Familial hypercholesterolemia (FH) (OMIM 143890) is an autosomal dominantly inherited disease mainly caused by mutations of the gene encoding the low density lipoprotein receptor (LDLR) and Apolipoprotein (Apo) B. First the common mutation R3500Q in ApoB gene was determined using PCR/RFLP method. Then the LDLR gene was screened for mutations using Touch-down PCR, SSCP and sequencing techniques. Furthermore, the secondary structure of the LDLR protein was predicted with ANTHEPROT5.0. The R3500Q mutation was absent in these two families. A heterozygous p.W483X mutation of LDLR gene was identified in family A which caused a premature stop codon, while a homozygous mutation p.A627T was found in family B. The predicted secondary structures of the mutant LDLR were altered. We identified two known mutations (p.W483X, p.A627T) of the LDLR gene in two Chinese FH families respectively.  相似文献

Pollution from the oil shale dump in Maoming, Guangdong Province, China, was a major social problem due to the great amount of environmental damage it caused. Therefore, a great deal of attention needed to be paid for the ecological restoration and reconstruction. The objective of this study was to investigate the species composition of soil fauna and its diversity in oil shale dumps after the application of different ecological restoration schemes in order to understand the impact they had on ecological restoration. Three plots were selected on an oil shale dump near the city of Maoming. The “north plot” was a newly-planted young forest mixed with various tree species, while the “south plot” was a 20-year-old Acacia auriculaeformis forest, and the “control plot” was a 20-year-old naturally-recovering grassland. Soil animals, mainly including macro-meso groups, were collected by hand-sorting and Tullgren funnel extraction. They were then identified to family or genus level with only a small portion to order (e.g. Chllopoda) or species (e.g. Isopoda) level. The specimens obtained in the present study was 11164 individuals, belonging to 27 orders and 110 families or genera. The Shannon index (H′), DGs (based on species) and DGg (based on groups) were used to analyze the diversity of soil animals between different plots. The major results were as follows: A total of 33 families or genera belonging to nine orders were found in the “north plot”. The main group was Caritermes, accounting for 63.4% of the total, followed by Tetramorium with 21.3%. Hymenoptera, mainly Formicidae, had more genera than others, accounting for 80% of the total genera in this group. The diversity of soil animals in this plot was very low because the H′ index was only 1.2, while the DGs index was 4.0 and the DGg index was 1.3. A total of 61 families or genera belonging to 23 orders were found in the “south plot”. Malmcoangelia and Tetramorium were the main groups, which accounted for 60.3% and 10.2%, respectively. Two genera of Annelda and two genera of Isopoda only accounted for 2.6% and 1.9%, respectively, but they were considered to be major groups due to their large body sizes and the distinct characteristics of their habitat. Acarina had a greater number of individuals and families or genera with its individual number accounting for 67.5% of the total, and the number of families or genera of this group account for 70% in this plot. The diversity indexes (H′, DGs and DGg) in this plot were significantly higher than those in the “north plot”, and were 1.65, 16.7 and 7.75, respectively. In the “control plot”, there were 67 families or genera of soil animals belonging to 23 orders. The main groups were Tetramorium (20%), Lasius (17.1%), Bothriomymex of Formicidae (13.8%), and Malmcoangelia of oribatid mites (14.5%). Formicidae of Hymenoptera was the group with the maximum number of individuals, accounting for 51.0%, while Diplopoda had the most families or genera. The H′ and DGs indexes, being 2.54 and 17.7, were higher than those in the “south plot”, while the DGg index of 7.20 was lower than that in the “south plot”. The results showed that the species composition and diversity indexes were higher in the “south plot” than in the “north plot” and the “control plot”, which demonstrated that using Acacia auriculaeformis forest to restore the oil shale dump was an effective approach in terms of soil biodiversity.  相似文献

Plant defensins are small, basic, cysteine‐rich peptides, belonging to the antimicrobial peptide superfamily, commonly found in the plant kingdom. In this study, we cloned and characterized a plant defensin gene from Jatropha curcas (JcDef). JcDef carried conserved receptor binding sites and a cysteine motif, and it was phylogenetically grouped together with defensin Ec‐AMP‐D2‐like in Elaeis guineensis. JcDef is localized to cytoplasm and highly expressed in young tissues with fast metabolism such as cotyledons and stem apexes. Transgenic expression of JcDef in tobacco showed enhanced resistance against sheath blight disease caused by R. solani, indicating the antibacterial function.  相似文献

Five new guaiane sesquiterpenes, 1 – 5 , were isolated from the culture broth of the endophytic fungus Xylaria sp. YM 311647, isolated from Azadirachta indica A. Juss . The structures of these compounds were elucidated on the basis of spectroscopic analyses, and their inhibitory activities against five pathogenic fungi were evaluated. All guaiane sesquiterpenes showed moderate or weak antifungal activities in a broth microdilution assay.  相似文献

丁香酚是中药丁香油中的主要作用成份,它具有多种复杂的药理作用,尤其在解热降温方面有显著作用.本研究室以往的电生理实验观察到,丁香酚可反转致热原作用下的PO/AH(视前区-下丘脑前部)温度敏感神经元的放电活动,说明丁香酚的解热作用是通过直接或间接地影响PO/AH神经元的放电活动而实现的.近年来,弓状核在体温调节方面的作用倍受观注.本研究室多年的研究表明,弓状核积极地参与了体温调节活动,因为毁损弓状核后大鼠体温发生明显紊乱.本实验对PO/AH与弓状核在体温调节中的作用进行比较性研究,同时观察家兔发热以及丁香酚解热时在家兔弓状核及PO/AH中PGE2和cAMP含量的变化.  相似文献

多态性蛋白Mad2是有丝分裂纺锤体检测点(SAC)的关键蛋白,也是多态性蛋白质家族中研究最广泛的成员之一.Mad2有两种不同的天然构象:O-Mad2和C-Mad2.Mad2构象间的转变及其与配体Cdc20间的相互作用对SAC发挥其生物学功能至关重要.本文利用荧光各向异性技术对O-Mad2和C-Mad2与配体TAMRA-Cdc20~(121-138)间相互作用的热力学及动力学过程进行了系统表征.结果表明:在无盐和低盐溶液(100 mmol/L NaCl)中,Mad2两种构象与Cdc20~(121-138)的平衡解离常数(K_D)均在10~(-6) mol/L,但C-Mad2与Cdc20~(121-138)结合的K_D值约为O-Mad2的1/5;在高盐(300 mmol/L NaCl)溶液中,Mad2两种构象与TAMRA-Cdc20~(121-138)结合的K_D值无明显差别.动力学实验结果显示,在同一种缓冲液中Mad2两种构象与Cdc20~(121-138)相互作用的解离速率常数k_d没有显著差别,而C-Mad2与Cdc20~(121-138)的结合速率常数k_a却比O-Mad2高一个数量级,这表明C-Mad2与Cdc20~(121-138)不仅结合力更强,且结合速率要快很多.Mad2与Cdc20~(121-138)突变体间的相互作用以及离子强度对二者相互作用的影响结果提示,Mad2和Cdc20间的相互作用不是通过静电相互作用,而可能是通过疏水相互作用来实现的.本研究为揭示多态性蛋白Mad2的构象转变机理及其在有丝分裂过程中的作用机制提供了重要的实验基础.  相似文献

Linear cationic α-helical antimicrobial peptides are referred to as one of the most likely substitutes for common antibiotics, due to their relatively simple structures (≤ 40 residues) and various antimicrobial activities against a wide range of pathogens. Of those, HP(2-20) was isolated from Helicobacter pylori ribosomal protein. To reveal a mechanical determinant that may mediate the antimicrobial activities, we examined the mechanical properties and structural stabilities of HP(2-20) and its four analogues of same chain length by steered molecular dynamics simulation. The results indicated the following: the resistance of H-bonds to the tensile extension mediated the early extensive stage; with the loss of H-bonds, the tensile force was dispensed to prompt the conformational phase transition; and Young's moduli (N/m(2)) of the peptides were about 4 ~ 8 × 10(9). These mechanical features were sensitive to the variation of the residue compositions. Furthermore, we found that the antimicrobial activity is rigidity-enhanced, that is, a harder peptide has stronger antimicrobial activity. It suggests that the molecular spring constant may be used to seek a new structure-activity relationship for different α-helical peptide groups. This exciting result was reasonably explained by a possible mechanical mechanism that regulates both the membrane pore formation and the peptide insertion.  相似文献

Deleted in liver cancer-1 (DLC-1), encoding a Rho GTPase-activating protein (GAP), is considered as a promising candidate tumor suppressor gene in nasopharyngeal carcinoma (NPC). The single-nucleotide polymorphism (SNP) -29A/T upstream of ATG start codon was found when gene mutation profile of DLC-1 in NPC was analyzed. To evaluate the correlation between SNP -29A/T in the promoter region of DLC-1 gene and risk of NPC, a total of 521 samples from a Chinese population, including 320 healthy individuals and 201 NPC patients, were collected for SNP analysis by PCR-single-strand conformation polymorphism and sequencing. The differences in allele and genotype frequencies between NPC patients and controls were tested using logistic regression statistical method. No significant differences were found in allele or genotype frequencies between NPC patients and controls or among different NPC clinical stages. Hence, our data indicate that the SNP -29A/T of DLC-1 gene is not associated with NPC susceptibility.  相似文献
