The hypothalamic peptide growth hormone-releasing factor (GRF) regulates the secretion and production of growth hormone from the anterior pituitary (M. C. Gelato and G. R. Merriam, Annu. Rev. Physiol. 48:569-591). To study GRF gene regulation, transgenic mice were generated that harbor the human GRF promoter fused to the coding sequences from the simian virus 40 early region. These mice had normal hypothalamic functions but unexpectedly suffered from severe thymic hyperplasia. Immunohistochemical analysis revealed that large T antigen was expressed in the thymic epithelial cells. These cells have endocrine properties and are known to produce thymic hormones [corrected]. The thymic hyperplasia was the apparent consequence of inappropriate production of T-cell maturation factors by epithelial cells and could involve increased self renewal of apparently normal T stem cells in the thymus.  相似文献

Mass spectrometric determinations of O2 affinities by the rumen fungus Neocallimastix patriciarum indicated a stable respiration under liquid phase O2 concentrations up to 10 M, the apparent K m for O2 under these conditions was 4.0 M. Exposure to O2 concentrations in excess of 10 M resulted in rapid inactivation of the observed respiration. Calculated H2 evolution rates for the organism are 8.1 nmol min-1 per mg of protein. Exposure to liquid-phase O2 concentrations in excess of 1.4 M caused 50% inhibition of H2 production. That superoxide and peroxide are amongst the products of respiration was shown by the use of ESR spectroscopy with the spin trapping agent 5,5-dimethyl-l-pyrroline-N-oxide. On 22 August 1985 a fire occurred on a Boeing 737 jet airliner at take off at Manchester Airport. One hundred and thirty seven passengers and crew were on board. Fifty two passengers died on the aircraft, 85 escaped. Most survivors had minor physical injuries, but 15 required admission to hospital because of smoke inhalation and two of these had severe burns. At presentation only one survivor required ventilation but within 12 hours a further five required ventilation. Although initially patients suffering from smoke inhalation may seem relatively well, lung function may deteriorate rapidly in the first 24 hours. Careful organisation and regular practice of procedures to deal with a major accident are essential to be able to respond adequately to such an event.  相似文献

In the course of discovering the first mutagen (X-rays) just over 60 years ago, Herman J. Muller asked whether X-rays induced single-gene mutations and/or chromosomal (multiple-gene) mutations. To a large extent, his question has set the agenda for mutagenesis research ever since. We explore historically the answers to this question, with special emphasis on recent developments in the field of mammalian cell mutagenesis. Studies indicate that ionizing radiation and many chemical mutagens/carcinogens induce both gene and chromosomal mutations; however, only certain genetic systems permit the recovery and analysis of both classes of mutations. Few chemical mutagens induce only gene mutations in mammalian cells; instead, most mutagens appear to induce both classes of mutations, with chromosomal mutations (especially multilocus deletions) predominating at high doses. These results have implications regarding the mechanisms of mutagenesis, the role of chromosomal mutations in carcinogenesis and hereditary disease, and the type of data required for risk assessment of physical and chemical mutagens/carcinogens.  相似文献

Use of tissue culture to bypass wheat hybrid necrosis   总被引:1,自引:0,他引:1  
Summary Hybrid necrosis in wheat is a barrier to gene transfer in wheat breeding practice. It is based on two complementary genes, Ne1 and Ne2. Recovery mutants (Re1, Re2 and Re3) which can grow to maturity were recovered from immature embryo cultures of necrotic hybrids between T. aestivum and T. durum. Cytological observation demonstrated that Re1 had 34 chromosomes instead of 35. This indicated that one of the chromosomes carrying the Ne genes was lost. Genetic study suggested that for Re1, the lost chromosome was chromosome 5B of the durum parental line. Re mutants are male sterile but can be maintained through a young ear culture method. Re mutants could be successfully pollinated by either parental line and the BC1 progeny is partially fertile. Re mutants were repeatedly induced in about 1% of the regenerated plants from immature embryo culture. This technique provides a practical way to bypass hybrid necrosis.  相似文献
