In this study we evaluated the capacity of a defined microbial consortium (five bacteria: Mycobacterium fortuitum, Bacillus cereus, Microbacterium sp., Gordonia polyisoprenivorans, Microbacteriaceae bacterium, Naphthalene-utilizing bacterium; and a fungus identified as Fusarium oxysporum) isolated from a PAHs contaminated landfarm site to degrade and mineralize different concentrations (0, 250, 500 and 1000 mg kg(-1)) of anthracene, phenanthrene and pyrene in soil. PAHs degradation and mineralization was evaluated by gas chromatography and respirometry, respectively. The microbial consortium degraded on average, 99%, 99% and 96% of the different concentrations of anthracene, phenanthrene and pyrene in the soil, in 70 days, respectively. This consortium mineralized 78%, on average, of the different concentrations of the 3 PAHs in soil after 70 days. Contrarily, the autochthonous soil microbial population showed no substantial mineralization of the PAHs. Bacterial and fungal isolates from the consortium, when inoculated separately to the soil, were less effective in anthracene mineralization compared to the consortium. This signifies synergistic promotion of PAHs mineralization by mixtures of the monoculture isolates (the microbial consortium).  相似文献

Iron may enhance polycyclic aromatic hydrocarbons (PAHs) degradation directly by increasing the activity of the enzymes involved in the aerobic biodegradation pathways for hydrocarbons, and indirectly by increasing the PAHs bioavailability due to the stimulation of biosurfactant production. In the present work, the PAH anthracene was used in order to study the effect of different forms and concentrations of iron on its biodegradation and surfactant production by Pseudomonas spp. isolates from a 14-years old petrochemical sludge landfarm site. Among the iron forms, iron nitrate was chosen based on its high solubility and effect on the increase in the growth of the isolate. Iron concentration of 0.1mM was selected as the limit between deficiency and toxicity for isolates growth and anthracene degradation. After 48 days Pseudomonas citronellolis isolate 222A degraded 72% of anthracene related to iron stimulation and surface tension decrease, indicating surfactant production. Pseudomonas aeruginosa isolate 332C was iron-stimulated but did not reduce surface tension while P. aeruginosa isolate 312A exhibited a noniron and surfactant dependence to degrade 72% of anthracene. Isolate 222A showed a direct dependence on iron to stimulate surfactant activity, which probably increased anthracene bioavailability. To our knowledge, this is the first report about the iron effect on anthracene degradation and surfactant production by a Pseudomonas sp. Based on the iron requirement and surfactant activity, the Pseudomonas isolates may be useful for bioremediation of PAHs.  相似文献

Material and methods: Fifty nine elders wearing complete dentures and living in retirement homes in Curitiba (southern Brazil), were divided into two groups: group #1, 26 patients with denture‐induced stomatitis and group #2, 33 patients without denture‐induced stomatitis. The two groups were evaluated in relation to the degree of denture‐induced stomatitis, salivary fungal loads, and secretion of some histolytic enzymes. Results: Patients from group #1 showed higher degrees of colonisation by Candida albicans (p = 0.031). Candida krusei, Candida tropicalis, and Candida parapsilosis were also isolated, but there were no differences between the groups (p > 0.05). Secretory aspartyl protease (Sap) and chondroitinase did not show significant differences among the isolated Candida spp. in the two groups. Phospholipase secretion rates were higher among the strains of C. albicans from group #2 (p = 0.036). The same behaviour was not detected for non‐albicans Candida species. Conclusions: The results could infer that differences in the secretion rates of candidal histolytic enzymes should not be imputed as imperative for the progress of denture‐induced stomatitis.  相似文献

Worldwide, fungal richness peaks in tropical forest biomes where they are the primary drivers of decomposition. Understanding how environmental and anthropogenic factors influence tropical macrofungal fruiting patterns should provide insight as to how, for example, climate change and deforestation may impact their long‐term demographic stability and evolutionary potential. However, in Amazonia no studies have yet to disentangle the effects of substrate, seasonality and forest history on phenology. Here, we quantitate spatial and temporal variation in community structure of fruiting macrofungi in relation to these factors at a long‐term forest management research site in central Amazonia: the Biomass and Nutrients of Tropical Rain Forest (BIONTE's). Basidiome surveys of four substrate classes (leaves, soil, branches and trunks) were conducted along 250 m2 transects in primary and secondary (managed) forests, between 2012–13. From the 669 basidiomes collected, 290 taxa were identified of which 44 percent were restricted to primary and 36 percent to secondary forests. Although species‐accumulation curves did not asymptote, rarefaction analyses and Fisher's alpha indicate contrasting differences in richness among forests in relation to substrate type. For example, leaf litter basidiome richness was higher in secondary forests, whereas the contrary was observed for soil communities, suggesting that variation in fruiting patterns in relation to disturbance is substrate‐dependent possibly due to differences in necromass quality and/or understory micro‐climates. Furthermore, secondary forests harbored significantly lower basidiome richness and abundance in dry months, suggesting synergistic impacts of seasonality and management history on fruiting regimes.  相似文献

It is well known that parasitoids are attracted to volatiles emitted by host‐damaged plants; however, this tritrophic interaction may change if plants are attacked by more than one herbivore species. The larval parasitoid Cotesia flavipesCameron (Hymenoptera: Braconidae) has been used intensively in Brazil to control the sugarcane borer, Diatraea saccharalisFabricius (Lepidoptera: Pyralidae) in sugarcane crops, where Spodoptera frugiperda (JE Smith) (Lepidoptera: Noctuidae), a non‐stemborer lepidopteran, is also a pest. Here, we investigated the ability of C. flavipes to discriminate between an unsuitable host (S. frugiperda) and a suitable host (D. saccharalis) based on herbivore‐induced plant volatiles (HIPVs) emitted by sugarcane, and whether multiple herbivory (D. saccharalis feeding on stalk + S. frugiperda feeding on leaves) in sugarcane affected the attractiveness of HIPVs to C. flavipes. Olfactometer assays indicated that volatiles of host and non‐host‐damaged plants were attractive to C. flavipes. Even though host‐ and non‐host‐damaged plants emitted considerably different volatile blends, neither naïve nor experienced wasps discriminated suitable and unsuitable hosts by means of HIPVs emitted by sugarcane. With regard to multiple herbivory, wasps innately preferred the odor blend emitted by sugarcane upon non‐host + host herbivory over host‐only damaged plants. Multiple herbivory caused a suppression of some volatiles relative to non‐host‐damaged sugarcane that may have resulted from the unaltered levels of jasmonic acid in host‐damaged plants, or from reduced palatability of host‐damaged plants to S. frugiperda. In conclusion, our study showed that C. flavipes responds to a wide range of plant volatile blends, and does not discriminate host from non‐host and non‐stemborer caterpillars based on HIPVs emitted from sugarcane. Moreover, we showed that multiple herbivory by the sugarcane borer and fall armyworm increases the attractiveness of sugarcane plants to the parasitoids.  相似文献

Bacillus sp. ES 29 (ATCC: BAA-696) is an efficient chromate reducing bacterium. We evaluated hexavalent chromium (Cr[VI]) reduction by immobilized intact cells and the cell-free enzyme extracts of Bacillus sp. ES 29 in a bioreactor system. Influences of different flow rates (3 to 14 mL h-1), Cr(VI) concentration (2 to 8 mg L-1), and immobilization support materials (Celite, amberlite, and Ca-alginate) on Cr(VI) reduction were examined. Both immobilized intact cells and the cell-free extract of Bacillus sp. ES 29 displayed substantial Cr(VI) reduction. Increasing flow rates from 3 to 6 mL h-1 did not affect the rate of Cr(VI) reduction, but above 6 mL h-1, the Cr(VI) reducing capacity of the immobilized intact cells and cell-free extract of Bacillus sp. ES 29 decreased. With both intact cells and the cell-free extracts, the rate of Cr(VI) reduction was inversely related to the concentration. Intact cells immobilized to Celite displayed the highest rate (k = 0.443 at 3 mL h-1) of Cr(VI) reduction. For the immobilized cell-free extract, maximal reduction (k = 0.689 at 3 mL h-1) was observed with Ca-alginate. Using initial Cr(VI) concentrations of 2 to 8 mg L-1 at flow rates of 3 to 6 mL h-1 both immobilized intact cells and the cell-free extracts reduced 84 to 98% of the influent Cr(VI). Results indicate that immobilized cells and the cell-free extracts of Bacillus sp. ES 29 could be used for large-scale removal of Cr(VI) from contaminated water and waste streams in containment systems.  相似文献

Capsular components of Cryptococcus neoformans induce several deleterious effects on T cells. However, it is unknown how the capsular components act on these lymphocytes. The present study characterized cellular and molecular events involved in immunoregulation of splenic CD4(+) T cells by C. neoformans capsular polysaccharides (CPSs). The results showed that CPSs induce proliferation of normal splenic CD4(+) T cells, but not of normal CD8(+) T or B lymphocytes. Such proliferation depended on physical contact between CPSs and viable splenic adherent cells (SAC) and CD40 ligand-induced intracellular signal transduction. The absence of lymphoproliferation after fixation of SAC with paraformaldehyde has discarded the hypothesis of a superantigen-like activation. The evaluation of a cytokine pattern produced by the responding CD4(+) T lymphocytes revealed that CPSs induce a dominant Th2 pattern, with high levels of IL-4 and IL-10 production and undetectable inflammatory cytokines, such as TNF-alpha and IFN-gamma. Blockade of CD40 ligand by relevant mAb down-regulated the CPS-induced anti-inflammatory cytokine production and abolished the enhancement of fungus growth in cocultures of SAC and CD4(+) T lymphocytes. Our findings suggest that CPSs induce proliferation and differentiation of normal CD4(+) T cells into a Th2 phenotype, which could favor parasite growth and thus important deleterious effects to the host.  相似文献

Abstract:  The objective of this work was to study the effect of different hosts and temperatures on Diaphorina citri Kuwayama (Hem., Psyllidae) biology. Citrus limonia (Rangpur lime), Murraya paniculata (orange jessamine) and Citrus sunki (Sunki mandarin) were used as hosts. Measurements included duration and viability of the egg and nymphal stages, sex ratio, fecundity and longevity. In order to verify the effect of temperature on D. citri biology, the duration and viability of its developmental stages and biological cycle were compared at seven different temperature conditions. Durations of the embryonic and nymphal stages were similar for Rangpur lime, orange jessamine and mandarin. However, the nymphal viability obtained on mandarin was lower than the values obtained on the other hosts. Fecundity was higher on orange jessamine and, in all instances, females had greater longevity than males. A fixed number of instars (five) was obtained on the three hosts. Rangpur lime and orange jessamine provided better D. citri development when compared with mandarin. Duration of the egg and nymphal stages varied from 2.6 to 7.7 and from 9.4 to 35.8 days, respectively, at temperatures from 18 to 32°C. Egg viability was higher than 81.6% at the temperature range studied (18–32°C); nymphal viability was higher than 70% at the 18–30°C range, differing from viability at 32°C, which was dramatically reduced. The D. citri lower temperature development threshold (TT) and thermal constant ( K ) values for the egg, nymphal and biological cycle (egg–adult) stages were 12.0°C and 52.6 Degree-Day (DD); 13.9°C and 156.9 DD; and 13.5°C and 210.9 DD respectively.  相似文献

The age of colonizing Hypothenemus hampei (Ferrari) females when they leave the native coffee fruit was determined under laboratory conditions. This biological parameter might be useful for planning experiments with this species because certain physiological statuses are expressed from or until a determined age, which in turn may determine the response of individuals to different treatments. An experimental device was used to simulate the conditions inside a coffee berry and to permit the observation of the abandonment behavior of the beetles. Virgin and mated females with or without melanized cuticles were used in the experiments. On average, colonizing coffee berry borers were 15-days-old at the moment of host abandonment. Females at this age were mated, had full dark cuticles, and were able to display flight and lay viable eggs. Interestingly, H. hampei females that mated before they acquire a fully dark cuticle abandoned the host 1.7 times faster than females that mated after they reach this physiological status. Further studies into food conditions and their impacts on the pre- abandonment of H. hampei females are encouraged.  相似文献

Saccharomyces cerevisiae has in several cases been proven to be a suitable host for the production of natural products and was recently exploited for the production of non‐ribosomal peptides. Synthesis of non‐ribosomal peptides (NRPs) is mediated by NRP synthetases (NRPSs), modular enzymes, which are often organized in enzyme complexes. In these complexes, partner NRPSs interact via communication‐mediating domains (COM domains). In order to test whether functional interaction between separate NRPS modules is possible in yeast we constructed a yeast strain expressing two modules with compatible COM domains from two plasmids. Successful production as well as secretion of the expected dipeptide was detected. This opens the possibility of using yeast as a eukaryotic platform for fast assessment of new module combinations for the development of novel NRP compounds. Biotechnol.
