In addition to Rb and p53, a third cellular protein, p120 in monkey and p118 in human cells, forms a specific complex with SV40 large T antigen (T). p118/120 is not a product of the Rb-gene. As was shown with T/Rb complex formation, the interaction between T and p120 is dependent on the intact nature of a ten residue, transformation-controlling domain in T (residues 105-114). In mouse cells, a readily detectable protein of 115 kd was detected, which, like murine Rb, also forms a stable complex with T. Like p118/120, p115 binding is also dependent on the intact nature of the 105-114 sequence. Given their similar size and T antigen binding sequence dependence, p115 and p118/120 may be products of the same gene in different species. These results suggest that interactions between T and p115/118/120, as well as T and Rb, contribute to the SV40 transforming mechanism. 相似文献

Pitting wear is a dominant form of polyethylene surface damage in total knee replacements, and may originate from surface cracks that propagate under repeated tribological contact. In the present study, stress intensity factors, KI and KII, were calculated for a surface crack in a polyethylene-CoCr-bone system in the presence of rolling or sliding contact pressures. Variations in crack length and load location were studied to determine probable crack propagation mechanisms and modes. The crack tip experienced a wide range of mixed-mode conditions that varied as a function of crack length, load location, and sliding friction. Positive KI values were observed for shorter cracks in rolling contact and for all crack lengths when the sliding load moved away from the crack. KII was greatest when the load was directly adjacent to the crack (g/a = +/- 1), where coincidental Mode I stresses were predominantly compressive. Sliding friction substantially increased both KImax and KIImax. The effective Mode I stress intensity factors, Keff, were greatest at g/a = +/- 1, illustrating the significance of high shear stresses generated by loads adjacent to surface cracks. Keff trends suggest mechanisms for surface pitting by which surface cracks propagate along their original plane under repeated reciprocating rolling or sliding, and turn in the direction of sliding under unidirectional sliding contact. 相似文献

The plasma noradrenaline (NA) and adrenaline (A) concentration responses of seven young male athletes [15 (SD 1) years] and
seven adult male athletes [25 (SD 6) years] were investigated together with the serum testosterone (Tes) concentration responses
in four different half-squatting exercises. The loads, number of repetitions, exercise intensity and recovery between the
sets were manipulated such that different types of metabolic demand could be expected. However, the amount of work done was
kept equal in each kind of exercise. After the most exhausting unit of exercise (E3; two sets of 30 repetitions with 50% of
1 repetition maximum and with 2-min recovery between the sets) the plasma NA concentration was significantly lower in the
younger than in the adult subjects [15.7 (SD 7.8) vs 32.7 (SD 13.2) nmol · l−1, P < 0.05], while the A concentrations were similar. In the other three exercises no differences in the plasma catecholamine
concentration responses among the groups were observed. The postexercise Tes concentrations, however, were significantly lower
in the younger than in the adult subjects in every exercise unit. No correlations between the plasma catecholamine and serum
Tes concentration responses were observed in any of the exercise units in either group. The results of the present study may
suggest reduced sympathetic nervous activity in the younger subjects compared to the adults in response to exhausting resistance
exercise. The results may also suggest that the catecholamines were less involved in eliciting an increase in Tes secretion
in these resistance exercises.

Southern Hemisphere passerines are generally thought to have long breeding seasons and high annual survivorship, but this may reflect a bias resulting from a disproportionate concentration on sedentary species. This study presents a detailed examination of the breeding biology and social organization of a migratory population of a passerine from the Southern Hemisphere – the Yellow‐faced Honeyeater Lichenostomus chrysops. It took place between 1997 and 2000 in the Coranderrk Reserve, Healesville, Australia. Following the birds’ return from migration, breeding commenced in mid‐November, roughly 2 months after arrival. The breeding season was short (3.5–4 months) compared with sedentary honeyeaters occupying the same habitat. Territories (0.19 ± 0.11 ha) were established by males but defended by both sexes against conspecific and heterospecific intruders. Within a breeding season, pairs were multibrooded (mean no. of clutches per season 2.14 ± 0.76, range 1–3) and always re‐nested within the same territory with the same partner. However, only 12.5% of pairs bred together in a subsequent breeding season despite, on many occasions, their partner from the previous season being alive (divorce rate of 75% for males and 66% for females). Females alone built the nest, incubated the eggs and brooded the nestlings. However, males contributed 44.5% of feeding visits to nestlings. The breeding cycle of Yellow‐faced Honeyeaters (clutch size 1–3 eggs, mean 2.4 ± 0.6, n = 84, incubation period 14.4 ± 0.7 days, n = 11, nestling period 13.0 ± 1.7 days, n = 6) was similar to that of sedentary honeyeaters (and Southern Hemisphere Corvidae in general), except that fledglings only remained on the parental territory for 2–3 weeks post‐fledging. At least one young fledged from 32.1% of nests (n = 156). Predation was the major cause of nest failure (74.5% of 106 failed nests). Only 4.4% of nestlings (n = 136) were observed on the study site in subsequent seasons (five males and one female). None of those males was observed on the study site until their second year of life. Adult annual survivorship (ignoring losses due to dispersal) was 43% for females and 51% for males. Our study suggests that if more data were available for the numerous migratory species from the Southern Hemisphere, some apparent differences from northern hemisphere passerines might disappear. 相似文献

Climate change is a major threat to species worldwide, yet it remains uncertain whether tropical or temperate species are more vulnerable to changing temperatures. To further our understanding of this, we used a standardised field protocol to (1) study the buffering ability (ability to regulate body temperature relative to surrounding air temperature) of neotropical (Panama) and temperate (the United Kingdom, Czech Republic and Austria) butterflies at the assemblage and family level, (2) determine if any differences in buffering ability were driven by morphological characteristics and (3) used ecologically relevant temperature measurements to investigate how butterflies use microclimates and behaviour to thermoregulate. We hypothesised that temperate butterflies would be better at buffering than neotropical butterflies as temperate species naturally experience a wider range of temperatures than their tropical counterparts. Contrary to our hypothesis, at the assemblage level, neotropical species (especially Nymphalidae) were better at buffering than temperate species, driven primarily by neotropical individuals cooling themselves more at higher air temperatures. Morphology was the main driver of differences in buffering ability between neotropical and temperate species as opposed to the thermal environment butterflies experienced. Temperate butterflies used postural thermoregulation to raise their body temperature more than neotropical butterflies, probably as an adaptation to temperate climates, but the selection of microclimates did not differ between regions. Our findings demonstrate that butterfly species have unique thermoregulatory strategies driven by behaviour and morphology, and that neotropical species are not likely to be more inherently vulnerable to warming than temperate species. 相似文献

Abstract The effects of infection by Salmonella enteritidis and S. typhimurium on the small and large intestines, liver, spleen and mesenteric nodules of rats were studied in vivo. Both Salmonella serotypes persisted and proliferated in the gastrointestinal tract and invaded sub-epithelial tissues, mainly the ileum, leading to the systemic distribution of these pathogens. Coincidental with the infection, the rate of crypt cell proliferation increased resulting in substantial growth of the small intestine. The extent of this and the accompanying accumulation of polyamines was particularly dramatic in the ileum where there was also some disruption of the villus epithelium. It is possible that these effects of the infection on the metabolism and morphology of the small bowel, which strongly resembled the changes induced by some plant lectins, may facilitate the colonisation and invasion of the gut by Salmonellae . 相似文献

Iqg1p is a component of the actomyosin contractile ring that is required for actin recruitment and septum deposition. Cells lacking Iqg1p function have an altered bud-neck structure and fail to form a functional actomyosin contractile ring resulting in a block to cytokinesis and septation. Here it is demonstrated that increased expression of the actin cytoskeleton associated protein Bsp1p bypasses the requirement for contractile ring function. This also correlates with reduced bud-neck width and remedial septum formation. Increased expression of this protein in a temperature-sensitive iqg1-1 background causes remedial septum formation at the bud neck that is reliant upon chitin synthase III activity and restores cell separation. The observed suppression correlates with a restoration of normal bud-neck structure. While Bsp1p is a component of the contractile ring, its recruitment to the bud neck is not required for the observed suppression. Loss of Bsp1p causes a brief delay in the redistribution of the actin cytoskeleton normally observed at the end of actin ring contraction. Compromise of Iqg1p function, in the absence of Bsp1p function, leads to a profound change in the distribution of actin and the pattern of cell growth accompanied by a failure to complete cytokinesis and cell separation. 相似文献

The enantiomerically pure alpha-methoxycarbaldehyde 3 was prepared from l-leucine in five steps and 31% overall yield. The aldehyde was subjected to a diastereoselective BF3-mediated crotylation with silane 4 and to various reagent-controlled addition reactions. The configuration of aldol addition products 19 and 20 was proven by single crystal X-ray crystallography. Based on these data spectral comparison allowed an unambiguous assignment of the desired anti,syn-crotylation product 2b and of the syn,syn-crotylation product 2a. Unexpectedly, product 2a of the BF3-mediated crotylation is formally the product of chelation-control.
