Caldicellulosiruptor bescii grows optimally at 78°C and is able to decompose high concentrations of lignocellulosic plant biomass without the need for thermochemical pretreatment. C. bescii ferments both C5 and C6 sugars primarily to hydrogen gas, lactate, acetate, and CO2 and is of particular interest for metabolic engineering applications given the recent availability of a genetic system. Developing optimal strains for technological use requires a detailed understanding of primary metabolism, particularly when the goal is to divert all available reductant (electrons) toward highly reduced products such as biofuels. During an analysis of the C. bescii genome sequence for oxidoreductase-type enzymes, evidence was uncovered to suggest that the primary redox metabolism of C. bescii has a completely uncharacterized aspect involving tungsten, a rarely used element in biology. An active tungsten utilization pathway in C. bescii was demonstrated by the heterologous production of a tungsten-requiring, aldehyde-oxidizing enzyme (AOR) from the hyperthermophilic archaeon Pyrococcus furiosus. Furthermore, C. bescii also contains a tungsten-based AOR-type enzyme, here termed XOR, which is phylogenetically unique, representing a completely new member of the AOR tungstoenzyme family. Moreover, in C. bescii, XOR represents ca. 2% of the cytoplasmic protein. XOR is proposed to play a key, but as yet undetermined, role in the primary redox metabolism of this cellulolytic microorganism.  相似文献

Social play between two rhesus monkey (Macaca mulatta) infants takes place mainly when they are both not in body contact with their mothers. This suggests that social play and mother-infant body contact are potential competitors in the infants' time budgets. We investigated whether the presence of a playmate changed the duration of mother-infant body contact during the first 6 months of life. A decrease in contact would favour play opportunity. Mother-infant pairs were observed alternately alone and together with another pair. Resting, which always occurs during on-mother, was not reduced in the presence of a peer. Body contact during activity phases was reduced in most playing pairs, but only to a large extent in pairs which showed relatively high levels of contact in the situation without a peer. Play opportunity was further increased by synchronization of the rest-activity cycles of the two infants; this occurred without a reduction in mother-infant interactions. No influences by mothers on play opportunity were demonstrated, except that strong maternal interference with resting reduced activity synchronization.  相似文献

The economy of predynastic Maadi, situated on a Pleistocene terrace, was largely based upon the exploitation of the Nile valley close by. Wheat and barley were the main crop plants. Melon (Cucumis melo) was also grown, but there is no firm evidence of local pulse crop and flax cultivation. The identification of wheat grains posed problems; it is not clear whether in addition to Triticum dicoccum, free-threshing wheat (T. durum/aestivum) is also represented. Both two-rowed barley (Hordeum distichum) and the six-rowed form (H. vulgare) could be demonstrated. The arable weed flora included Lolium (temulentum), Vicia spp., Rumex (dentatus), Phalaris (paradoxa), Lathyrus hirsuta and Medicago (tuberculata). A concentration of Acacia seeds including pod segments points to the use of Acacia pods for tanning. The Nile valley produced most of the timber used by the Maadi inhabitants: Acacia, Tamarix, Salix. Scarce remains of coniferous wood (Cedrus, Juniperus, Pinus) are evidence of foreign trade.  相似文献

Laudan's thesis that conceptual problem solving is at least as important as empirical problem solving in scientific research is given support by a study of the relation between the chromosome theory and the Mendelian research program. It will be shown that there existed a conceptual tension between the chromosome theory and the Mendelian program. This tension was to be resolved by changing the constraints of the Mendelian program. The relation between the chromosome theory and the Mendelian program is shown to be a good illustration of the influence of science itself on the rational standards governing scientific development.I thank A. One of the surprising insights gained from research in evolutionary developmental biology (evo-devo) is that increasing diversity in body plans and morphology in organisms across animal phyla are not reflected in similarly dramatic changes at the level of gene composition of their genomes. For instance, simplicity at the tissue level of organization often contrasts with a high degree of genetic complexity. Also intriguing is the observation that the coding regions of several genes of invertebrates show high sequence similarity to those in humans. This lack of change (conservation) indicates that evolutionary novelties may arise more frequently through combinatorial processes, such as changes in gene regulation and the recruitment of novel genes into existing regulatory gene networks (co-option), and less often through adaptive evolutionary processes in the coding portions of a gene. As a consequence, it is of great interest to examine whether the widespread conservation of the genetic machinery implies the same developmental function in a last common ancestor, or whether homologous genes acquired new developmental roles in structures of independent phylogenetic origin. To distinguish between these two possibilities one must refer to current concepts of phylogeny reconstruction and carefully investigate homology relationships. Particularly problematic in terms of homology decisions is the use of gene expression patterns of a given structure. In the future, research on more organisms other than the typical model systems will be required since these can provide insights that are not easily obtained from comparisons among only a few distantly related model species.  相似文献

The City of Fredericton, New Brunswick, Canada obtains its water from the Saint John River-recharged Fredericton aquifer. Water quality improves as it flows from the river into the aquifer in a process called bank filtration. Microorganisms contribute to water quality improvements during bank filtration by removing organic carbon. In the Fredericton aquifer, microbially catalyzed reductive dissolution of manganese oxide minerals negatively affects water quality. Aerobic and anaerobic microorganisms were cultured from Fredericton aquifer production well water, from associated bedrock groundwater, and from Saint John River sediment core and sediment groundwater samples. Aerobes were the largest culturable groups in all samples. The ratio of aerobes to other microbial groups, i.e., those reducing nitrate, Mn4 +, Fe3 +, or sulfate, did not vary significantly along the water flow path from the river to the aquifer. Analysis of microbial community composition along the flow path indicated an essentially identical community except in the immediate vicinity of the aquifer. This is in agreement with the absence of macroscale redox zones in the sediment below the Saint John River as determined by groundwater geochemical data. Bacteria isolated from groundwater samples, identified by 16S ribosomal RNA gene sequencing, were α -, β -, γ -, and δ -Proteobacteria, Actinobacteria, and Firmicutes. In contrast to groundwater samples, the majority of bacteria isolated from sediments were γ -Proteobacteria. Numbers of manganese-reducing bacteria, including Aeromonas spp., were small, however Mn4 + reduction ability was widespread in bacteria enriched and isolated with other electron acceptors. The diverse Fredericton aquifer microbial community likely uses manganese oxide minerals as a sink for electrons derived from organic carbon oxidation.  相似文献

Alkaline phosphatase fusions were used to study the membrane topology of DcrA, a protein of 668 amino acids fromDesulfovibrio vulgaris Hildenborough, which has two potentially membrane-spanning hydrophobic sequences at residues 11 to 29 and 188 to 207. A fusion at amino acid residue 170 in the proposed periplasmic domain exhibited high alkaline phosphatase activity, while low activity was observed for a fusion at amino acid residue 284 in the proposed cytoplasmic domain. The data support a topological model for DcrA similar to that of the methyl-accepting chemotaxis proteins of the enteric bacteria.  相似文献
