Background  

Synapsins are neuronal phosphoproteins involved in several functions correlated with both neurotransmitter release and synaptogenesis. The comprehension of the basal role of the synapsin family is hampered in vertebrates by the existence of multiple synapsin genes. Therefore, studying homologous genes in basal chordates, devoid of genome duplication, could help to achieve a better understanding of the complex functions of these proteins.  相似文献

Aedes aegypti (L.), the main vector of dengue fever in Brazil, has been controlled with the use of massive chemical products, contributing to the development of resistance and decreasing the insect control efficiency. The control of dipterans with bioinsecticides based on Bacillus thuringiensis has been satisfactory, due to the production of insecticidal proteins denominated Cry (crystal), Cyt (cytolytic) toxins and Chi (chitinase), and to the synergistic effects among them. The present work aimed to select B. thuringiensis isolates efficient against A. aegypti larvae. A bacterial collection containing 1,073 isolates of B. thuringiensis, obtained from different locations of Brazilian territory, had the DNA isolated and submitted to PCR amplifications using specific primers for cry4Aa, cry4Ba, cry11Aa, cry11Ba, cyt1Aa, cyt1Ab, cyt2Aa and chi genes. For the LC50 and LC90 determination, the entomopathogenic isolates were evaluated by selective and quantitative bioassays. Only 45 isolates (4.2%) presented amplicons for the cry and cyt genes. The chi gene sequence was detected in 25 (54.3%) of those isolates. From the 45 isolates submitted to the selective bioassays, 13 caused 100% mortality of A. aegypti larvae. The identification of cry, cyt and chi genes of B. thuringiensis and the toxicity analysis on A. aegypti led to the selection of a set of isolates that have the potential to be used in the formulation of new bioinsecticides.  相似文献

Cowpea crops are severely attacked by Callosobruchus maculatus, a Coleopteran that at the larval stage penetrates into stored seeds and feeds on cotyledons. Cowpea weevil control could be based in utilization of bacteria and fungi to reduce pest development. Entomopathogenic fungi, such as Metarhizium anisopliae, are able to control insect-pests and are widely applied in biological control. This report evaluated ten M. anisopliae isolates according to their virulence, correlating chitinolytic, proteolytic and alpha-amylolytic activities, as well proteomic analysis by two dimensional gels of fungal secretions in response to an induced medium containing C. maculatus shells, indicating novel biotechnological tools capable of improving cowpea crop resistance.  相似文献

This study investigates whether a 6-wk intermittent hypoxia training (IHT), designed to avoid reductions in training loads and intensities, improves the endurance performance capacity of competitive distance runners. Eighteen athletes were randomly assigned to train in normoxia [Nor group; n = 9; maximal oxygen uptake (VO2 max) = 61.5 +/- 1.1 ml x kg(-1) x min(-1)] or intermittently in hypoxia (Hyp group; n = 9; VO2 max = 64.2 +/- 1.2 ml x kg(-1) x min(-1)). Into their usual normoxic training schedule, athletes included two weekly high-intensity (second ventilatory threshold) and moderate-duration (24-40 min) training sessions, performed either in normoxia [inspired O2 fraction (FiO2) = 20.9%] or in normobaric hypoxia (FiO2) = 14.5%). Before and after training, all athletes realized 1) a normoxic and hypoxic incremental test to determine VO2 max and ventilatory thresholds (first and second ventilatory threshold), and 2) an all-out test at the pretraining minimal velocity eliciting VO2 max to determine their time to exhaustion (T(lim)) and the parameters of O2 uptake (VO2) kinetics. Only the Hyp group significantly improved VO2 max (+5% at both FiO2, P < 0.05), without changes in blood O2-carrying capacity. Moreover, T(lim) lengthened in the Hyp group only (+35%, P < 0.001), without significant modifications of VO2 kinetics. Despite similar training load, the Nor group displayed no such improvements, with unchanged VO2 max (+1%, nonsignificant), T(lim) (+10%, nonsignificant), and VO2 kinetics. In addition, T(lim) improvements in the Hyp group were not correlated with concomitant modifications of other parameters, including VO2 max or VO2 kinetics. The present IHT model, involving specific high-intensity and moderate-duration hypoxic sessions, may potentialize the metabolic stimuli of training in already trained athletes and elicit peripheral muscle adaptations, resulting in increased endurance performance capacity.  相似文献

One of the most important morphological changes occurring in arbuscular mycorrhizal (AM) roots takes place when the plant plasma membrane (PM) invaginates around the fungal arbuscular structures resulting in the periarbuscular membrane formation. To investigate whether AM symbiosis-specific proteins accumulate at this stage, two complementary MS approaches targeting the root PM from the model legume Medicago truncatula were designed. Membrane extracts were first enriched in PM using a discontinuous sucrose gradient method. The resulting PM fractions were further analysed with (i) an automated 2-D LC-MS/MS using a strong cation exchange and RP chromatography, and (ii) SDS-PAGE combined with a systematic LC-MS/MS analysis. Seventy-eight proteins, including hydrophobic ones, were reproducibly identified in the PM fraction from non-inoculated roots, representing the first survey of the M. truncatula root PM proteome. Comparison between non-inoculated and Glomus intraradices-inoculated roots revealed two proteins that differed in the mycorrhizal root PM fraction. They corresponded to an H(+)-ATPase (Mtha1) and a predicted glycosylphosphatidylinositol-anchored blue copper-binding protein (MtBcp1), both potentially located on the periarbuscular membrane. The exact role of MtBcp1 in AM symbiosis remains to be investigated.  相似文献

Carbon isotope discrimination during photosynthetic CO2 assimilation has been extensively studied and rigorous models have been developed, while the fractionations during photorespiratory and dark respiratory processes have been less well investigated. Whilst models of discrimination have included specific factors for fractionation during respiration (e) and photorespiration (f), these effects have been considered to be very small, i.e. not significantly modifying the net discrimination expressed in organic material. On this paper we consider the fractionation effects associated with specific reactions set against the overall discrimination which occurs during source-product transformations. We review the studies which have recently shown that discrimination occurs during respiration at night in intact C3 leaves, leading to the production of CO2 enriched in 13C (i.e., e = ?6‰ ), and modifying the signature of the remaining plant material. Under photorespiratory conditions (i.e. increased oxygen concentration and high temperature), the photorespiratory fractionation factor may be high (with f around +10‰ ), and significantly alters the observed net photosynthetic discrimination measured during gas exchange. Fractionation factors for both respiration and photorespiration have been shown to be variable among species and with environmental conditions, and we suggest that the term `apparent fractionation' be used to describe the net effect for each process. In this paper we review the fractionations during photorespiration and dark respiration and the metabolic origin of the CO2 released during these processes, and we discuss the ecological implications of such fractionations.  相似文献

As local-area workstation networks are widely available, the idea of offering a software distributed shared memory (SDSM) system across interconnects of clusters is quite an attractive alternative for compute-intensive applications. However, the higher cost of sending a message over an inter-cluster link compared to an intra-cluster one can limit applications' performance on a multi-cluster SDSM system. In this paper, we present the extensions that we have added to the SDSM TreadMarks, which provides the lazy release consistency (LRC) memory model, in order to adapt it to a loosely-coupled cluster-based platform. We have implemented a logical per-cluster cache that exploits cluster locality. By accessing the cache of its cluster, a processor can share data previously requested by a second processor of its cluster, thereby, minimizing, the cost of inter-cluster communication.  相似文献

Mounting evidence suggests that average telomere length reflects previous stress and predicts subsequent survival across vertebrate species. In humans, leucocyte telomere length (LTL) is consistently shorter during adulthood in males than in females, although the causes of this sex difference and its generality to other mammals remain unknown. Here, we measured LTL in a cross‐sectional sample of free‐living Soay sheep and found shorter telomeres in males than in females in later adulthood (>3 years of age), but not in early life. This observation was not related to sex differences in growth or parasite burden, but we did find evidence for reduced LTL associated with increased horn growth in early life in males. Variation in LTL was independent of variation in the proportions of different leucocyte cell types, which are known to differ in telomere length. Our results provide the first evidence of sex differences in LTL from a wild mammal, but longitudinal studies are now required to determine whether telomere attrition rates or selective disappearance are responsible for these observed differences.  相似文献

Greenhouse gases (GHG) can be affected by grazing intensity, soil, and climate variables. This study aimed at assessing GHG emissions from a tropical pasture of Brazil to evaluate (i) how the grazing intensity affects the magnitude of GHG emissions; (ii) how season influences GHG production and consumption; and (iii) what are the key driving variables associated with GHG emissions. We measured under field conditions, during two years in a palisade-grass pasture managed with 3 grazing intensities: heavy (15 cm height), moderate (25 cm height), and light (35 cm height) N2O, CH4 and CO2 fluxes using static closed chambers and chromatographic quantification. The greater emissions occurred in the summer and the lower in the winter. N2O, CH4, and CO2 fluxes varied according to the season and were correlated with pasture grazing intensity, temperature, precipitation, % WFPS (water-filled pores space), and soil inorganic N. The explanatory variables differ according to the gas and season. Grazing intensity had a negative linear effect on annual cumulative N2O emissions and a positive linear effect on annual cumulative CO2 emissions. Grazing intensity, season, and year affected N2O, CH4, and CO2 emissions. Tropical grassland can be a large sink of N2O and CH4. GHG emissions were explained for different key driving variables according to the season.  相似文献

The biochemical properties of the ferredoxin/thioredoxin transduction pathway regulating the activity of key carbon-fixation enzymes through post-translational modifications are well characterized but little is known about the regulation of the different genes. In the present study, we investigated in Chlamydomonas reinhardtii the regulation of the expression of ferredoxin, thioredoxin m, ferredoxin-NADP reductase, phosphoribulokinase, as well as that of cytosolic thioredoxin h, the function of which is still largely unknown. The effects of light, the circadian clock and active cell division were investigated by northern blotting. The five genes were found to be regulated by light and the circadian clock but with different kinetics and amplitudes. This leads for the first time to the proposal that an extra-chloroplastic thioredoxin is possibly implicated in light and/or circadian-related processes. An interplay between several light-transduction pathways in controlling the expression of the genes is suggested by the expression studies and the theoretical analysis of the promoters.
