Observations that cytoskeletal proteins assemble in vivo close to the time and site of synthesis have been confirmed and extended by an in vitro translation system. HeLa cytoskeletons prepared with Triton in a translation-extraction buffer without reticulocyte or wheat germ lysate efficiently incorporate 35S-methionine into polypeptides, and are stable during this translation. Cytoskeletal proteins translated in this way associate with the HeLa cytoskeleton independent of the concentration of soluble proteins. These associations are puromycin-resistant before the proteins are complete; the protein associations made in vitro show only minor differences from those made in vivo. The protein associations are not simply a consequence of protein solubility in the buffers used, as the associations require initiation in vivo. These results indicate that many cytoskeletal proteins associate with the cytoskeleton during translation.  相似文献

Abstract Although most reptiles have polygynous mating systems without long‐term pair bonds, one lineage of large scincid lizards in Australia is exceptional in this respect. Reports of complex sociality in the genus Egernia led us to conduct the first radiotelemetric field study of a species within this group. Land mullets (Egernia major) are large (60 cm total length), viviparous lizards from rainforest habitats in south‐eastern Australia. To document the spatial ecology and social organization of this species, we captured 12 adult lizards in the Barrington Tops area of eastern New South Wales and implanted them with miniature radiotransmitters. The lizards were released at their sites of capture and located daily for the next 6 weeks. All of the radiotracked lizards had discrete home ranges of approximately 10 000 m2, based around well‐defined core areas (approximately 2000–3000 m2). Females tended to move further, and to range over wider areas, than did males. All of the radiotracked lizards lived in social groups consisting of one or more adult males and females plus juveniles of all age classes. Subgroups were apparent within one group of five radio‐tagged lizards: individual animals consistently shared their shelter sites and home ranges with one or more specific individuals. Male/female pairings were more frequent than expected under the null hypothesis of random association among individuals. The data in the present study support anecdotal reports of pair bonds in E. major and support suggestions that the social systems of species in this genus are more complex than those of previously studied reptiles.  相似文献

In sediments, the bioavailability and toxicity of Ni are strongly influenced by its sorption to manganese (Mn) oxides, which largely originate from the redox metabolism of microbes. However, microbes are concurrently susceptible to the toxic effects of Ni, which establishes complex interactions between toxicity and redox processes. This study measured the effect of Ni on growth, pellicle biofilm formation and oxidation of the Mn-oxidizing bacteria Pseudomonas putida GB-1. In liquid media, Ni exposure decreased the intrinsic growth rate but allowed growth to the stationary phase in all intermediate treatments. Manganese oxidation was 67% less than control for bacteria exposed to 5 μM Ni and completely ceased in all treatments above 50 μM. Pellicle biofilm development decreased exponentially with Ni concentration (maximum 92% reduction) and was replaced by planktonic growth in higher Ni treatments. In solid media assays, growth was unaffected by Ni exposure, but Mn oxidation completely ceased in treatments above 10 μM of Ni. Our results show that sublethal Ni concentrations substantially alter Mn oxidation rates and pellicle biofilm development in P. putida GB-1, which has implications for toxic metal bioavailability to the entire benthic community and the environmental consequences of metal contamination.  相似文献

A monoclonal antibody (ISU-cA) was produced that recognized certain alloantigens of the chicken A blood group locus. Antigens produced by alleles A3, A4 and A8 were positive, and those produced by A2 and A5 were negative, by haemagglutination. The specificity of ISU-cA for chicken A blood group antigens was demonstrated by serologic analyses, genetic crosses and competitive inhibition of binding by anti-A alloantisera. To our knowledge, this is the first reported monoclonal antibody against a chicken blood group alloantigen system other than the B complex.  相似文献
