Mycosphaerella nubilosa is one of the most important Eucalyptus leaf pathogens, causing premature defoliation and stunting of growth. The aim of this study was to develop polymorphic microsatellite markers for M. nubilosa. Fifteen primer sets were developed and evaluated for polymorphism. Two primers were monomorphic, three primers did not amplify the desired region and 10 primer pairs were polymorphic. These microsatellite markers will be applied to population biology studies of M. nubilosa collections from several countries. These studies will promote an understanding of the genetics and the global movement of M. nubilosa that is severely limiting plantation development.  相似文献

The evolutionary importance of cryptic taxa is well documented, yet few studies have examined them with a view to conservation. In the present study, the significance of cryptic speciation in freshwater crabs is examined. Allozyme and 16S rRNA sequence data were used to explore the degree of population differentiation between mountain stream populations of two distinct freshwater crab species. Marked patterns of differentiation were evident among populations; those in close geographical proximity were characterized by the fixation of alternate alleles at certain loci, indicating that currently there is no gene flow among populations. Both allozyme and sequence data provide evidence for the recognition of at least five distinct evolutionary lineages with pronounced levels of genetic differentiation. Morphometric data failed to detect any distinct geographically intermediate population groupings. Mature second leaves of Lolium perenne L. cv. Vigor, were sampledin a spring and summer regrowth period. Effects of CO2enrichmentand increased air temperature on stomatal density, stomatalindex, guard cell length, epidermal cell density, epidermalcell length and mesophyll cell area were examined for differentpositions on the leaf and seasons of growth. Leaf stomatal density was smaller in spring but greater in summerin elevated CO2and higher in both seasons in elevated temperatureand in elevated CO2xtemperature relative to the respective controls.In spring, leaf stomatal index was reduced in elevated CO2butin summer it varied with position on the leaf. In elevated temperature,stomatal index in both seasons was lower at the tip/middle ofthe leaf but slightly higher at the base. In elevated CO2xtemperature,stomatal index varied with position on the leaf and betweenseasons. Leaf epidermal cell density was higher in all treatmentsrelative to controls except in elevated CO2(spring) and elevatedCO2xtemperature (summer), it was reduced at the leaf base. Inall treatments, stomatal density and epidermal cell densitydeclined from leaf tip to base, whilst guard cell length showedan inverse relationship, increasing towards the base. Leaf epidermalcell length and mesophyll cell area increased in elevated CO2inspring and decreased in summer. In elevated CO2xtemperatureleaf epidermal cell length remained unaltered in spring comparedto the control but decreased in summer. Stomatal conductancewas lower in all treatments except in summer in elevated CO2itwas higher than in the ambient CO2. These contrasting responses in anatomy to elevated CO2and temperatureprovide information that might account for differences in seasonalleaf area development observed in L. perenne under the sameconditions. Four field experiments were sown with AMV‐infected or healthy seed of burr medic (Medicago polymorpha) and grazed by sheep; two were sown with cv. Circle Valley and two with cv. Santiago. Seed‐infected plants acted as primary sources for virus spread by naturally occurring aphids. Insecticides and admixture with annual ryegrass (Lolium rigidum), a non‐host of AMV, suppressed virus spread to different extents in the plots sown with infected seed. Effects of the different amounts of virus spread obtained on overall concentration of the oestrogenic compound coumestrol (dihydroxycoumestan) in dry stems and pods, and on seed production were measured in the medic. With cv. Santiago, stem and pod coumestrol concentration values for plots sown with healthy seed were significantly smaller than those for all plots sown with infected seed regardless of whether they were sprayed. With cv. Circle Valley, the coumestrol values for stems from plots sown with healthy seed were significantly smaller than those for unsprayed plots sown with infected seed but not than those for sprayed plots or ones with grass admixtures, and there were no significant differences with pods. There was always a significant positive relationship between concentration of coumestrol in medic stems and percentage AMV infection of swards; this was also so with pods in two experiments. A linear model best fitted this relationship with cv. Circle Valley but a logarithmic model did so with cv Santiago. In glasshouse grown plants, the coumestrol content of dried medic shoots was increased 11 (cv. Circle Valley) and five (cv. Santiago) times by AMV infection. AMV increased mean coumestrol concentrations up to 256 ppm (field) and 237 ppm (glasshouse) in stems and 223 ppm in pods (field). Sowing healthy seed in new pasture swards was an effective strategy for minimising coumestrol accumulation in burr medic swards. Two, but not single, applications of a newer generation pyrethroid insecticide to swards in which AMV was spreading significantly diminished coumestrol accumulation but applying organophosphorus insecticide twice and carbamate insecticide repeatedly did not. Medic seed yields and individual seed weights were sometimes significantly increased by the treatments, suppression of AMV spread by regular carbamate sprays being sufficient to increase seed yield by 55%.  相似文献

Spring wheat (Triticum aestivumL., ‘Chablis’) wasgrown under field conditions from sowing until harvest maturity,except for a 12-d period [70–82 days after sowing (DAS)coinciding with anthesis] during which replicated crop areaswere exposed to a range of temperatures within two pairs ofpolyethylene-covered temperature gradient tunnels. At 82 DAS,an increase in mean temperature from 16 to 25 °C duringthis treatment period had no effect on above-ground biomass,but increased ear dry weight from 223 to 327 g m-2and, at 83DAS, reduced root biomass from 141 to 63 g m-2. Mean temperatureover the treatment period had no effect on either above-groundbiomass or grain yield at maturity. However, the number of grainsper ear at maturity declined with increasing maximum temperaturerecorded over the mid-anthesis period (76–79 DAS) and,more significantly, with maximum temperature 1 d after 50% anthesis(78 DAS). Grain yield and harvest index also declined sharplywith maximum temperature at 78 DAS. Grain yield declined by350 g m-2at harvest maturity with a 10 °C increase in maximumtemperature at 78 DAS and was related to a 40% reduction inthe number of grains per ear. Grain yield was also negativelyrelated to thermal time accumulated above a base temperatureof 31 °C (over 8 d of the treatment from 5 d before to 2d after 50% anthesis). Some praying mantids have sensitive ultrasonic hearing arising from a unique 'cyclopean' ear located in the ventral metathorax. The present study explores the evolutionary history of the mantis auditory system by integrating large anatomical, neurophysiological, behavioural, and molecular databases. Using an 'auditory phylogeny' based on 13 morphological characters, we identified a primitively earless form of metathoracic anatomy in several extant taxa. In addition, there are five distinct mantis auditory systems. Three of these can be identified anatomically, and the other two can only be detected neurophysiologically. Superimposing these results onto a phylogenetic tree derived from molecular data from seven genes shows that the cyclopean mantis ear evolved once approximately 120 Mya. All the other auditory system types are either varying degrees of secondary loss, or are recent innovations that each occurred independently multiple times. The neurophysiological response to ultrasound is remarkably consistent across all taxa tested, as is the multicomponent, in-flight behaviour triggered by ultrasound. Thus, mantids have an ancient, highly conserved auditory neural–behavioural system. Although ultrasonic hearing in several insect groups evolved in response to bat predation, mantis hearing predates the appearance of bats (approximately 63 Mya) and must originally have functioned in communication, prey detection, or avoidance of nonbat predators.  © 2008 The Linnean Society of London, Biological Journal of the Linnean Society , 2008, 94 , 541–568.  相似文献

1. Changes in nutrients and climate have occurred over approximately the same timescales in many European lake catchments. Here, we attempt to interpret the sedimentary diatom record of a large shallow lake, Loch Leven, in relation to these pressures using information gained from analysis of long‐term data sets of water quality, climate and planktonic diatoms. 2. The core data indicate the enrichment of Loch Leven starting in c. 1800–1850, most likely from agricultural practices in the catchment, with a more marked phase since c. 1940–1950 caused by increased phosphorus inputs from sewage treatment works, land drainage and a woollen mill. 3. While the recent diatom plankton remains are dominated by taxa associated with nutrient‐rich conditions, an increase in Aulacoseira subarctica relative to Stephanodiscus taxa since the mid‐1980s suggests that reductions in external catchment sources of nutrients (since 1985) may have resulted in partial recovery. This observation accords well with the long‐term monitoring series of water chemistry and phytoplankton. 4. On a decadal‐centennial scale, the eutrophication signal in the sediment record outweighs any evidence of climate as a control on the diatom community. However, at an inter‐annual scale, while the diatom data exhibit high variability, there are several changes in species composition in the recent fossil record that may be attributed to climatic controls. 5. The study highlights the value of a palaeolimnological approach, particularly when coupled with long‐term data sets, for developing our understanding of environmental change at a range of temporal scales. The diatom record in the sediment can be used effectively to track recovery from eutrophication, but requires greater understanding of contemporary ecology to fully interpret climate impacts. 6. The study illustrates the complexity of ecosystem response to synchronous changes in nutrients and climate, and the difficulty of disentangling the effects of these multiple, interacting pressures.  相似文献

The European alder species, Alnus cordata and A. glutinosa have sympatric distributions on Corsica and southern Italy, where they often grow in mixed populations. An assessment was made of the levels of genetic variation in the chloroplast and nuclear genomes of individual trees from 22 alder populations from Corsica and Italy. The distribution of 10 chloroplast DNA polymorphisms generated by the PCR-RFLP method shows strong geographic structuring of cpDNA haplotypes. Conversely, variation in the nuclear genome, as determined by ISSR analysis, is strongly correlated to the taxonomic affinities of the individual trees analysed. Approximately 50% of ISSR variation (P= 0.00, AMOVA) could be attributed to differences between species. Possible reasons for the incongruence between variation in the chloroplast and nuclear genomes are discussed.  相似文献
