In recent years the enzymes of the C3 photosynthetic carbon reduction (PCR) cycle have been studied using the techniques of molecular biology. In this review we discuss the primary protein sequences and structural predictions that have been made for a number of these enzymes, which, with the input of crystallographic analysis, gives the opportunity to understand the mechanisms of enzyme activity.The genome organisation and gene structure of the PCR enzymes is another area which has recently expanded, and we discuss the regulation of the genes encoding these enzymes and the complex interaction of various factors which influence their expression.  相似文献

Adaptive landscapes have served as fruitful guides to evolutionary research for nearly a century. Current methods guided by landscape frameworks mostly utilize evolutionary modeling (e.g., fitting data to Ornstein–Uhlenbeck models) to make inferences about adaptive peaks. Recent alternative methods utilize known relationships between phenotypes and functional performance to derive information about adaptive landscapes; this information can then help explain the distribution of species in phenotypic space and help infer the relative importance of various functions for guiding diversification. Here, data on performance for three turtle shell functions–strength, hydrodynamic efficiency, and self‐righting ability–are used to develop a set of predicted performance optima in shell shape space. The distribution of performance optima shows significant similarity to the distribution of existing turtle species and helps explain the absence of shells in otherwise anomalously empty regions of morphospace. The method outperforms a modeling‐based approach in inferring the location of reasonable adaptive peaks and in explaining the shape of the phenotypic distributions of turtle shells. Performance surface‐based methods allow researchers to more directly connect functional performance with macroevolutionary diversification, and to explain the distribution of species (including presences and absences) across phenotypic space.  相似文献

Performance in the model refinement category of the 13th round of Critical Assessment of Structure Prediction (CASP13) is assessed, showing that some groups consistently improve most starting models whereas the majority of participants continue to degrade the starting model on average. Using the ranking formula developed for CASP12, it is shown that only 7 of 32 groups perform better than a “naïve predictor” who just submits the starting model. Common features in their approaches include a dependence on physics-based force fields to judge alternative conformations and the use of molecular dynamics to relax models to local minima, usually with some restraints to prevent excessively large movements. In addition to the traditional CASP metrics that focus largely on the quality of the overall fold, alternative metrics are evaluated, including comparisons of the main-chain and side-chain torsion angles, and the utility of the models for solving crystal structures by the molecular replacement method. It is proposed that the introduction of these metrics, as well as consideration of the accuracy of coordinate error estimates, would improve the discrimination between good and very good models.  相似文献

The functions of the actin-myosin–based contractile ring in cytokinesis remain to be elucidated. Recent findings show that in the fission yeast Schizosaccharomyces pombe, cleavage furrow ingression is driven by polymerization of cell wall fibers outside the plasma membrane, not by the contractile ring. Here we show that one function of the ring is to spatially coordinate septum cell wall assembly. We develop an improved method for live-cell imaging of the division apparatus by orienting the rod-shaped cells vertically using microfabricated wells. We observe that the septum hole and ring are circular and centered in wild-type cells and that in the absence of a functional ring, the septum continues to ingress but in a disorganized and asymmetric manner. By manipulating the cleavage furrow into different shapes, we show that the ring promotes local septum growth in a curvature-dependent manner, allowing even a misshapen septum to grow into a more regular shape. This curvature-dependent growth suggests a model in which contractile forces of the ring shape the septum cell wall by stimulating the cell wall machinery in a mechanosensitive manner. Mechanical regulation of the cell wall assembly may have general relevance to the morphogenesis of walled cells.  相似文献
