Iron regulatory proteins (IRPs), the cytosolic proteins involved in the maintenance of cellular iron homeostasis, bind to stem loop structures found in the mRNA of key proteins involved iron uptake, storage, and metabolism and regulate the expression of these proteins in response to changes in cellular iron needs. We have shown previously that HFE-expressing fWTHFE/tTA HeLa cells have slightly increased transferrin receptor levels and dramatically reduced ferritin levels when compared to the same clonal cell line without HFE (Gross et al., 1998, J Biol Chem 273:22068-22074). While HFE does not alter transferrin receptor trafficking or non-transferrin mediated iron uptake, it does specifically reduce (55)Fe uptake from transferrin (Roy et al., 1999, J Biol Chem 274:9022-9028). In this report, we show that IRP RNA binding activity is increased by up to 5-fold in HFE-expressing cells through the activation of both IRP isoforms. Calcein measurements show a 45% decrease in the intracellular labile iron pool in HFE-expressing cells, which is in keeping with the IRP activation. These results all point to the direct effect of the interaction of HFE with transferrin receptor in lowering the intracellular labile iron pool and establishing a new set point for iron regulation within the cell.  相似文献

There is increasing public, governmental and commercial interest in the welfare of intensively farmed fish and stocking density has been highlighted as an area of particular concern. Here we draw scientific attention and debate to this emerging research field by reviewing the evidence for effects of density on rainbow trout. Although no explicit reference to ‘welfare’ has been made, there are 43 studies which have examined the effects of density on production and physiological parameters of rainbow trout. Increasing stocking density does not appear to cause prolonged crowding stress in rainbow trout. However, commonly reported effects of increasing density are reductions in food conversion efficiency, nutritional condition and growth, and an increase in fin erosion. Such changes are indicative of a reduced welfare status—although the magnitude of the effects has tended to be dependent upon study‐specific conditions. Systematic observations on large scale commercial farms are therefore required, rather than extrapolation of these mainly small‐scale experimental findings. There is dispute as to the cause of the observed effects of increasing density, with water quality deterioration and/or an increase in aggressive behaviour being variously proposed. Both causes can theoretically generate the observed effects of increasing density, and the relative contribution of the two causes may depend upon the specific conditions. However, documentation of the relationship between density and the effects of aggressive behaviour at relevant commercial densities is lacking. Consequently only inferential evidence exists that aggressive behaviour generates the observed effects of increasing density, whereas there is direct experimental evidence that water quality degradation is responsible. Nevertheless, there are contradictory recommendations in the literature for key water quality parameters to ensure adequate welfare status. The potential for welfare to be detrimentally affected by non‐aggressive behavioural interactions (abrasion, collision, obstruction) and low densities (due to excessive aggressive behaviour and a poor feeding response) have been largely overlooked. Legislation directly limiting stocking density is likely to be unworkable, and a more practical option might be to prescribe acceptable levels of water quality, health, nutritional condition and behavioural indicators.  相似文献

Technological computation is entering the quantum realm, focusing attention on biomolecular information processing systems such as proteins, as presaged by the work of Michael Conrad. Protein conformational dynamics and pharmacological evidence suggest that protein conformational states-fundamental information units ('bits') in biological systems-are governed by quantum events, and are thus perhaps akin to quantum bits ('qubits') as utilized in quantum computation. 'Real time' dynamic activities within cells are regulated by the cell cytoskeleton, particularly microtubules (MTs) which are cylindrical lattice polymers of the protein tubulin. Recent evidence shows signaling, communication and conductivity in MTs, and theoretical models have predicted both classical and quantum information processing in MTs. In this paper we show conduction pathways for electron mobility and possible quantum tunneling and superconductivity among aromatic amino acids in tubulins. The pathways within tubulin match helical patterns in the microtubule lattice structure, which lend themselves to topological quantum effects resistant to decoherence. The Penrose-Hameroff 'Orch OR' model of consciousness is reviewed as an example of the possible utility of quantum computation in MTs.  相似文献

The effects of elevated [CO2] on 25 variables describing soybean physiology, growth and yield are reviewed using meta‐analytic techniques. This is the first meta‐analysis to our knowledge performed on a single crop species and summarizes the effects of 111 studies. These primary studies include numerous soybean growth forms, various stress and experimental treatments, and a range of elevated [CO2] levels (from 450 to 1250 p.p.m.), with a mean of 689 p.p.m. across all studies. Stimulation of soybean leaf CO2 assimilation rate with growth at elevated [CO2] was 39%, despite a 40% decrease in stomatal conductance and a 11% decrease in Rubisco activity. Increased leaf CO2 uptake combined with an 18% stimulation in leaf area to provide a 59% increase in canopy photosynthetic rate. The increase in total dry weight was lower at 37%, and seed yield still lower at 24%. This shows that even in an agronomic species selected for maximum investment in seed, several plant level feedbacks prevent additional investment in reproduction, such that yield fails to reflect fully the increase in whole plant carbon uptake. Large soil containers (> 9 L) have been considered adequate for assessing plant responses to elevated [CO2]. However, in open‐top chamber experiments, soybeans grown in large pots showed a significant threefold smaller stimulation in yield than soybeans grown in the ground. This suggests that conclusions about plant yield based on pot studies, even when using very large containers, are a poor reflection of performance in the absence of any physical restriction on root growth. This review supports a number of current paradigms of plant responses to elevated [CO2]. Namely, stimulation of photosynthesis is greater in plants that fix N and have additional carbohydrate sinks in nodules. This supports the notion that photosynthetic capacity decreases when plants are N‐limited, but not when plants have adequate N and sink strength. The root : shoot ratio did not change with growth at elevated [CO2], sustaining the charge that biomass allocation is unaffected by growth at elevated [CO2] when plant size and ontogeny are considered.  相似文献

Local niche complementarity among species (the partitioning of species based upon niche differentiation) is predicted to affect local ecosystem functioning positively. However, recent theory predicts that greater local diversity may hinder local ecosystem functioning when diversity is enhanced through source–sink dynamics. We suggest community assembly as a way to incorporate both the local and regional processes that determine biodiversity and its consequent effects on ecosystem functioning. From this, we propose a hump-shaped relationship between diversity and ecosystem functioning at local scales, but a linear increase of functioning with diversity at regional scales due to regional complementarity.  相似文献

Forty seven vibracores and fifteen radiocarbon dates have beenobtained to outline the Holocene history of the North Inlet saltmarsh basin. Marsh deposits date from about 3500 years BP and havetransgressed over a Late Pleistocene beach-ridge terrain that waspartly eroded by Late Holocene tidal channel meandering. Marsh mudalso has prograded southward over shallow subtidal estuarine Macomamuds which date from about 4500 years BP and which are stillaccumulating in adjacent Winyah Bay. The southward migration of themarsh environment probably is due to the southward migration ofboth North Inlet and the mouth of Winyah Bay. The stratigraphy ofthe North Inlet basin offers no evidence for Late Holocene sea-level oscillations.Application of this model of marsh history to the study long-term ecosystem succession driven by slowly rising sea level isdiscussed.  相似文献

Despite an established link between epilepsy and sleep behavior, it remains unclear how specific epileptogenic mutations affect sleep and subsequently influence seizure susceptibility. Recently, Sun et al. (2012) created a fly knock-in model of human generalized epilepsy with febrile seizures plus (GEFS+), a wide-spectrum disorder characterized by fever-associated seizing in childhood and lifelong affliction. GEFS+ flies carry a disease-causing mutation in their voltage-gated sodium channel (VGSC) gene and display semidominant heat-induced seizing, likely due to reduced GABAergic inhibitory activity at high temperature. Here, we show that at room temperature the GEFS+ mutation dominantly modifies sleep, with mutants exhibiting rapid sleep onset at dusk and increased nighttime sleep as compared to controls. These characteristics of GEFS+ sleep were observed regardless of sex, mating status, and genetic background. GEFS+ mutant sleep phenotypes were more resistant to pharmacologic reduction of GABA transmission by carbamazepine (CBZ) than controls, and were mitigated by reducing GABAA receptor expression specifically in wake-promoting pigment dispersing factor (PDF) neurons. These findings are consistent with increased GABAergic transmission to PDF neurons being mainly responsible for the enhanced nighttime sleep of GEFS+ mutants. Additionally, analyses under other light conditions suggested that the GEFS+ mutation led to reduced buffering of behavioral responses to light on and off stimuli, which contributed to characteristic GEFS+ sleep phenotypes. We further found that GEFS+ mutants had normal circadian rhythms in free-running dark conditions. Interestingly, the mutants lacked a homeostatic rebound following mechanical sleep deprivation, and whereas deprivation treatment increased heat-induced seizure susceptibility in control flies, it unexpectedly reduced seizure activity in GEFS+ mutants. Our study has revealed the sleep architecture of a Drosophila VGSC mutant that harbors a human GEFS+ mutation, and provided unique insight into the relationship between sleep and epilepsy.  相似文献
