An infestation of a sooty tern colony on Bird Island, Seychelles, with the ixodid tick Amblovomma loculosum was studied during visits to the colony in 1993 to 1995. During the infestation, adult ticks were numerous during the birds' incubation, larvae were abundant when young chicks were present, and there was some evidence that nymphs were found when older chicks were present. The life cycle of the tick appeared closely synchronized with that of its host. Following a breeding failure of the sooty terns, caused by adverse weather in August-September 1994, few adult ticks were found during the birds' breeding season of 1995, suggesting that the absence of chicks late in the 1994 breeding season led to starvation of the ticks and their failure to survive their diapause until the onset of the birds' next breeding season.  相似文献

Viet nam is known as an endemic area of melioidosis but its etiologic agent originated in Viet nam was not extensively studied. For the first time, we analyzed the cellular lipid and fatty acid compositions of 15 Vietnamese isolates of Burkholderia pseudomallei, 10 from humans and 5 from the environment. Cellular lipid compositions were analyzed by two-dimensional thin-layer chromatography on silica gel G plates. Cellular fatty acid methyl esters were analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). The major lipids in all the isolates were phosphatidylglycerol (PG), two forms of phosphatidylethanolamine (PE-1 and PE-2), and two forms of ornithine-containing lipid (OL-1 and OL-2). PE-1 contained non-hydroxy fatty acids at both sn-1 and ?2 positions, while PE-2 possessed 2-hydroxy fatty acids and non-hydroxy fatty acids in a ratio of 1: 1. Since snake venom phospholipase A2 digestion of PE-2 liberated 2-hydroxy fatty acids, it was confirmed that these acids are at the sn-2 position of glycerol moiety. In both OL-1 and OL-2, amide-linked fatty acid was 3-hydroxy palmitic acid (3-OH-C16: 0), while ester-linked fatty acids were non-hydroxy acids in OL-1 and 2-hydroxy acids in OL-2. The total cellular fatty acid compositions of the test strains were characterized by the presence of 2-hydroxy palmitic (2-OH-C16: 0), 2-hydroxy hexadecenoic (2-OH-C16: 1), 2-hydroxy octadecenoic (2-OH-C18: 1), 2-hydroxy methylene octadecanoic (2-OH-C19CPA), 3-hydroxy myristic (3-OH-C14: 0) and 3-hydroxy palmitic (3-OH-C16: 0) acids. There were significant differences in the concentration of hexadecenoic (C16: 1), methylene hexadecanoic (C17CPA), octadecenoic (C18: 1) and methylene octadecanoic (C19CPA) acids among the Vietnamese isolates of B. pseudomallei. However, no significant difference was observed in cellular lipid and fatty acid components between strains of human and environmental origins.  相似文献

Abstract The tandem mini-exon gene repeat is an ideal diagnostic target for trypanosomatids because it includes sequences that are conserved absolutely coupled with regions of extreme variability. We have exploited these features and the polymerase chain reaction to differentiate Phytomonas strains isolated from phloem, fruit or latex of various host plants. While the transcribed regions are nearly identical, the intergenic sequences are variable in size and content (130–332 base pairs). The mini-exon genes of these phytomonads can therefore be distinguished from each other and from the corresponding genes in insect trypanosomes, with which they are oft confused.  相似文献

Floating marshes occur over 70% of the western Terrebonne Basin, Louisiana, USA, freshwater coastal wetlands. They are of several types: A free-floating thick-mat (45–60 cm) marsh dominated by Panicum hemitomon and Sagittaria lancifolia; a thick mat marsh dominated by Panicum hemitomon and Sagittaria lancifolia that floats part of the year, but whose vertical floating range is damped compared to adjacent water; and an irregularly-floating thin mat (< 30 cm) dominated by Eleocharis spp. in the spring and Ludwigia leptocarpa and Bidens laevis in the summer and fall. Floating mats must be almost entirely organic in order to be buoyant enough to float. The western Terrebonne wetlands receive large winter/spring supplies of suspended sediments from the Atchafalaya River. Even though sediment concentrations in the adjacent bayou are as high as 100 mg l–1, the Panicum hemitomon/Sagittaria lancifolia free-floating marsh probably receives no over-surface sediments since it floats continuously. The bulk density data of the damped-floating marsh, however, suggest some mineral sediment input, probably during winter when this marsh is submerged. These two types of floating marsh could not have developed in the present sediment regime of the Atchafalaya River, but as long as they remain floating can continue to exist. Thin floating mats are found in areas receiving the least sediment (<20 mg 1–1 suspended sediment concentration in adjacent bayous). This low sediment environment probably made possible their formation within the past 20 years. They may represent a transitional stage in mat succession from (1) existing thick-mat floating marsh to a degrading floating marsh, or (2) a floating marsh developing in shallow open water.Corresponding editor: D. Neisseria meningitidis pili are filamentous protein structures that are essential adhesins in capsulate bacteria. Pili of adhesion variants of meningococcal strain C311 contain glycosyl residues on pilin (PilE), their major structural subunit. Despite the presence of three potential N -linked glycosylation sites, none appears to be occupied in these pilins. Instead, a novel O -linked trisaccharide substituent, not previously found as a constituent of glycoproteins, is present within a peptide spanning amino acid residues 45 to 73 of the PilE molecule. This structure contains a terminal 1-4-linked digalactose moiety covalently linked to a 2,4-diacetamido-2,4,6-trideoxyhexose sugar which is directly attached to pilin. Pilins derived from galactose epimerase ( galE ) mutants lack the digalactosyl moiety, but retain the diacetamidotrideoxyhexose substitution. Both parental (#3) pilins and those derived from a hyper-adherent variant (#16) contained identical sugar substitutions in this region of pilin, and galE mutants of #3 were similar to the parental phenotype in their adherence to host cells. These studies have confirmed our previous observations that meningococcal pili are glycosylated and provided the first structural evidence for the presence of covalently linked carbohydrate on pili. Summary Conjugation, the process of genetic transfer requiring cell-to-cell contact, has been the focus of many investigations. In recent years, the molecular aspect of conjugation has been questioned. Since it has been shown that during exponential growth plasmid DNA forms a complex with the folded chromosomal complex (FCC), the relationship of R64drd11 plasmid DNA to the FCC (chromosome plus membrane) during conjugal replication was examined. A cell system was used which allowed specific observation of conjugal events as they occurred in the donor cell. Evidence is presented to show that conjugally replicating R64drd11 covalently closed circular molecules co-sediment with the FCC in neutral sucrose gradients. The use of density gradients to separate DNA from membrane-bound DNA from free membrane, indicate that the membrane is the preferential structure for conjugally replicating plasmid DNA association.  相似文献

In two experiments 48 prepuberal Merino ewe lambs were injected with oestradiol-17 beta (E2) or saline to study the effect of E2 on their plasma LH levels and on oestrus and ovulation. In the three groups which received 30 (experiment I), 50 and 30 (experiment II) microgram E2 respectively, 27 out of 28 lambs showed an LH response, the corresponding mean LH peaks being 64.3 +/0 22.5, 153.6 +/-33.4 and 91.7 +/- 16.9 ng/ml at mean intervals of 11.1, 11.2 and 10.5 h, respectively, after injection. None of the 20 lambs in the control groups had an LH level higher than 18 ng/ml 12 h after injection. In the three E2 groups, 41.7, 62.5 and 37.5% of animals showed oestrus within 26 h of injection while in the control groups only one animal showed oestrus. Of 13 animals showing oestrus in the E2 groups, 11 failed to ovulate. The mean pre-injection plasma FSH level in experiment I was 102.7 ng/ml, and in four 5--7-month-old lambs over several weeks uas 155.3 ng/ml. Despite these high pre-injection levels of FSH, it appears that the follicles were unable to respond to the LH peak which followed the E2 injection.  相似文献

The conformational behaviour of a cyclic disaccharide, di-β-d-glucopyranose 1,6′:1′,6-dianhydride hexaacetate, has been investigated. Because this molecule can exist only with the glucose rings in the unusual flexible forms, such conformational parameters as pseudorotation phase-angles have been used. Within a given number of approximations, the conformational space available for the whole system can be explored by considering only one two-dimensional map. Detailed investigations have shown that three stable conformations may be proposed. Among these, two correspond to minima found in the solid state. In one form, the six-membered rings adopt a boat conformation, whereas a skew conformation is found for the other form. However, these two conformations cannot be considered to be unique models of the conformation in solution; they both produce sets of proton-proton coupling-constants inconsistent with observed n.m.r.-spectroscopic results. At least the third form, having the six-membered rings in skew conformations, has to be taken into account. Deviations from coupling constants-molecular conformation relationships are thought to originate from ring strain.  相似文献
