA) depletion or deletion and multiorgan defects, named POLG disorders, for which an effective therapy is still needed. By applying antisense strategies, ENU- and CRISPR/Cas9-based mutagenesis, we have generated embryonic, larval-lethal and adult-viable zebrafish Polg models. Morphological and functional characterizations detected a set of phenotypes remarkably associated to POLG disorders, including cardiac, skeletal muscle, hepatic and gonadal defects, as well as mitochondrial dysfunctions and, notably, a perturbed mitochondria-to-nucleus retrograde signaling (CREB and Hypoxia pathways). Next, taking advantage of preliminary evidence on the candidate molecule Clofilium tosylate (CLO), we tested CLO toxicity and then its efficacy in our zebrafish lines. Interestingly, at well tolerated doses, the CLO drug could successfully rescue mtDNA and Complex I respiratory activity to normal levels, even in mutant phenotypes worsened by treatment with Ethidium Bromide. In addition, the CLO drug could efficiently restore cardio-skeletal parameters and mitochondrial mass back to normal values. Altogether, these evidences point to zebrafish as a valuable vertebrate organism to faithfully phenocopy multiple defects detected in POLG patients. Moreover, this model represents an excellent platform to screen, at the whole-animal level, candidate molecules with therapeutic effects in POLG disorders.Subject terms: Energy metabolism, Disease model, Experimental models of disease  相似文献
