N-Bromosuccinimide cleavage of in vivo 32P-labelled lysine-rich histone isolated from rapidly dividing cells has been studied. N-Bromosuccinimide cleaves F1-histone into two fragments, a small N-terminal piece and a larger C-terminal portion. The phosphate-induced microheterogeneity and associated radioactivity which has been linked to cell replication, is found in the carboxyterminal fragment. No phosphorous is found associated with the amino-terminal fragment when histone phosphorylation is associated with cell division. The specific tryptic phosphopeptides obtained from in vivo labelled F1 are clearly different from those obtained from in vitro incubations of free F1-histone and cytoplasmic protein kinase.  相似文献

O 6-(carboxymethyl)guanine (O 6-CMG) and O 6-(4-oxo-4-(3-pyridyl)butyl)guanine (O 6-pobG) are toxic lesions formed in DNA following exposure to alkylating agents. O 6-CMG results from exposure to nitrosated glycine or nitrosated bile acid conjugates and may be associated with diets rich in red meat. O 6-pobG lesions are derived from alkylating agents found in tobacco smoke. Efficient syntheses of oligodeoxyribonucleotides (ODNs) containing O 6-CMG and O 6-pobG are described that involve nucleophilic displacement by the appropriate alcohol on a common synthetic ODN containing the reactive base 2-amino-6-methylsulfonylpurine. ODNs containing O 6-pobG and O 6-CMG were found to be good substrates for the S. pombe alkyltransferase-like protein Atl1.

By examining serial serum samples from patients undergoing surgical removal of malignant tumours an increased incidence of tumour-specific antibodies has been detected. The mass digitization of books is changing the way information is created, disseminated and displayed. Electronic book readers (e-readers) generally refer to two main display technologies: the electronic ink (E-ink) and the liquid crystal display (LCD). Both technologies have advantages and disadvantages, but the question whether one or the other triggers less visual fatigue is still open. The aim of the present research was to study the effects of the display technology on visual fatigue. To this end, participants performed a longitudinal study in which two last generation e-readers (LCD, E-ink) and paper book were tested in three different prolonged reading sessions separated by - on average - ten days. Results from both objective (Blinks per second) and subjective (Visual Fatigue Scale) measures suggested that reading on the LCD (Kindle Fire HD) triggers higher visual fatigue with respect to both the E-ink (Kindle Paperwhite) and the paper book. The absence of differences between E-ink and paper suggests that, concerning visual fatigue, the E-ink is indeed very similar to the paper.  相似文献

Little of the historical extent of tallgrass prairie ecosystems remains in North America, and therefore there is strong interest in restoring prairies. However, slow‐growing prairie plants are initially weak competitors with the fast‐growing yet short‐lived weedy plant species that are typically abundant in recently established prairie restorations. One way to aid establishment of slow‐growing plant species is through adding soil amendments to prairie restorations before planting. Arbuscular mycorrhizal (AM) fungi form mutualisms with the roots of most terrestrial plants and are particularly important for the growth of slow‐growing prairie plant species. As prairie ecosystems are adapted to fires that leave biochar (charred organic material) in the soil, adding biochar as well as AM fungal strains from undisturbed remnant prairies into the soil of prairie restorations may improve restoration outcomes. Here, we test this prediction during the first four growing seasons of a prairie restoration. When prairie plant seedlings were inoculated prior to planting into the field with AM fungi derived from remnant prairies, that one‐time inoculation significantly increased growth of five of the nine tested plant species through at least two growing seasons. This long‐term benefit of AM fungal inoculation was unaffected by biochar addition to the soil. Biochar application rates of at least 10 tons/ha significantly decreased Coreopsis tripteris growth but acted synergistically with AM fungal inoculation to significantly improve survival of Schizachyrium scoparium. Overall, inoculation with native AM fungi can help promote prairie plant establishment, but concomitant use of biochar soil amendments had relatively little effect.  相似文献
