BACKGROUND: Both intrinsic and acquired multidrug resistance play an important role in the insurgence of tuberculosis. Detailed knowledge of the molecular basis of drug recognition and transport by multidrug transport systems is required for the development of new antibiotics that are not extruded or of inhibitors that block the multidrug transporter and allow traditional antibiotics to be effective. MATERIALS AND METHODS: We have undertaken the inventory of the drug transporters subfamily, included in the major facilitator superfamily (MFS), encoded by the complete genome of Mycobacterium tuberculosis (MTB). These proteins were identified on the basis of their characteristic stretches of amino acids and transmembrane segments (TMS) number. CONCLUSIONS: Genome analysis and searches of homology between the identified transporters and proteins characterized in other organisms revealed 16 open reading frames encoding putative drug efflux pumps belonging to MFS. In the case of two of them, we also have demonstrated that they function as drug efflux proteins.  相似文献

We documented and assessed the influence of chimpanzee activity on group size and composition of Colobus guereza in Kyambura Gorge, southwest Uganda, from July to September, 1994 and in February and March 1996. The population density of colobus is very high: 347 individuals per km 2. Density differed outside activity centers of chimpanzees (525 individuals per km 2 ) and within the centers (186 per km 2 ). We identified a total of 24 colobus groups, ranging between 3 and 13 individuals. Of the 24 groups, 22 were one-male groups. Groups were smaller and the percentage of subadults and juveniles was lower in groups within chimpanzee activity centers. Estimates of home ranges are between 1.7 and 6.2 ha, but neighboring home ranges overlapped 80%. Only a territory of approximately 0.5 ha was defended by the alpha male.  相似文献

BACKGROUND: Spermatozoa acquire active fertilizing competence only after deposition in the female tract and subsequent capacitation. Recent studies on the cellular location of major sperm phosphoproteins suggest that capacitation is associated with tyrosine phosphorylation of proteins exposed on the sperm surface. However, these changes have not yet been quantified objectively. A calcium influx seems to be required for the completion of tyrosine phosphorylation in some species; however, the exact temporal coordination between these processes is still poorly understood. METHODS: Flow cytometry was used to quantify the degree of phosphorylation of the sperm surface proteins by probing with fluorescein isothiocyanate-conjugated anti-phosphotyrosine (pY) antibody raised in mouse. Dynamic changes in other sperm parameters (calcium influx, membrane integrity, and spontaneous acrosome reaction) were assessed to analyze their temporal coordination. RESULTS:: The changes in specific phosphotyrosine (pY) fluorescence signal detected in live, nonpermeabilized boar cell suspensions were biphasic during incubation under capacitating conditions. After 120 min of incubation, the degree of pY fluorescence increased threefold, indicating the changes in proteins exposed on sperm surface. At the same time there was a gradual increase in cytosolic calcium ion levels with the maximal rate at 60 min of incubation. This rate slowed immediately before the onset of the massive rise in tyrosine phosphorylation and decreased by 90% after its completion. The integrity of plasma and acrosome membranes decreased only slowly, illustrating that the changes observed were not due to the process of spontaneous acrosome reaction. CONCLUSIONS: These data provide quantitative evidence for the appearance of tyrosine-phosphorylated proteins on the surface of live boar spermatozoa during capacitation. An exact temporal coordination exists between cytosolic calcium ion content and protein tyrosine phosphorylation under these conditions. This novel approach has the advantage of making possible a precise quantification and kinetic comparison of molecular processes in different cell subpopulations.  相似文献

The mammalian endoplasmic reticulum (ER)-to-cytosol degradation pathway for disposal of misfolded proteins is an attractive target for therapeutic intervention in diseases that are characterized by impaired protein degradation. The ability to do so is hampered by the small number of specific inhibitors available and by our limited understanding of the individual steps involved in this pathway. Cells that express a class I major histocompatibility complex (MHC) heavy chain-enhanced green fluorescent protein (EGFP) fusion protein and the human cytomegalovirus protein US11, which catalyzes dislocation of the class I MHC EGFP reporter, show only little fluorescence. Treatment with proteasome inhibitors increases their fluorescence by stabilizing EGFP-tagged MHC class I molecules. We used this change in signal intensity as a readout to screen a chemical library of 16,320 compounds and identified two structurally related compounds (eeyarestatin I and II) that interfered with the degradation of both EGFP-heavy chain and its endogenous unmodified class I MHC heavy chain counterpart. Eeyarestatin I also inhibited degradation of a second misfolded type I membrane protein, T-cell receptor alpha. Both compounds stabilize these dislocation substrates in the ER membrane, without preventing proteasomal turnover of cytosolic substrates. The new inhibitors must therefore interfere with a step that precedes proteasomal degradation. The use of eeyarestatin I thus allows the definition of a new intermediate in dislocation.  相似文献

The brown alga Spatoglossum schroederi contains three fractions of sulfated polysaccharides. One of them was purified by acetone fractionation, ion exchange, and molecular sieving chromatography. It has a molecular size of 21.5 kDa and contains fucose, xylose, galactose, and sulfate in a molar ratio of 1.0:0.5:2.0:2.0 and contains trace amounts of glucuronic acid. Chemical analyses, methylation studies, and NMR spectroscopy showed that the polysaccharide has a unique structure, composed of a central core formed mainly by 4-linked beta-galactose units, partially sulfated at the 3-O position. Approximately 25% of these units contain branches of oligosaccharides (mostly tetrasaccharides) composed of 3-sulfated, 4-linked alpha-fucose and one or two nonsulfated, 4-linked beta-xylose units at the reducing and nonreducing end, respectively. This sulfated galactofucan showed no anticoagulant activity on several "in vitro" assays. Nevertheless, it had a potent antithrombotic activity on an animal model of experimental venous thrombosis. This effect is time-dependent, reaching the maximum 8 h after its administration compared with the more transient action of heparin. The effect was not observed with the desulfated molecule. Furthermore, the sulfated galactofucan was 2-fold more potent than heparin in stimulating the synthesis of an antithrombotic heparan sulfate by endothelial cells. Again, this action was also abolished by desulfation of the polysaccharide. Because this sulfated galactofucan has no anticoagulant activity but strongly stimulates the synthesis of heparan sulfate by endothelial cells, we suggested that this last effect may be related to the "in vivo" antithrombotic activity of this polysaccharide. In this case the highly sulfated heparan sulfate produced by the endothelial cells is in fact the antithrombotic agent. Our results suggested that this sulfated galactofucan may have a potential application as an antithrombotic drug.  相似文献

New methods are needed for rapid and sensitive assessment of sperm function. As the ability to fertilize an oocyte is acquired during the capacitation process, assessments of sperm function have to be performed under fertilizing conditions. In this study, we monitored the dynamics of the temporal response of sperm from ejaculates of both fertile and subfertile boars to capacitating conditions in vitro (responsiveness) by following the changes in the response to calcium ionophore treatment and in [Ca(2+)](i). The differences between individual males were also investigated. Ionophore-induced changes and increased intracellular calcium ion content in boar spermatozoa were found to progress as a function of time during incubation under capacitating conditions. After primary kinetic analysis, 120 min was chosen as the point in time for assessment of responsiveness. Intra-boar variability in responsiveness parameters was relatively high (variation coefficient CV>30%), especially in the response to ionophore treatment, indicating that an isolated test may be inadequate for the evaluation of sperm function. Despite this high variability, there were markedly significant individual differences with respect to changes during capacitation, and there were significant correlations between conventional and responsiveness sperm parameters. The population of samples from subfertile boars, was found to be heterogeneous in regard to sperm responsiveness to capacitating conditions. There were two significantly different classes of subfertile boars ("low" and "high" responders), indicating that fertility may be associated with suboptimal rather than maximal response (both too rapid and too slow membrane changes). Therefore, criteria for quality judgement should include both the low and upper limits of responsiveness. The use of responsiveness parameters together with conventional spermatological parameters improved the prediction level of multiple regression models for farrowing rate and litter size. It can be concluded that the combination of sperm responsiveness parameters applied here is a suitable tool for the evaluation of sperm function.  相似文献

The mammalian oocyte is encased by a transparent extracellular matrix, the zona pellucida (ZP), which consists of three glycoproteins, ZPA, ZPB, and ZPC. The glycan structures of the porcine ZP and the complete N-glycosylation pattern of the ZPB/ZPC oligomer has been recently described. Here we report the N-glycan pattern and N-glycosylation sites of the porcine ZP glycoprotein ZPA of an immature oocyte population as determined by a mass spectrometric approach. In-gel deglycosylation of the electrophoretically separated ZPA protein and comparison of the pattern obtained from the native, the desialylated and the endo-beta-galactosidase-treated glycoprotein allowed the assignment of the glycan structures by MALDI-TOF MS by considering the reported oligosaccharide structures. The major N-glycans are neutral biantennary complex structures containing one or two terminal galactose residues. Complex N-glycans carrying N-acetyllactosamine repeats are minor components and are mostly sialylated. A significant signal corresponding to a high-mannose type chain appeared in the three glycan maps. MS/MS analysis confirmed its identity as a pentamannosyl N-glycan. By the combination of tryptic digestion of the endo-beta-galactosidase-treated ZP glycoprotein mixture and in-gel digestion of ZPA with lectin affinity chromatography and reverse-phase HPLC, five of six N-glycosylation sites at Asn(84/93), Asn268, Asn316, Asn323, and Asn530 were identified by MS. Only one site was found to be glycosylated in the N-terminal tryptic glycopeptide with Asn(84/93.) N-glycosidase F treatment of the isolated glycopeptides and MS analysis resulted in the identification of the corresponding deglycosylated peptides.  相似文献

We studied the dynamics of the proteome of influenza virus A/PR/8/34 (H1N1) infected Madin-Darby canine kidney cells up to 12 hours post infection by mass spectrometry based quantitative proteomics using the approach of stable isotope labeling by amino acids in cell culture (SILAC). We identified 1311 cell proteins and, apart from the proton channel M2, all major virus proteins. Based on their abundance two groups of virus proteins could be distinguished being in line with the function of the proteins in genesis and formation of new virions. Further, the data indicate a correlation between the amount of proteins synthesized and their previously determined copy number inside the viral particle. We employed bioinformatic approaches such as functional clustering, gene ontology, and pathway (KEGG) enrichment tests to uncover co-regulated cellular protein sets, assigned the individual subsets to their biological function, and determined their interrelation within the progression of viral infection. For the first time we are able to describe dynamic changes of the cellular and, of note, the viral proteome in a time dependent manner simultaneously. Through cluster analysis, time dependent patterns of protein abundances revealed highly dynamic up- and/or down-regulation processes. Taken together our study provides strong evidence that virus infection has a major impact on the cell status at the protein level.  相似文献
