Background

Toll-like receptors (TLR) and cytokines play a central role in the pathogen clearance as well as in pathological processes. Recently, we reported that TLR2, TLR4 and TLR9 are differentially modulated in injured livers from BALB/c and C57BL/6 (B6) mice during Trypanosoma cruzi infection. However, the molecular and cellular mechanisms involved in local immune response remain unclear.

Methodology/Principal Findings

In this study, we demonstrate that hepatic leukocytes from infected B6 mice produced higher amounts of pro-inflammatory cytokines than BALB/c mice, whereas IL10 and TGFβ were only released by hepatic leukocytes from BALB/c. Strikingly, a higher expression of TLR2 and TLR4 was observed in hepatocytes of infected BALB/c mice. However, in infected B6 mice, the strong pro-inflammatory response was associated with a high and sustained expression of TLR9 and iNOS in leukocytes and hepatic tissue respectively. Additionally, co-expression of gp91- and p47-phox NADPH oxidase subunits were detected in liver tissue of infected B6 mice. Notably, the pre-treatment previous to infection with Pam3CSK4, TLR2-agonist, induced a significant reduction of transaminase activity levels and inflammatory foci number in livers of infected B6 mice. Moreover, lower pro-inflammatory cytokines and increased TGFβ levels were detected in purified hepatic leukocytes from TLR2-agonist pre-treated B6 mice.

Conclusions/Significance

Our results describe some of the main injurious signals involved in liver immune response during the T. cruzi acute infection. Additionally we show that the administration of Pam3CSk4, previous to infection, can attenuate the exacerbated inflammatory response of livers in B6 mice. These results could be useful to understand and design novel immune strategies in controlling liver pathologies.  相似文献

Proteases from a Chilean clone of Alexandrium catenella were studied using gelatin–zymogram gel and protease fluorescent substrate to facilitate their visual identification in vitro and in vivo, respectively. Proteolytic activity bands were grouped arbitrarily according to their molecular weight as P1 (150 and 120 kDa), P2 (100 kDa), P3 (70 and 65 kDa), P4 (60, 55 and 50 kDa) and P5 (25 kDa). Protease inhibitors affect differentially P2 and P3 proteases. Only P2 activity increased in the presence of 1–10 phenanthroline (σPhe), pepstatin A (pepA), leupeptin (leup) and phenylmethanesulfonyl fluoride (PMSF), while P2 and P3 become inactivated with ρ-aminobenzamidine. The protease inhibitors lethal dose was determined by incubating cells with different concentration of the protease inhibitor and evaluating their effect on cell viability. Furthermore, cells treated for 4 h with one lethal dose of 1–10 phenanthroline and ρ-aminobenzamidine, caused serious damage to the intracellular vacuolar system and nuclear material. Live cysts also die when treated independently with these two protease inhibitors. Future work will be aimed at chemically designing species-specific inhibitors for their potential use in killing cysts transported within the sediment of ship ballast water before washing them off to the environment.  相似文献

BACKGROUND: The signal for the onset of septum formation in the fission yeast Schizosaccharomyces pombe is transduced by the septation initiation network (SIN). Many of the components of the SIN are located on the spindle pole body during mitosis, from where it is presumed that the signal for septum formation is delivered. Cdc11 mutants are defective in SIN signaling, but the role of cdc11 in the pathway has remained enigmatic. RESULTS: We have cloned the cdc11 gene by a combination of chromosome walking and transfection of cosmids into a cdc11 mutant. Cdc11p most closely resembles Saccharomyces cerevisiae Nud1p and is essential for septum formation. Cdc11p is a phosphoprotein, which becomes hyperphosphorylated during anaphase. It localizes to the spindle pole body at all stages of the cell cycle, in a sid4p-dependent manner, and cdc11p is required for the localization of all the known SIN components, except sid4p, to the SPB. Cdc11p and sid4p can be coimmunoprecipitated from cell extracts. Finally, like its S. cerevisiae ortholog Nud1p, cdc11p is involved in the proper organization of astral microtubules during mitosis. CONCLUSIONS: We propose that cdc11p acts as a bridge between sid4p and the other SIN proteins, mediating their association with the spindle pole body.  相似文献

In this paper we discuss recent work on the physiological, molecular, and mechanical mechanisms that underlie the capacity of root caps to modulate the properties of the rhizosphere and thereby foster plant growth and development. The root cap initially defines the rhizosphere by its direction of growth, which in turn occurs in response to gradients in soil conditions and gravity. The ability of the root cap to modulate its environment is largely a result of the release of exudates and border cells, and so provides a potential method to engineer the rhizosphere. Factors affecting the release of border cells from the outer surface of the root cap, and function of these cells and their exudates in the rhizosphere, are considered in detail. Release of border cells into the rhizosphere depends on soil matric potential and mechanical impedance, in addition to a host of other environmental conditions. There is good evidence of unidentified feedback signals between border cells and the root cap meristem, and some potential mechanisms are discussed. Root border cells play a significant mechanical role in decreasing frictional resistance to root penetration, and a conceptual model for this function is discussed. Root and border cell exudates influence specific interactions between plant hosts and soil organisms, including pathogenic fungi. The area of exudates and border cell function in soil is an exciting and developing one that awaits the production of appropriate mutant and transgenic lines for further study in the soil environment.  相似文献

Data were obtained on the population structure and reproduction of Atlantoscia floridana, one of the most common species of terrestrial isopods in the restinga (coastal dune) forests of the state of Rio Grande do Sul, southern Brazil. During a 19-month period, a total of 7833 individuals were sampled: 2792 males, 3400 females and 1691 mancas. There was a significant difference between the size of both males and females collected in 2000 and 2001: the mean size was smaller in the second year when individuals in the larger size classes were lacking. Population density varied with season. The minimum population was 131 ind per m2 individuals, the maximum 1040 ind per m2 and the mean 450  per m2. While the overall sex ratio was clearly female biased, the operational sex ratio favored males, and showed no changes with season. Because both ovigerous and post-ovigerous females were present throughout the year, reproduction is considered continuous; however, reproduction peaked during autumn and spring. Ovigerous females were measured (CW = cephalothorax width) and the number of eggs was counted. Fecundity (F) varied from 5 to 23 eggs ( = 11.18 ± 4) per female, and was expressed by the regression F = –18.48 + 22.59 CW, with the female cephalothorax width varying from 1.04 to 1.68 mm. Marsupial mortality was only 0.9%. Egg production was 588 eggs per m2 in spring and 660 eggs per m2 in autumn. Recruitment occurred in all months, and eggs, embryos and marsupial mancas were also present year-round. A. floridana is the dominant species of terrestrial isopod in the study area. Its most remarkable characteristic is its high reproductive investment.  相似文献

We aimed at determining involvement of extracellular matrix proteins (ECMp) and an ECM-binding adhesin (32-kDa protein) from Paracoccidioides brasiliensis, in the course of experimental paracoccidioidomycosis. BALB/c mice were infected with P. brasiliensis conidia previously incubated with soluble laminin, fibronectin and fibrinogen or a mAb against the fungal adhesin. Inflammatory response, chitin levels and cytokine production at different postinfection periods were determined. Chitin was significantly decreased in lungs of mice infected with ECMp-treated conidia when compared with controls at week 8, especially with laminin and fibrinogen. Contrariwise, when animals were infected with mAb-treated conidia no differences in chitin content were found. The observed inflammatory reaction in lungs was equivalent in all cases. IFN-gamma increased significantly in lungs from mice infected with soluble ECMp - (at day 4 and week 12) or mAb-treated conidia (at week 12) when compared with animals infected with untreated conidia. Significant increased levels of tumour necrosis factor-alpha were observed at 8 weeks in animals infected with ECMp-treated conidia while no differences were observed during the remaining periods. These findings point toward an inhibitory effect of ECMp on P. brasiliensis conidia infectivity and suggest that these proteins may interfere with conidia initial adhesion to host tissues probably modulating the immune response in paracoccidioidomycosis.  相似文献

Abstract. Freshwater crayfishes of the genus Parastacus are intersex, i.e., show characteristics of both sexes in the same individual; also, intersexuality has been documented in hermaphroditic species. The aim of this study was to analyze the gonads of Parastacus varicosus , characterizing its sexual pattern. The animals were collected at Cova do Touro, Gravataí, Rio Grande do Sul, Brazil. Three sexual forms were identified: intersex females, intersex males, and transitional specimens showing an ovotestes gonad that could only be identified by means of histological analysis. All specimens had two pairs of gonopores and gonoducts. The oviduct had a wider diameter in females, whereas in males the vasa deferentia were more developed. Gonads were composed of two parallel structures located in the cephalothorax. Female gonads were classified into three stages. The presence of transitional specimens may indicate that sex change occurs, with protandric hermaphroditism, as observed in other species of Parastacus . Because previous studies had demonstrated that P. varicosus is a gonochoristic species in a population from Uruguay, the present study addressed the possibility that the sexual pattern depends on differences in environmental factors among populations. To improve understanding of evolution of hermaphroditism in these crustaceans, their reproductive dynamics should be studied, including identification of the factors stimulating male and female functions.  相似文献

Wilson disease (WND), an autosomal recessive disorder of copper transport with a broad range of genotypic and phenotypic characteristics, results from mutations in the ATP7B gene. ATP7B encodes a copper transporting P-type ATPase involved in the transport of copper into the plasma protein ceruloplasmin, and for excretion of copper from the liver. Defects in ATP7B lead to copper storage in liver, brain and kidney. Mutation analysis was carried out on 300 WND patients of various origins, and new mutations not previously reported were identified: European white (p.L217X, c.918_931, c.1073delG, c.3082_3085delAAGAinsCG, p.V536A, p.S657R, p.A971V, p.T974M, p.Q1004P, p.D1164N, p.E1173G, p.I1230V, p.M1359I, c.2355+4A>G), Sephardic Jewish (p.Q286X), Filipino (p.G1149A), Lebanese (p.R1228T), Japanese (p.D1267V) and Taiwanese (p.A1328T). All but one missense variant have strong evidence for classification as disease-causing mutations. In the patients reported here, we also identified 20 nucleotide substitutions, six not previously reported, which cause silent amino acid changes or intronic changes. Documentation and characterization of all variants is essential for accurate DNA diagnosis in WND because of the wide range of clinical and biochemical variability.  相似文献
