The synthesis and molecular structure of prolame, N-(3-hydroxy-1,3,5(10)-estratrien-17 beta-yl)-3-hydroxypropylamine, is described. It was characterized by ir, nmr, mass spectrometry and chemical analysis. The crystal structure of this compound was determined by single-crystal x-ray diffraction. Prolame belongs to space group P212121. Cell dimensions are: a = 8.356(2), b = 13.343(4) and c = 16.119(4) A. Z = 4; R = 4.1%.  相似文献

Genes specifying DNA primases (pri) are common in all IncP plasmids examined so far. These plasmids suppress the thermosensitive character of the Escherichia coli dnaG3 mutation. The mechanism of suppression appears to be identical to that known for RP4 and IncI alpha plasmids. The DNA primases of both these plasmid types can substitute for the dnaG protein in chromosomal DNA replication. The pri genes of the alpha and beta subgroup of IncP plasmids are related to each other as judged from Southern hybridization and immunological data. Extensive DNA and protein sequence homology has been detected although the gene products of the alpha and beta subgroups exhibit substantial differences in size. The arrangement of overlapping genes at the pri locus of IncP alpha plasmids also appears to be present in the IncP beta group.  相似文献

Strain LBG A3136 of Nocardia mediterranei (ETH Collection) was found to contain a low-copy-number covalently closed circular extrachromosomal element, pMEA 100, which could only be isolated from mycelium grown on agar plates. pMEA 100 could not be isolated from the closely related strain ATCC 13685. Hybridization experiments showed that pMEA 100 is present in strain LBG A3136 in the free as well as in the integrated form whereas in strain ATCC 13685 only an integrated form was detected. Excision and reintegration in strain LBG A3136 seemed to be site specific. pMEA100 was found to be self-transmissible, eliciting the lethal zygosis phenotype, and is possibly involved in fertility in N. mediterranei.  相似文献

The 17 base pair operator O R 3 oligonucleotide, which is the preferential binding site for the Cro repressor of phage , was studied by two-dimensional NMR spectroscopy. A sequential assignment procedure based on two-dimensional Nuclear Overhauser Effect (NOESY) and scalar coupling correlated (COSY) NMR spectroscopy, together with the knowledge of the oligodesoxynucleotide sequence, made it possible to assign the non-exhangeable base protons and the H1 and the H2-H2 sugar protons of the O R 3 operator DNA. The pattern of the observed NOE connectivities is consistent with a right-handed helical DNA structure. Summary The parameters affecting the formation in vivo of -aminoadipyl-cysteinyl-valine (ACV), an intermediate in penicillin biosynthesis, have been established in low- and high-penicillin producing strains ofPenicillium chrysogenum. ACV was found both in cell extracts and in the culture broth filtrates. (14C)valine, -(14C)aminoadipic acid and (14C)cysteine were efficiently incorporated into ACV. Formation of ACV was stimulated by phenylacetic acid when added during the growth of the culture. ACV biosynthesis was enhanced when protein synthesis was blocked with cycloheximide or anisomicin. The ACV-synthesising activity of the culture increased between 24 and 48 h of the culture preceeding penicillin biosynthesis, and remained constant thereafter. A decay of ACV-forming activity was observed when de novo protein synthesis was inhibited with cycloheximide. The apparent half-life of the ACV-synthesising enzyme system was 2.5 h.  相似文献

Energy metabolism during dark respiration of the green alga Chlorella fusca was investigated by 31P NMR spectroscopy. The kinetics of the transition from anaerobic to aerobic conditions (and vice versa) was followed with a temporal resolution of 16 s. This transition is accompanied by a shift of the cytoplasmic pH from 6.8 to 7.4, while the vacuolar pH remains constant. Simultaneously, an increase in the concentration of nucleoside-triphosphates and a decrease in the concentration of cytoplasmic orthophosphate take place, as well as the formation of mobile polyphosphates. The concentration of ATP and P i reach steady-state levels within 30 s. Upon the reverse transition, from aerobic to anaerobic conditions, steady-state concentrations are obtained only after 3 min.  相似文献

Summary Cells ofRhodospirillum rubrum have been immobilized in various gels and tested for photobiological hydrogen production. Agar proved to be the best immobilizing agent with respect to production rates as well as stability. Agar immobilized cells were also superior compared to liquid suspension cultures. Growth conditions of the cells prior to immobilization, e.g. cell age, light intensity or nutrient composition, were of primary importance for the activity in the later immobilized state. A reactor with agar immobilized cells has been operated successfully over 3000 h with a loss of the activity of about 60%. Mean rates for hydrogen production for immobilized cells in this work during the first 60 to 70 hours after immobilization were in the range of 18 to 34 μl H2 mg−1 d.w. h−1 and thus by a factor of up to 2 higher than liquid cultures under the same conditions. Maximal rates of hydrogen production (57 μl H2 ml−1 immobilized cell suspension) were reached in agar gel beads with cells immobilized after 70 h growth in liquid culture in the light and a cell density of 1.0 mg ml−1, 70 h after immobilization.  相似文献

Summary The new enzyme d-2-hydroxyisocaproate dehydrogenase (NAD+-dependent) was detected in strains of the genus Lactobacillus and related genera. Straight and branched chain aliphatic as well as aromatic 2-ketocarboxylic acids are stereospecifically reduced to the corresponding d-2-hydroxycarboxylic acids according to the following equation:R-CO-COOH + NADH + H+ R-CHOH-COOH + NAD+ The enzyme is called d-hydroxyisocaproate dehydrogenase by us because 2-ketoisocaproate is the substrate with the lowest KM-value. NAD(H) as a cofactor cannot be replaced by NADP(H). Because of its broad substrate specificity we chose the strain Lactobacillus casei ssp. pseudoplantarum (DSM 20 008) for enzyme production and characterization. d-2-hydroxyisocaproate dehydrogenase could be purified 180-fold starting with 500 g of wet cells.The purification procedure involved liquid-liquid extraction with aqueous two-phase systems and ion-exchange chromatography. At this stage the enzyme has a specific activity of 25 U/mg and can be used for technical applications. Further purification up to a homogeneous protein with a specific activity of 110 U/mg can be achieved by chromatography on Amberlite CG 50 at pH 3.5. Properties important for technical application of the d-HicDH were investigated, especially the substrate specificity and the optimum pH- and temperature ranges for activity and stability of the catalist.  相似文献

Summary The oxidation of propene by resting-cells of ethene-grown Mycobacterium E3 was inactivated by 1,2-epoxypropane. Inactivation increased with increasing epoxide concentrations with 50% inactivation at approximately 30 mM epoxide. Other lower epoxides as epoxyethane and 1,2-epoxybutane also inactivated oxidation of propene as well as of other alkenes. Propene oxidation by resting-cells of ethane-grown Mycobacterium E20 and resting-cells of methane-grown Methylosinus trichosporium OB3b was inactivated for 50% at much lower 1,2-epoxypropane concentrations of approximately 1 and 3 mM respectively. It was demonstrated that in vivo the predominant effect of 1,2-epoxypropane was on the epoxidizing enzyme, i.e. alkene mono-oxygenase (strain E3), alkane mono-oxygenase (strain E20) and methane mono-oxygenase (methylotroph) and that the effect of the epoxide on the alkene mono-oxygenase was irreversible.  相似文献
