Assessment of eutrophication pressure on lakes using littoral invertebrates   总被引:1,自引:1,他引:0  
Until the E.U. Water Framework Directive listed benthic invertebrates as a biotic element to be used for ecological classification of lakes, techniques for the assessment of the response of littoral invertebrates to anthropogenic pressures were extremely limited compared with those of rivers and lake profundal zones. We describe here the development of an ecological classification model based on changes of littoral invertebrate assemblages across a gradient of eutrophication, which is the most widespread anthropogenic pressure on lakes across Europe. The model comprises three derived parameters, two of which were developed from taxon-specific optima along a total phosphorus gradient calculated using canonical correspondence analysis, and the third based on invertebrate abundance. Combining the parameter metrics, we can estimate the ecological quality ratio (EQR), relative to those from paleolimnologically-confirmed reference lakes. The model was tested using independent samples collected from both hard and soft substrata and across two seasons from 45 lakes, comprising three alkalinity groups (n = 15 in each), and across gradients in water column total phosphorus concentrations. For hard substrata, EQRs were related consistently and highly significantly to water column concentrations of total phosphorus, accounting for the majority of the variance in every alkalinity group. For samples taken from soft substrata, a significant relationship was found only for high alkalinity lakes, accounting for a moderate proportion of the variability in water column total phosphorus concentrations. Our results compare highly favourably with those from other aquatic ecological assessment methods, irrespective of the faunal or floral group upon which they are based, demonstrating that littoral invertebrate assemblages can provide a statistically robust prediction of nutrient status when samples are collected from hard substrata. While the method was developed specifically to assess nutrient pressures on littoral invertebrates, many lakes are subject to multiple pressures. The development of classification models that incorporate multiple pressures presents a particularly significant challenge for the implementation of the Water Framework Directive, requiring both reliable identification of minimally-impacted reference states and incorporation of pressures that are unlikely to interact in predictable ways.  相似文献

Renal fibrosis is the common histological feature of advanced glomerular and tubulointerstitial disease leading to end-stage renal disease (ESRD). However, specific antifibrotic therapies to slow down the evolution to ESRD are still absent. Because persistent inflammation is a key event in the development of fibrosis, we hypothesized that the proinflammatory kinin B1 receptor (B1R) could be such a new target. Here we show that, in the unilateral ureteral obstruction model of renal fibrosis, the B1R is overexpressed and that delayed treatment with an orally active nonpeptide B1R antagonist blocks macrophage infiltration, leading to a reversal of the level of renal fibrosis. In vivo bone marrow transplantation studies as well as in vitro studies on renal cells show that part of this antifibrotic mechanism of B1R blockade involves a direct effect on resident renal cells by inhibiting chemokine CCL2 and CCL7 expression. These findings suggest that blocking the B1R is a promising antifibrotic therapy.  相似文献

Niemann-Pick type C1 (NPC) disease is an autosomal recessive neurodegenerative disorder. One feature of the mouse model of NPC1 is it’s infertility. We have made transgenic mice which express the Npc1 protein exclusively in fibrillary astrocytes, using the glial fibrillary acidic protein (GFAP) promoter. This selective expression ofNpc1 corrects sterility in GFAP-Npc1E,Npc1 -/- mice. Counts of acidophils in the pituitary of GFAP-Npc1E,Npcl -/- mice, as compared toNpcl -/- mice, and measurements of dopamine D2 receptor (DRD2) mRNA in the pituitary, suggest mechanisms for fertility enhancement. We conclude that the correction of sterility in GFAP-Npc1E,Npc1 -/- mice is a result of restoring hypothalamic control of the pituitary.  相似文献

Increased sediment loading comprises one of the most important and pervasive anthropogenic impacts on aquatic ecosystems globally. In spite of this, little is known of the overall effects of increased sediment loads on lakes. By modifying both bottom-up and top-down ecological processes and restructuring energy flow pathways, increased sediment loads not only alter biotic assemblage structure and ecological functioning significantly, but frequently result in reduced biological diversity and productivity. Although lake food-webs can be subsidised to some extent by the adsorption of organic carbon to fine sediments, trophic structure and the composition of biotic assemblages remain likely to be modified considerably. The mineralogy and particle size of sediments and the availability of nutrients, by influencing both the scale and nature of impacts, are key determinants of the overall effects of increased sediment loads on lake ecosystems. Although interactions with other global anthropogenic pressures, such as invasion by exotic species and climate change, are likely to be significant, little remains known about the nature or likely strength of those interactions. Widespread increases in sediment loading to lakes have, therefore, profound implications for the conservation and management of global aquatic biological diversity.  相似文献

Background  

Toxoplasma gondii is a zoonotic parasite of global importance. In common with many protozoan parasites it has the capacity for sexual recombination, but current evidence suggests this is rarely employed. The global population structure is dominated by a small number of clonal genotypes, which exhibit biallelic variation and limited intralineage divergence. Cannabinoid type-1 (CB(1)) receptor ligands, derived from the 1,5-diarylpyrazole core template of rimonabant (Acomplia), have been the focus of several studies aimed at examining structure-activity relationships (SARs). The purpose of this study was to design and synthesize a set of compounds based on the 1,5-diarylpyrazole template while focusing on the potential for discovery of CB(1) receptor radioligands that might be used as probes with in vivo molecular imaging. Each synthesized ligand was evaluated for potency as an antagonist at CB(1) and cannabinoid type-2 (CB(2)) receptors in vitro using a GTPgamma(35)S-binding assay. clog P values were calculated with Pallas 3.0. The antagonist binding affinities (K(B)) at CB(1) receptors ranged from 11 to >16,000 nM, CB(1) versus CB(2) selectivities from 0.6 to 773, and clog Ps from 3.61 to 6.25. An interesting new ligand, namely N-(piperidin-1-yl)-1-(2-bromophenyl)-5-(4-methoxyphenyl)-4-methyl-1H-pyrazole-3-carboxamide (9j), emerged from the synthesized set with appealing properties (K(B)=11 nM; CB(1) selectivity>773; clog P=5.85), for labeling with carbon-11 and development as a radioligand for imaging brain CB(1) receptors in vivo with positron emission tomography (PET).  相似文献

In theory, adaptive divergence can increase intrinsic post‐zygotic reproductive isolation (RI), either directly via selection on loci associated with RI, or indirectly via linkage of incompatibility loci with loci under selection. To test this hypothesis, we measured RI at five intrinsic post‐zygotic reproductive barriers between 18 taxa from the genera Cakile and Erucaria (Brassicaceae). Using a comparative framework, we tested whether the magnitude of RI was associated with genetic distance, geographic distance, ecological divergence and parental mating system. Early stages of post‐zygotic RI related to F1 viability (i.e. initial seed set) tended to be stronger than later stages related to F1 fecundity (i.e. flower number, fruit number). Mating system significantly influenced early stages of RI, such that RI was lowest when the mother was selfing and father was outcrossing, consistent with an imbalance between sink strength and resistance to provisioning. We found little evidence that adaptive divergence accelerates the evolution of intrinsic post‐zygotic RI, consistent with a nonecological model of evolution that predicts the nonlinear accumulation of RI and RI asymmetry with time (i.e. genetic distance), irrespective of adaptive divergence. Thus, although certain aspects of ecological divergence do not appear to have contributed strongly to the evolution of RI in this system, divergence in mating system actually reduced RI, suggesting that mating system evolution may play a significant role in speciation dynamics.  相似文献

We study model selection for clustered data, when the focus is on cluster specific inference. Such data are often modelled using random effects, and conditional Akaike information was proposed in Vaida & Blanchard (2005) and used to derive an information criterion under linear mixed models. Here we extend the approach to generalized linear and proportional hazards mixed models. Outside the normal linear mixed models, exact calculations are not available and we resort to asymptotic approximations. In the presence of nuisance parameters, a profile conditional Akaike information is proposed. Bootstrap methods are considered for their potential advantage in finite samples. Simulations show that the performance of the bootstrap and the analytic criteria are comparable, with bootstrap demonstrating some advantages for larger cluster sizes. The proposed criteria are applied to two cancer datasets to select models when the cluster-specific inference is of interest.  相似文献
