The majority of black Aspergilli (Aspergillus section Nigri), including Aspergillus niger, as well as many other Ascomycetes fail to germinate on d-galactose as a sole carbon source. Here, we provide evidence that the ability of A. niger to transport D-galactose is growth stage dependent, being absent in the conidiospores but present in the mycelia. Despite earlier claims, we could identify galactokinase activity in growing cells and all genes of the Leloir pathway (responsible for channelling D-galactose into the EMP pathway) are well induced on D-galactose (and also on lactose, D-xylose and L-arabinose) in the mycelial stage. Expression of all Leloir pathway genes was also detectable in conidiospores, although galE (encoding a galactokinase) and galD (encoding a galactose-1-phosphate uridylyl transferase) were expressed poorly. These results suggest that the D-galactose-negative phenotype of A. niger conidiospores may be due to the lack of inducer uptake.  相似文献

Members of the aldo–keto reductase family including aldose reductases are involved in antioxidant defense by metabolizing a wide range of lipid peroxidation-derived cytotoxic compounds. Therefore, we produced transgenic wheat genotypes over-expressing the cDNA of alfalfa aldose reductase gene. These plants consequently exhibit 1.5–4.3 times higher detoxification activity for the aldehyde substrate. Permanent drought stress was generated in the greenhouse by growing wheat plants in soil with 20 % water capacity. The control and stressed plants were monitored by a semi automatic phenotyping platform providing computer-controlled watering, digital and thermal imaging. Calculation of biomass values was based on the correlation (R 2 = 0.7556) between fresh weight and green pixel-based shoot surface area. The green biomass production by plants of the three transgenic lines was 12–26–41 % higher than the non-transgenic plants’ grown under water limitation. Thermal imaging of stressed non-transgenic plants indicated an elevation in the leaf temperature. The thermal status of transformants was similar at both normal and suboptimal water regime. In drought, the transgenic plants used more water during the growing season. The described phenotyping platform provided a comprehensive data set demonstrating the improved physiological condition of the drought stressed transgenic wheat plants in the vegetative growth phase. In soil with reduced water capacity two transgenic genotypes showed higher seed weight per plant than the control non-transgenic one. Limitation of greenhouse-based phenotyping in analysis of yield potential is discussed.  相似文献

The ARTEMIS Global Antifungal Susceptibility Program provides the collection of epidemiological data and the results of the fluconazole and voriconazole susceptibility testing of yeast isolates. Participating in this study, a total of 7318 clinical yeast isolates were tested from different geographical areas in Hungary in the period 2001 to 2003. The species isolated most frequently was C. albicans (68.8%), followed by C. glabrata (11.8%), C. tropicalis (5.7%) and C. krusei (4.6%). Isolates of C. albicans, C. kefyr, C. lusitaniae, C. tropicalis and C. parapsilosis were highly susceptible to fluconazole (78.9-100%). The rates of isolation of fluconazole-resistant C. glabrata and C. krusei were higher in our study than the global mean in 2001 (28.2% and 87.5% vs. 18.3% and 70.2%, respectively). Differences were detected in the distribution of fluconazole-susceptibility data of C. glabrata isolates in the different counties of Hungary: most of the resistant isolates were observed in the eastern part of the country.  相似文献

In the present study, white truffle ectomycorrhizae (EM) collected in deciduous forests (Populus, Quercus, and Fagus) from Hungary were characterized by morphological–anatomical and molecular methods. Our investigations suggest that the EM of white truffles (e.g., Tuber rapaeodorum, Tuber puberulum, Tuber rufum) are common and abundant members of the forest communities in the area. The ITS sequences of 14 EM specimens and 46 additional fruitbody sequences from the GenBank were clustered into four main groups in phylogenetic analyses. In the ITS-1 region, a characteristic indel pattern was found, which supports the clades. Although our analyses indicate definite genetic distance between the groups of the phylogenetic tree, these clades do not correspond to the traditional taxons identified by fruitbody characteristics. Comparison of the ectomycorrhizae shows that neither is mycorrhizal anatomy a good tool to separate the groups, because the characters (like the epidermoid or angular mantle structure, cell wall thickness, the sape and size of cystidia) are too variable and overlap between the clades. The interspecific similarity, observed both in ectomycorrhizal and fruitbody characters, strengthen the sensu lato morpho-species concept of this group. Our study, which combines comprehensive molecular and anatomical approach to characterize and identify ectomycorrhizae of white truffles from natural samples, stress out the need of the taxonomical revision of this group.  相似文献

Variable ventilation (VV), characterized by breath-to-breath variation of tidal volume (Vt) and breathing rate (f), has been shown to improve lung mechanics and blood oxygenation during acute lung injury in many species compared with conventional ventilation (CV), characterized by constant Vt and f. During CV as well as VV, the lungs of mice tend to collapse over time; therefore, the goal of this study was to develop a new VV mode (VV(N)) with an optimized distribution of Vt to maximize recruitment. Groups of normal and HCl-injured mice were subjected to 1 h of CV, original VV (VV(O)), CV with periodic large breaths (CV(LB)), and VV(N), and the effects of ventilation modes on respiratory mechanics, airway pressure, blood oxygenation, and IL-1beta were assessed. During CV and VV(O), normal and injured mice showed regional lung collapse with increased airway pressures and poor oxygenation. CV(LB) and VV(N) resulted in a stable dynamic equilibrium with significantly improved respiratory mechanics and oxygenation. Nevertheless, VV(N) provided a consistently better physiological response. In injured mice, VV(O) and VV(N), but not CV(LB), were able to reduce the IL-1beta-related inflammatory response compared with CV. In conclusion, our results suggest that application of higher Vt values than the single Vt currently used in clinical situations helps stabilize lung function. In addition, variable stretch patterns delivered to the lung by VV can reduce the progression of lung injury due to ventilation in injured mice.  相似文献

Summary In this paper, the connection between morphology, cephalosporin-C production and alternative respiration of Acremonium chrysogenum is examined. As demonstrated by chemostat experiments, the ratio of the filamentous and the yeast-like forms depended on the growth rate. The yeast-like form, but not the filamentous form exhibited cyanide-resistant alternative respiration. As a consequence, the yeast-like form was regarded to be more suitable for antibiotic overproduction.  相似文献

We have analysed the influence of human malignant, inflammatory and infectious disease on the capacity of peripheral lymphocytes to mediate antibody-dependent haemolysis. The application of an enzyme-like kinetic model for measurement of maximal cytotoxic function has permitted reproducible and sensitive determinations of the K-cell function.
