Reports of changes in DNA content of certain types of cells following exposure to conditions of stress has led to the suggestion that two kinds of DNA may be present. One is genetic DNA, and the other is called "metabolic" DNA. In a further attempt to investigate the possibility of this phenomenon, determinations of DNA content were made on Feulgen-stained nuclei of adrenal glands and kidneys in cold-treated rats. Feulgen-stained nuclei were measured by two-wavelength microspectrophotometry. Particular attention was given to the handling of the smears in hydrolysis and staining. Mean values of Feulgen-DNA contents in a total of 720 nuclei demonstrated (a) a constancy of DNA content within 2% in individual nuclei both in adrenal medulla and kidney cortex, (b) no more than an average of 2% difference in DNA content between control and experimental nuclei, and (c) no more than an average of 1.5% difference in DNA content between normal kidney cortex nuclei and normal adrenal medulla nuclei. These results confirm the view that the more precise the measurement, the more accurately the constancy rule is obeyed. Moreover, there is no support for the concept of a metabolic DNA in the rat adrenal medulla.  相似文献

Attempts to modify experimentally the variegated phenotypes of the mutantsz inDrosophila melanogaster andw m2 inD. hydei, were largely unsuccessful. Transplanted eyes ofz were found to be more intensely pigmented, but this effect was not influenced by treatment of the imaginal disks or by the host's genotype. Inw m2, increased pigmentation was observed after treatment with 5-bromo-uracil.Thew m2 phenotype is strongly modified by supernumerary Y chromosomes. One Y shifts the eye colour from yellow to brown, two Y's, to dark red. In the Malpighian tubules, two Y's have a similarly strong effect, but one Y a slight effect only. Malpighian tubules ofw m2 larvae were used for a quantitative analysis of mottling. An average clonal cluster size of 1.1 was found. This indicates that the state of pigmentation is determined around the last cell division in the embryo.  相似文献

Analyses of plumules from seedlings of a winter wheat variety (Elgin) and a spring wheat variety (Red Bobs) indicated that SH and SS contents based on protein levels were similar in the whiter and spring wheat tissues grown at 20 C. Boll) varieties, when grown nt 3 C, contained greater concentrations of SH and SS groups, based on protein levels, than those present in the tissues grown at 20 C. The spring wheat tissues at 3 C maintained relatively constant levels of SH and SS during the growth period. Sulfhydryl (SH) and SS groups based on fresh weights were present in greater quantities in the tissues of the spring wheat grown at 20 C than in the winter wheat tissues grown at 20 C. The high SH and SS concentrations present in the warm-grown spring wheat tissues were due to a high concentration of protein extracted from that tissue. The SH and SS contents per unit fresh weight were quite similar in the cold-grown tissues of both varieties during the growth period. Both varieties at both temperatures showed a similar decrease in SH and SS contents, based on fresh weights, as maturation progressed. The spring wheats grown at 3 C and 20 C and the winter wheat grown at 3 C all showed similar characteristics and differed considerably from the winter wheat grown at 20 C. These data were positively correlated with the flowering potential of the wheat varieties.  相似文献

The effect of penicillin on the morphology and reproduction of some strains ofA. chroococcum was studied on a number of solid media. When the growth was not entirely suppressed by the penicillin, filamentous cells and spheroplasts were formed. The formation of spheroplasts was stimulated by peptone. Gonidia were sometimes formed inside the spheroplasts and also inside giant cells. They were released from the cell after disruption or after lysis of the cell wall. In some cases they produced dwarf cells. Under certain conditions groups of gonidia present in a cell fused and formed one or more normal-looking cells inside the mother cell. Sometimes one or moreAzotobacter cells developed inside a spheroplast or at the site of a spheroplast with a lysed cell wall. Microcolonies consisting of small cocci representing gonidia and dwarf cells were also observed occasionally at the sites of spheroplasts with lysed cell walls. Occasionally tiny groups of small elements with a less marked structure were found at such sites, probably representing debris of lysed cells. The production of normal-looking cells inside filamentous cells was greatly stimulated on a medium containing 10 percent horse serum, with a drop of sterile water containing 200 or 250 I.U. penicillin added in the centre of the plate. The growth ofA. chroococcum was greatly retarded when the medium contained 10 U/ml penicillin and seemed to be checked entirely at concentrations of 20 U/ml penicillin or higher. Occasionally, however, even at concentrations of 100 and 300 U/ml penicillin, a few filamentous cells were found and also a few microcolonies, visible only through the microscope, consisting of gonidia or regenerative rods. By repeated exposure ofAzotobacter to penicillin populations could be obtained that were adapted to high concentrations of this antibiotic.  相似文献

Four allelic pairs were found to segregate from the cross between two mung bean varieties:rrGGSSdd×RRggssDD. R.rr stands for red versus green cotyledons, hypocotyl and top of leaflet stalk, and for beige yellow versus bright yellow flowers. Seed coat differences areG.-gg for green versus yellow (or brownish) basic colour,S.-ss for spotted (mottled) versus non-spotted (self colour), andD.-dd for dull versus shiny surface.In the spotted class (S.), R.-rr conditions spot covered (SS) and dark spotted (Ss) versus light spotted (SS andSs).
