All three HLA class II families (DR, DQ, and DP) are involved in restriction of helper T cell (Th) recognition of nominal antigens including CMV. Only limited studies have been described previously to determine whether restricting determinants of DR and especially DQ are subtypic to the serologically defined DR and DQ specificities, and to what extent restricting determinants are associated with Dw specificities defined in alloresponses. In the present report, we describe a large number of CMV-specific Th clones derived from two different individuals who are seropositive for CMV. Clones were classified as being DR-, DQ-, or DP-reactive based on blocking with monoclonal antibodies. DR- and DQ-restricted clones were then examined in panel studies using antigen-presenting cells (APC) expressing the Dw subtype of the restricting DR-DQ haplotype, as well as APC expressing different Dw subtypes associated with the serologically defined specificity. Unrelated specificities were also included. Our findings show that not only for DR but for DQ as well, the primary restricting determinants appear to be subtypic to the serologically defined antigen; furthermore, subtype restriction for both DR and DQ is very closely associated with single Dw specificities. In several cases in which cross-reactivity among restricting Dw specificities was observed in association with a given DR or DQ haplotype, a molecular basis could be suggested to explain the cross-reacting determinants. A small minority of the clones appeared to be CMV specific, but was restricted by a determinant(s) that is either monomorphic or minimally polymorphic.  相似文献

Sources of inorganic carbon (Ci) for photosynthesis and calcification and the mechanisms involved in their uptake in scleractinian corals were investigated in microcolonies of Galaxea fascicularis. Direct measurements of Ca2+, pH and O2 on the surface and inside the polyp's coelenteron were made with microsensors. Gross photosynthesis (Pg) and net photosynthesis (Pn) were measured on the surface. Light respiration (LR) was calculated from Pg and Pn. The effect of light/dark and dark/light switches on Ca2+ and pH dynamics on the surface and inside the coelenteron were followed. To evaluate the different sources of Ci for photosynthesis and calcification, Ci-free seawater and 6-Ethoxyzolamide and Acetazolamide, inhibitors for carbonic anhydrase (CA) were used.In normal seawater, Pg was about seven times higher than Pn, the LR was ca. 80-90% of the Pg. Thus, most of the O2 produced in Pg are immediately consumed in respiration, indicating the presence of a highly active internal C-cycle. As the internal C-cycle is highly active, a large part of the Ci for calcification will have passed through the metabolism of the symbiont. The high LR provides ATP for energy requiring processes in light.Ci for photosynthesis and calcification can come from seawater in the form of free Ci, respiration of photosynthates (internal C-cycle) or respiration of the ingested plankton. These sources form a common carbon pool (C-pool) that is used for the different processes.In Ci-free seawater, Pg decreased by about 12.5%, indicating that most of the photosynthetically fixed Ci can temporarily be supplied from internal sources. The initial decalcification, observed directly upon the switch to Ci-free seawater, showed that the Ca-pools in the coral are exchangeable. Part of the Pg in Ci-free seawater may depend on this decalcification for its Ci supply.Three localities of CA were defined. One on the surface facing seawater and one on endodermal cells facing the coelenteron, while the third is intracellular. The inhibition of CA decreased Pg by about 30%, while it increased the concentration of Ca2+ as a result of a decrease in its precipitation. The reduction of photosynthesis and calcification by CA inhibition demonstrated that both processes need the enzyme for the supply of Ci. The pH on the surface and inside the coelenteron decreased upon 6-Ethoxyzolamide addition indicating a role of CA in pH control.  相似文献

Group I metabotropic glutamate receptors (mGluRs) have been demonstrated to play a role in synaptic plasticity via a rapamycin-sensitive mRNA translation signaling pathway. Various growth factors can stimulate this pathway, leading to the phosphorylation and activation of mammalian target of rapamycin (mTOR), a serine/threonine protein kinase that modulates the activity of several translation regulatory factors, such as p70S6 kinase. However, little is known about the cellular and molecular mechanisms that bring the plastic changes of synaptic transmission after stimulation of group I mGluRs. Here, we investigated the role of the mTOR-p70S6K and the ERK1/2-p70S6K pathways in rat striatal and hippocampal synaptoneurosomes after group I mGluR stimulation. Our findings show that (S)-3,5-dihydroxyphenylglycine (DHPG) increases significantly the activation of mTOR and p70S6K (Thr389, controlled by mTOR) in both brain areas. The mTOR activation is dose-dependent and requires the stimulation of mGluR1 subtype receptors as for the p70S6K activation observed in striatum and hippocampus. In addition, the p70S6K (Thr421/Ser424) activation via the ERK1/2 activation is increased and involved also mGluR1 receptors. These results demonstrate that group I mGluRs are coupled to mTOR-p70S6K and ERK1/2-p70S6K pathways in striatal and hippocampal synaptoneurosomes. The translational factor p70S6K could be involved in the group I mGluRs-modulated synaptic efficacy.  相似文献

Thyrotropin (TSH) and the gonadotropins; follitropin (FSH), lutropin (LH) and human chorionic gonadotropin (hCG) are a family of heterodimeric glycoprotein hormones. These hormones composed of two noncovalently linked subunits; a common alpha and a hormone specific beta subunits. Assembly of the subunits is vital to the function of these hormones. However, genetic fusion of the alpha and beta subunits of hFSH, hCG and hTSH resulted in active polypeptides. The glycoprotein hormone subunits contain one (TSH and LH) or two (alpha, FSHbeta and hCGbeta) asparagine-linked (N-linked) oligosaccharides. CGbeta subunit is distinguished among the beta subunits because of the presence of a carboxyl-terminal peptide (CTP) bearing four O-linked oligosaccharide chains. To examine the role of the oligosaccharide chains on the structure-function of glycoprotein hormones, chemical, enzymatic and site-directed mutagenesis were used. The results indicated that O-linked oligosaccharides play a minor role in receptor binding and signal transduction of the glycoprotein hormones. In contrast, the O-linked oligosaccharides are critical for in vivo half-life and bioactivity. Ligation of the CTP bearing four O-linked oligosaccharide sites to different proteins, resulted in enhancing the in vivo bioactivity and half-life of the proteins. The N-linked oligosaccharide chains have a minor role in receptor binding of glycoprotein hormones, but they are critical for bioactivity. Moreover, glycoprotein hormones lacking N-linked oligosaccharides behave as antagonists. In conclusion, the O-linked oligosaccharides are not important for in vitro bioactivity or receptor binding, but they play an important role in the in vivo bioactivity and half-life of the glycoprotein hormones. Addition of the O-linked oligosaccharide chains to the backbone of glycoprotein hormones could be an interesting strategy for designing long acting agonists of glycoprotein hormones. On the other hand, the N-linked oligosaccharides are not important for receptor binding, but they are critical for bioactivity of glycoprotein hormones. Deletion of the N-linked oligosaccharides resulted in the development of glycoprotein hormone antagonists. In the case of hTSH, development of an antagonist may offer a novel therapeutic strategy in the treatment of thyrotoxicosis caused by Graves' disease and TSH secreting pituitary adenoma.  相似文献

Fine mapping of quantitative trait loci (QTL) associated with resistance to the gastrointestinal parasite Heligmosomoides polygyrus was achieved on F6/F7 offspring (1076 mice) from resistant (SWR) and susceptible (CBA) mouse strains by selective genotyping (top and bottom 20% selected on total worm count in week 6). Fecal egg counts were recorded at weeks 2, 4, and 6, and the average was also analyzed. Blood packed cell volume in weeks 3 and 6 and five immunological traits (mucosal mast cell protease 1, granuloma score, IgG1 against adult worm, IgG1, and IgE to L4 antigen) were also recorded. On Chromosome 1 single-trait analyses identified a QTL with effects on eight traits located at about 24 cM on the F2 mouse genome database (MGD) linkage map, with a 95% confidence interval (CI) of 20-32 cM established from a multitrait analysis. On Chromosome 17 a QTL with effects on nine traits was located at about 18 cM on the MGD map (CI 17.9-18.4 cM). Strong candidate genes for the QTL position on Chromosome 1 include genes known to be involved in regulating immune responses and on Chromosome 17 genes within the MHC, notably the Class II molecules and tumor necrosis factor.  相似文献

Synaptobothrioides sardinellae n. g., n. sp. is described from Sardinella melanura off Fiji. The new genus differs from other plerurine hemiurid genera in having entire or angular, rather than lobed to digitiform, vitelline masses. It appears closest to Synaptobothrium, but differs in the lack of a presomatic pit and the presence of tegumental plications. Lecithochirium kawakawa from Euthynnus affinis and Thulinia microrchis from Siganus spinus are reported from off Fiji.  相似文献

Tight junction (TJ) permeability responds to changes of extracellular Ca(2+) concentration. This can be gauged through changes of the transepithelial electrical conductance (G) determined in the absence of apical Na(+). The early events of TJ dynamics were evaluated by the fast Ca(2+) switch assay (FCSA) (Lacaz-Vieira, 2000), which consists of opening the TJs by removing basal calcium (Ca(2+)(bl)) and closing by returning Ca(2+)(bl) to normal values. Oscillations of TJ permeability were observed when Ca(2+)(bl) is removed in the presence of apical calcium (Ca(2+)(ap)) and were interpreted as resulting from oscillations of a feedback control loop which involves: (a) a sensor (the Ca(2+) binding sites of zonula adhaerens), (b) a control unit (the cell signaling machinery), and (c) an effector (the TJs). A mathematical model to explain the dynamical behavior of the TJs and oscillations was developed. The extracellular route (ER), which comprises the paracellular space in series with the submucosal interstitial fluid, was modeled as a continuous aqueous medium having the TJ as a controlled barrier located at its apical end. The ER was approximated as a linear array of cells. The most apical cell is separated from the apical solution by the TJ and this cell bears the Ca(2+) binding sites of zonula adhaerens that control the TJs. According to the model, the control unit receives information from the Ca(2+) binding sites and delivers a signal that regulates the TJ barrier. Ca(2+) moves along the ER according to one-dimensional diffusion following Fick's second law. Across the TJ, Ca(2+) diffusion follows Fick's first law. Our first approach was to simulate the experimental results in a semiquantitative way. The model tested against experiment results performed in the frog urinary bladder adequately predicts the responses obtained in different experimental conditions, such as: (a) TJ opening and closing in a FCSA, (b) opening by the presence of apical Ca(2+) and attainment of a new steady-state, (c) the escape phase which follows the halt of TJ opening induced by apical Ca(2+), (d) the oscillations of TJ permeability, and (e) the effect of Ca(2+)(ap) concentration on the frequency of oscillations.  相似文献

The new species Diploproctodaeum arothroni (Lepocreadiidae: Diploproctodaeinae) is described from the tetraodontid fish Arothron hispidus from Fiji. It differs from its closest relatives, D. hakofugu, D. oviforme and D. momoaafata, in having 42–54 ovarian lobes, a vitellarium reaching to the pharynx and tegumental spination reaching almost to the posterior extremity. A revised key to the 17 recognised species of Diploproctodaeum is given.  相似文献
