The characteristics of signal PII proteins identified in bacteria, archaea, and plants are presented. Special attention is paid to the properties and functions of PII proteins in microorganisms with oxygenic photosynthesis type: cyanobacteria and unicellular green algae. The structure and regulation of PII proteins are reviewed. The mechanisms of interactions of PII proteins with effectors and cellular targets are analyzed in detail. The conservative properties and unique characteristics of proteins from representatives of pro-and eukaryotic phototrophs are discussed.  相似文献

A monoclonal antibody that can immunoprecipitate proteins containing phosphotyrosine has been isolated and characterized. To identify proteins that can act as substrates for tyrosine kinases in intact cells, extracts of phosphate-labeled NIH cells that had been treated with the phosphotyrosyl phosphatase inhibitor, vanadate, were precipitated with the antibody, and the immunoprecipitates were analyzed by two-dimensional gel electrophoresis. Numerous proteins were specifically precipitated from vanadate-treated NIH 3T3 cells by the antibody. The high level of phosphotyrosine detected in vanadate-treated cells is presumably primarily due to phosphatase inhibition, but approx. 2-fold increased tyrosine kinase activities were also detected in extracts of the cells after treatment with vanadate. The enhanced tyrosine kinase activity may contribute to the generation of the transformed phenotype seen in response to treatment with vanadate.  相似文献

Summary A method is presented for the preparation of immune sera and detection by immunofluorescence ofC. immitis, S. schenckii, B. dermatitidis, C. neoformans, andC. albicans in surgical and autopsy material. Formalin fixation does not affect the antigens of the mycotic agents. There are no cross reactions except withC. immitis andC. neoformans, which can be differentiated by the site of the specific fluorescence in each organism.  相似文献

Regulation of mRNA stability and translation plays a critical role in determining protein abundance within cells. Processing bodies (P‐bodies) are critical regulators of these processes. Here, we report that the Pim1 and 3 protein kinases bind to the P‐body protein enhancer of mRNA decapping 3 (EDC3) and phosphorylate EDC3 on serine (S)161, thereby modifying P‐body assembly. EDC3 phosphorylation is highly elevated in many tumor types, is reduced upon treatment of cells with kinase inhibitors, and blocks the localization of EDC3 to P‐bodies. Prostate cancer cells harboring an EDC3 S161A mutation show markedly decreased growth, migration, and invasion in tissue culture and in xenograft models. Consistent with these phenotypic changes, the expression of integrin β1 and α6 mRNA and protein is reduced in these mutated cells. These results demonstrate that EDC3 phosphorylation regulates multiple cancer‐relevant functions and suggest that modulation of P‐body activity may represent a new paradigm for cancer treatment.  相似文献

We present a model on plant-deer climate interactions developed for improving our understanding of the temporal dynamics of deer abundance and, in particular, how intrinsic (density-dependent) and extrinsic (plants, climate) factors influence these dynamics. The model was tested statistically by analysing the dynamics of five Norwegian red deer populations between 1964 and 1993. Direct and delayed density-dependence significantly influenced the development of the populations: delayed density-dependence primarily operated through female density, whereas direct density-dependence acted through both female and male densities. Furthermore, population dynamics of Norwegian red deer were significantly affected by climate (as measured by the global weather phenomenon, the North Atlantic Oscillation: NAO). Warm, snowy winters (high NAO) were associated with decreased deer abundance, whereas the delayed (two-year) effect of warm, snowy winters had a positive effect on deer abundance. Our analyses are argued to have profound implications for the general understanding of climate change and terrestrial ecosystem functioning.  相似文献
