Plants were regenerated by somatic embryogenesis from endosperm tissue of open-pollinated seeds of Juglans regia L. cv Manregian. These plants were obtained by growing endosperm tissue on media similar to those used for plant regeneration from walnut cotyledons (Tulecke and McGranahan 1985). The plants appear morphologically uniform and have a triploid chromosome number of 3n=48. Nine plants have been grown to a young sapling stage in soil. This embryogenic line from endosperm has been maintained in culture for two years by the process of repetitive somatic embryogenesis.  相似文献

Hepatitis C virus (HCV) is a positive-strand RNA virus of the Flaviviridae family and a major cause of liver disease worldwide. HCV replicates in the cytoplasm, and the synthesis of viral proteins induces extensive rearrangements of host cell membranes producing structures, collectively termed the membranous web (MW). The MW contains the sites of viral replication and assembly, and we have identified distinct membrane fractions derived from HCV-infected cells that contain replication and assembly complexes enriched for viral RNA and infectious virus, respectively. The complex membrane structure of the MW is thought to protect the viral genome limiting its interactions with cytoplasmic pattern recognition receptors (PRRs) and thereby preventing activation of cellular innate immune responses. Here we show that PRRs, including RIG-I and MDA5, and ribosomes are excluded from viral replication and assembly centers within the MW. Furthermore, we present evidence that components of the nuclear transport machinery regulate access of proteins to MW compartments. We show that the restricted assess of RIG-I to the MW can be overcome by the addition of a nuclear localization signal sequence, and that expression of a NLS-RIG-I construct leads to increased immune activation and the inhibition of viral replication.  相似文献

Proteolysis within the lipid bilayer is poorly understood, in particular the regulation of substrate cleavage. Rhomboids are a family of ubiquitous intramembrane serine proteases that harbour a buried active site and are known to cleave transmembrane substrates with broad specificity. In vitro gel and Förster resonance energy transfer (FRET)‐based kinetic assays were developed to analyse cleavage of the transmembrane substrate psTatA (TatA from Providencia stuartii). We demonstrate significant differences in catalytic efficiency (kcat/K0.5) values for transmembrane substrate psTatA (TatA from Providencia stuartii) cleavage for three rhomboids: AarA from P. stuartii, ecGlpG from Escherichia coli and hiGlpG from Haemophilus influenzae demonstrating that rhomboids specifically recognize this substrate. Furthermore, binding of psTatA occurs with positive cooperativity. Competitive binding studies reveal an exosite‐mediated mode of substrate binding, indicating allostery plays a role in substrate catalysis. We reveal that exosite formation is dependent on the oligomeric state of rhomboids, and when dimers are dissociated, allosteric substrate activation is not observed. We present a novel mechanism for specific substrate cleavage involving several dynamic processes including positive cooperativity and homotropic allostery for this interesting class of intramembrane proteases.  相似文献

The new antiestrogen toremifene (TOR) is currently on the market for the treatment of advanced breast cancer in postmenopausal women. TOR is known to exhibit a similar efficacy profile as tamoxifen (TAM) in the treatment of advanced breast cancer and there are studies to suggest that the beneficial side effects of TAM on bone and blood lipids are also achieved with TOR. However, the data concerning the action of TOR on the endometrium is sorely lacking. In light of the estrogenic effect of TAM on the uterus and the 2–3-fold increased incidence in endometrial carcinoma detected in patients receiving TAM therapy, it is imperative to investigate the effect of TOR on endometrial carcinoma. We compared the actions of TAM and TOR on the EnCa101 human endometrial tumor model and find that both antiestrogens have similar growth stimulatory effects. To investigate a potential mechanism of antiestrogen-stimulated endometrial tumor growth, we have examined known activators of the AP-1 signal transduction pathway, the protein kinase C (PKC) family of isozymes, in the EnCa101 human endometrial tumor model. We find that increased PKC isozyme expression correlates with hormone-independent breast cancer as well as antiestrogen-stimulated endometrial cancer.  相似文献

Bone marrow transplantation is being used with increasing frequency as therapy for a number of life-threatening diseases. Data reported to the International Bone Marrow Transplant Registry are presented to show the pattern of growth of bone marrow transplantation worldwide as well as current results in leukemia and aplastic anemia. Risk factors are identified that may predict the development of major complications after bone marrow transplantation.  相似文献

Mucuna (Fabaceae) species possess gullet-type flowers that open explosively and which are thought to be specifically adapted for bat- or bird-pollination. However, recent studies have shown that non-flying mammals are also important pollinators of this genus in Asia. Here we report on the pollination system of Mucuna championii (endemic in southeast China) in Hong Kong. As is typical for the genus, explosive opening is essential for fruit set, but flowers are unable to open in the absence of manipulation by an effective pollinator. Camera trap surveys of three individuals revealed both chestnut spiny rats (Niviventer fulvescens) and short-nosed fruit bats (Cynopterus sphinx) to be capable of triggering explosive opening. The number of flowers opened by each species did not differ significantly, and both removed most pollen grains from the flowers they visited, but either species visited different individuals. Sucrose-rich nectar was secreted by flowers throughout the day. Our results reveal that M. championii can be pollinated by both rats and bats, with this representing only the second report of rat-pollination in tropical Asia. The sympatric M. birdwoodiana often occurs in close proximity to M. championii and has an overlapping flowering season, suggesting that pollinator segregation may have played a role in shaping the evolutionary ecology of these two species.  相似文献

Total dark respiration (vt), cytochrome pathway (veyt), alternativepathway (valt) and residual (vres) respiration were measuredin Lemna gibba plants, by the use of pathway inhibitors. NaCNwas used to inhibit veyt and SHAM (salicylhydroxamic acid) toinhibit valt. Residual respiration (vres) was about 5% of vt.The effect of high (100 Pa) and low (0 Pa) carbon dioxide partialpressure ([CO2]) on vt, veyt and valt was determined from bothCO2 efflux and O2 uptake measurements. The higher [CO2] suppressedvt by about 30%. When respiration operated through the cytochromepathway only, in the absence of valt, it was suppressed by about12% as measured by the O2 uptake of submerged Lemna fronds orby about 40% as measured by CO2 efflux from floating fronds.The higher [CO2] treatment had only a small effect on respiration,when valt alone operated. There was no evidence of a specificsuppression of the valt pathway by high [CO2]. Succinic dehydrogenaseactivity of the mitochondria of roots of Medicago sativum wasreduced by 18%, when the mitochondria were pre-treated with120 as compared to 34 Pa [CO2]. There was no such effect oncytochrome c oxidase activity of mitochondria under the sameconditions.
