A procedure is described for the extensive purification of hydroxypyruvate:l-alanine transaminase from rabbit liver. On the basis of gel filtration studies, the molecular weight of the enzyme is estimated to be about 41,000 daltons. A similar value was obtained when the enzyme was subjected to gel electrophoresis in the presence of sodium dodecyl sulfate indicating that the enzyme consists of a single polypeptide chain.The purified enzyme catalyzes the transamination of glyoxylate as well as hydroxypyruvate with l-alanine as the preferred amino donor for both substrates. The two enzymatic activities were not separated during purification nor by Chromatographic or electrophoretic procedures. Kinetic studies demonstrated that the two α-keto acids are competitive substrates. The above data are consistent with the fact that a single enzyme catalyzes the transamination of both glyoxylate and hydroxypyruvate. The effects of various inhibitors on enzymatic activity were investigated. The enzyme is inhibited by glyceraldehyde-3-phosphate and other aldehydes.The possible role of hydroxypyruvate:l-alanine transaminase in gluconeogenesis is discussed.  相似文献

Antibacterial peptides are important components of the innate immune system, used by the host to protect itself from different types of pathogenic bacteria. Over the last few decades, the search for new drugs and drug targets has prompted an interest in these antibacterial peptides. This study investigated the effects of expectancies concerning the controllability of outcomes in a biofeedback task. Forty-eight college undergraduates were subjects. Frontal (forehead) electromyographic (EMG) responses were measured during baseline (no treatment), pretreatment, and test sessions. During pretreatment, subjects were assigned to one of three groups. Using a fictitious blood vessel control task, the success group received false feedback and instructions that conveyed that outcomes had been successfully controlled. The failure group received feedback and instructions that conveyed that outcomes had not been controlled. A control group was given no specific task. During the subsequent test stage, all subjects were asked to reduce frontal muscle tension levels through relaxation while assisted with true EMG feedback. Relative to the success and control groups, EMG levels of the failure group reflected more rapid acquisition of frontal muscle relaxation. However, performance on a cognitive task was not affected by the pretreatment. It was concluded that expectancies generated during the false feedback pretreatment were related to later biofeedback performance. The results were discussed in terms of concepts of locus of control and theories of learned helplessness and reactance.A version of this paper was presented at the annual meeting of the Biofeedback Society of America, San Diego, 1979.  相似文献

The renal hemodynamic and tubular effects of ANF were investigated using the Sperber technique in chickens. This technique takes advantage of the unique portal circulation of the avian kidney and permits direct access to the renal peritubular space independent of renal arterial blood flow and glomerular filtration. Infusion of ANF into the avian renal portal system increased urine flow rate and sodium excretion by as much as 300% and 100%, respectively. These changes occurred in the absence of significant alterations in glomerular filtration rate or renal plasma flow. There was no significant difference in urine flow, sodium excretion or glomerular filtration rate between the ANF-infused kidney and the contralateral, non-infused kidney. We conclude that the diuretic and natriuretic effects of ANF do not depend on changes in glomerular filtration rate and that the site of action of ANF is the renal medulla.  相似文献
