In many marine invertebrates, long‐distance dispersal is achieved during an extended pelagic larval phase. Although such dispersal should result in high gene flow over broad spatial scales, fine‐scale genetic structure has often been reported, a pattern attributed to interfamilial variance in reproductive success and limited homogenization during dispersal. To examine this hypothesis, the genetic diversity of dispersing larvae must be compared with the postdispersal stages, that is benthic recruits and adults. Such data remain, however, scarce due to the difficulty to sample and analyse larvae of minute size. Here, we carried out such an investigation using the marine gastropod Crepidula fornicata. Field sampling of three to four larval pools was conducted over the reproductive season and repeated over 3 years. The genetic composition of larval pools, obtained with 16 microsatellite loci, was compared with that of recruits and adults sampled from the same site and years. In contrast to samples of juveniles and adults, large genetic temporal variations between larval pools produced at different times of the same reproductive season were observed. In addition, full‐ and half‐sibs were detected in early larvae and postdispersal juveniles, pointing to correlated dispersal paths between several pairs of individuals. Inbred larvae were also identified. Such collective larval dispersal was unexpected given the long larval duration of the study species. Our results suggest that each larval pool is produced by a small effective number of reproducers but that, over a reproductive season, the whole larval pool is produced by large numbers of reproducers across space and time.  相似文献

Parthenium hysterophorus L. (Asteraceae) is a weed of national significance in Australia. Among the several arthropod agents introduced into Australia to control populations of P. hysterophorus biologically, Epiblema strenuana Walker (Lepidoptera: Tortricidae) is the most widespread and abundant agent. By intercepting the normal transport mechanisms of P. hysterophorus, the larvae of E. strenuana drain nutrients, other metabolic products, and energy, and place the host plant under intense metabolic stress. In this study, determinations of total non‐structural carbohydrates (TNC) levels and carbon and nitrogen isotope ratios of fixed products in different parts of the plant tissue, including the gall, have been made to establish the function of gall as a sink for the nutrients. Values of δ13C and δ15N in galls were significantly different than those in proximal and distal stems, whereas the TNC levels were insignificant, when measured in the total population of P. hysterophorus, regardless of plant age. However, carbon, nitrogen, and TNC signatures presented significant results, when assayed in different developmental stages of P. hysterophorus. Carbon isotope ratios in galls were consistently more negative than those from the compared plant organs. Nitrogen isotope ratios in galls, on the contrary, were either similar to or less negative than the compared plant organs, especially within a single host‐plant stage population (i.e., either rosette, preflowering, or flowering stage). TNC levels varied within compared plant populations. The stem distal to the gall functioned more efficiently as a nodal channel than the stem proximal to the gall, especially in the translocation of nitrogenous nutrients. Our findings indicate that the gall induced by E. strenuana functions as a sink for the assayed nutrients, although some variations have been observed in the patterns of nutrient mobilization. By creating a sink for the nutrients in the gall, E. strenuana is able to place the overall plant metabolism under stress, and this ability indicates E. strenuana has the necessary potential for use as a biological‐control agent.  相似文献

Novel microbial cultivation platforms are of increasing interest to researchers in academia and industry. The development of materials with specialized chemical and geometric properties has opened up new possibilities in the study of previously unculturable microorganisms and has facilitated the design of elegant, high-throughput experimental set-ups. Within the context of the international Genetically Engineered Machine (iGEM) competition, we set out to design, manufacture, and implement a flow device that can accommodate multiple growth platforms, that is, a silicon nitride based microsieve and a porous aluminium oxide based microdish. It provides control over (co-)culturing conditions similar to a chemostat, while allowing organisms to be observed microscopically. The device was designed to be affordable, reusable, and above all, versatile. To test its functionality and general utility, we performed multiple experiments with Escherichia coli cells harboring synthetic gene circuits and were able to quantitatively study emerging expression dynamics in real-time via fluorescence microscopy. Furthermore, we demonstrated that the device provides a unique environment for the cultivation of nematodes, suggesting that the device could also prove useful in microscopy studies of multicellular microorganisms.  相似文献

Introduction

Both nonclassical and intermediate monocytes have been implicated in different inflammatory conditions. We hypothesized that these monocytes would increase during pregnancy, a condition associated with generalized activation of inflammatory responses and that they would increase even more during preeclampsia, in which inflammatory responses are further stimulated. In the present study we investigated changes in monocyte subsets during healthy pregnancy and preeclampsia in humans and rats.

Methods

Blood monocyte subsets of nonpregnant, preeclamptic and healthy pregnant women were identified with CD14 and CD16. In nonpregnant and pregnant rats, blood monocytes were identified with CD172a and CD43, as well as in rats infused with adenosine triphosphate (ATP), a pro-inflammatory stimulus known to induce preeclampsia-like symptoms. Total and CD206-positive macrophages were quantified in placentas of these animals.

Results

Lower percentages of classical monocytes were found in pregnant women (91%–[83–98%]) compared to nonpregnant women (94%–[90–98%]) and even less in preeclamptic patients (90%–[61–92%]). In contrast, the percentage of combined nonclassical/intermediate monocytes was higher in pregnant women (8.5%–[2.3–16.6%] vs. 5.6%–[1.9–9.5%]) and even higher in preeclamptic patients (9.9%–[7.8–38.7%]), which was caused by a selective increase of intermediate monocytes. In rats, we also found lower percentages of classical monocytes and higher percentages of nonclassical monocytes in pregnant versus nonpregnant rats. ATP infusion increased the percentage of nonclassical monocytes in pregnant rats even further but not in nonpregnant rats. These nonclassical monocytes showed a more activated phenotype in pregnant ATP-infused rats only. Mesometrial triangles of ATP-infused rats had less CD206-positive macrophages as compared to those of saline-infused rats.

Conclusion

The higher percentage of nonclassical/intermediate monocytes found in pregnancy and preeclampsia confirms their association with inflammatory responses. The observation that ATP stimulated numbers/activation of nonclassical monocytes in pregnant rats only, suggests that nonclassical monocytes are specifically altered in pregnancy and may play a role in the pathophysiology of preeclampsia.  相似文献

Riparian ecosystems are among the most degraded systems in the landscape, and there has been substantial investment in their restoration. Consequently, monitoring restoration interventions offers opportunities to further develop the science of riparian restoration, particularly how to move from small‐scale implementation to a broader landscape scale. Here, we report on a broad range of riparian revegetation projects in two regions of south‐western Victoria, the Corangamite and Glenelg‐Hopkins Catchment Management Areas. The objectives of restoration interventions in these regions have been stated quite broadly, for example, to reinstate terrestrial habitat and biodiversity, control erosion and improve water quality. This study reports on tree and shrub composition, structure and recruitment after restoration works compared with remnant vegetation found regionally. Within each catchment, a total of 57 sites from six subcatchments were identified, representing three age‐classes: <4, 4–8 and >8–12 years after treatment, as well as untreated (control) sites. Treatments comprised fencing to exclude stock, spraying or slashing to reduce weed cover, followed by planting with tube stock. Across the six subcatchments, 12 reference (remnant) sites were used to provide a benchmark for species richness, structural and recruitment characteristics and to aid interpretation of the effects of the restoration intervention. Vegetation structure was well developed in the treated sites by 4–8 years after treatment. However, structural complexity was higher at remnant sites than at treated or untreated sites due to a higher richness of small shrubs. Tree and shrub recruitment occurred in all remnant sites and at 64% of sites treated >4 years ago. Most seedling recruitment at treatment sites was by Acacia spp. This assessment provides data on species richness, structure and recruitment characteristics following restoration interventions. Data from this study will contribute to longitudinal studies of vegetation processes in riparian landscapes of south‐western Victoria.  相似文献

Although a series of signaling cascades involved in spatial memory have been identified, their link to spatial memory and strain-dependent expression has not been reported so far. Hippocampal levels of the abovementioned signaling proteins were determined in laboratory inbred strain C57BL/6J, the wild-derived inbred strain PWD/PhJ and the wild caught mouse Apodemus sylvaticus (AS) by immunoblotting. The resulting hippocampal protein levels were correlated with results from MWM. Hippocampal signaling protein (hSP) levels were tested also in yoked controls. Within-strain comparison between trained and yoked controls revealed significant differences between levels of Phospho-CaMKII (alpha), Phospho-CREB, Egr-1, c-Src, Phospho-ERK5, Phospho-MEK5 and NOS1 in all of the three strains tested. In addition, the three strains revealed different involvement of individual hSP levels clearly indicating that individual mouse strains were linked to individual hSPs in spatial memory. Phospho-ERK5 levels were not detectable in hippocampi of yoked controls of each strain. We learn from this study that a series of hSPs are associated with spatial memory and that different hSPs are linked to spatial memory in different strains that show different outcome in the MWM. Even correlational patterns in the individual hSPs differed between mouse strains. This is of importance for the interpretation of previous studies on the abovementioned signaling cascades as well as for the design of future studies on these hippocampal proteins. It is intriguing that individual mouse strains, laboratory or wild caught, may use different signaling pathways for spatial memory in the Morris water maze.  相似文献

The evolutionary function and maintenance of variation in animal personality is still under debate. Variation in the size of metabolic organs has recently been suggested to cause and maintain variation in personality. Here, we examine two main underlying notions: (i) that organ sizes vary consistently between individuals and cause consistent behavioural patterns, and (ii) that a more exploratory personality is associated with reduced survival. Exploratory behaviour of captive red knots (Calidris canutus, a migrant shorebird) was negatively rather than positively correlated with digestive organ (gizzard) mass, as well as with body mass. In an experiment, we reciprocally reduced and increased individual gizzard masses and found that exploration scores were unaffected. Whether or not these birds were resighted locally over the 19 months after release was negatively correlated with their exploration scores. Moreover, a long-term mark–recapture effort on free-living red knots with known gizzard masses at capture confirmed that local resighting probability (an inverse measure of exploratory behaviour) was correlated with gizzard mass without detrimental effects on survival. We conclude that personality drives physiological adjustments, rather than the other way around, and suggest that physiological adjustments mitigate the survival costs of exploratory behaviour. Our results show that we need to reconsider hypotheses explaining personality variation based on organ sizes and differential survival.  相似文献

The quinone secretions of 6 Tribolium species were analyzed qualitatively and quantitatively by gas chromatography. Procedures were developed for collecting and weighing the secretion which prevented its contamination and oxidation of the quinones. Secretions of all species contained 2-methyl- and 2-ethyl-1,4-benzoquinones in amounts ranging from 50.5% in T. brevicornis to 73% in T. confusum. Ratios of 2-methyl- to 2-ethyl-1,4-benzoquinone were 1:1.7 in 2 species and 1:2.6 to 1:2.7 in 4 species. The most prevalent non-quinone in secretions of all species was 1-pentadecene, varying from 15.6% in T. confusum to 19.9% in T. brevicornis. Three unidentified compounds, probably long-chain hydrocarbons, were found and designated biosolvents. BS-2 was present in secretions of all species, BS-1a in 4 species and traces of BS-1b in 2 species. The primary function of 1-pentadecene, and probably the unidentified biosolvents, is quinone solvency. This is discussed in relation to other previously proposed functions and to the recently discovered activity as a sex pheromone.  相似文献

Demographic rates such as recruitment and survival probability can vary considerably among populations of the same species due to variation in underlying environmental processes. If environmental processes are spatially correlated, nearby populations are expected to have more similar demographic rates than those further apart. Breeding populations and foraging ranges are spatially segregated in colonial seabirds, making them ideal for studying spatial patterns in demographic rates and their effects on local population dynamics. Here we explored variation in age-dependent survival probabilities across 14 colonies of Herring Gulls Larus argentatus breeding along the Dutch North Sea coast. We used long-term mark–recapture data of marked fledglings to estimate survival, and estimated spatial autocorrelation of survival probabilities. We assessed whether survival until recruitment age or until 10 years old (close to their expected lifespan) explained variation in population trajectories of each colony. Juvenile and adult survival showed a strong, but different, north-to-south gradient in survival probability, with lower juvenile but higher adult survival in northern colonies than southern colonies, whereas the spatial pattern of immature survival was less distinct. Neither recruitment nor the proportion of 10-year-old adults alive predicted whether a colony collapsed, declined, remained stable or increased. The distinct spatial pattern in survival suggests variation in regional food availability, which do not seem to drive local population dynamics. The absence of a link between survival and colony trajectories implies that connectivity between populations plays an important role affecting population dynamics.  相似文献
