近年来, 生态系统服务评估逐渐地被运用到城市绿色基础设施的规划建设当中, 发掘能提供高生态效益的绿色屋顶潜力区域是增加城市生态空间有效途径。以提供城市空气净化、雨水截留、生物栖息地、热环境调节四个方面的生态服务为目标, 借助GIS平台确定醴陵市绿色屋顶生态空间潜力区域并提出规划策略, 为醴陵市以绿色屋顶形式完善城市生态空间结构提供参考。研究表明: (1)醴陵市潜在绿色屋顶生态系统服务评估中, 有20.3 km2的区域被划分为高效益服务区域, 约占研究区域面积的63.8%。(2)建筑年龄、结构和资金是小城市建设绿色屋顶的主要限制因素, 现状建筑中, 不适宜改建的建筑约占总建筑面积46.25%, 低限制改建的建筑占15.86%。 (3)通过叠加分析, 提取了高效益区域内的低限制改建建筑, 作为醴陵市绿色屋顶生态空间潜力区域, 面积为0.53 km2。 (4) 结合醴陵市绿地系统规划, 以“生态源地-廊道-节点”为结构, 以“分期规划, 重点建设”、“空间互补, 全域网络”两大原则, 形成“五重要节点、四优先区域、五主要廊道、全域网络”的醴陵市绿色屋顶生态空间格局。所提出的潜力评估和规划策略, 不仅可以为醴陵市建设绿色屋顶、优化城市空间构架提供依据, 还可为其他小城市以绿色屋顶形式增加生态空间的规划建设提供参考。  相似文献

A unique serpin, kallistatin, displays vasodilatory, antiangiogenic, anti-inflammatory, and antioxidant activity. Difficulty and low efficacy of obtaining recombinant kallistatin limit the wide investigation of its biological and pathological function. The present study employed a codon optimization algorithm to redesign the kallistatin gene and achieved a high yield of recombinant kallistatin protein. The kallistatin codons were redesigned for a more suitable Escherichia coli host without altering amino acids. Base composition and GC% content were compared between synthetic optimized kallistatin (opti-kallistatin) and wild-type kallistatin (wt-kallistatin). Both opti-kallistatin and wt-kallistatin were purified using Ni-NTA His-binding resins through fast protein liquid chromatography (FPLC). The identity and purity of kallistatin were confirmed by Coomassie blue staining, sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), and Western blot analysis. The output of opti-kallistatin protein was ~2-fold increase (2.09 ± 0.23 mg/L) compared to wt-kallistatin (1.05 ± 0.2 mg/L). These results suggest that more common codon optimization in the E. coli host significantly increases the yield of heterologous human protein yields. This approach will remarkably facilitate the further investigation of kallistatin in vitro and in vivo.  相似文献

The epidermal growth factor receptor (EGFR) signaling pathway regulates cell proliferation, differentiation, and survival, and is frequently dysregulated in esophageal and gastric cancers. Few studies have comprehensively examined the association between germline genetic variants in the EGFR pathway and risk of esophageal and gastric cancers. Based on a genome-wide association study in a Han Chinese population, we examined 3443 SNPs in 127 genes in the EGFR pathway for 1942 esophageal squamous cell carcinomas (ESCCs), 1758 gastric cancers (GCs), and 2111 controls. SNP-level analyses were conducted using logistic regression models. We applied the resampling-based adaptive rank truncated product approach to determine the gene- and pathway-level associations. The EGFR pathway was significantly associated with GC risk (P = 2.16×10−3). Gene-level analyses found 10 genes to be associated with GC, including FYN, MAPK8, MAP2K4, GNAI3, MAP2K1, TLN1, PRLR, PLCG2, RPS6KB2, and PIK3R3 (P<0.05). For ESCC, we did not observe a significant pathway-level association (P = 0.72), but gene-level analyses suggested associations between GNAI3, CHRNE, PAK4, WASL, and ITCH, and ESCC (P<0.05). Our data suggest an association between specific genes in the EGFR signaling pathway and risk of GC and ESCC. Further studies are warranted to validate these associations and to investigate underlying mechanisms.  相似文献

Radix Paeoniae Alba is widely used in Chinese traditional medicine to treat various diseases such as gastrointestinal disorders, immunomodulatory, cancer, and other diseases. In this paper, a novel acidic polysaccharide RPAPS purified from Radix Paeoniae Alba was evaluated for its structural features and potential of immunomodulatory and antioxidant activities. RPAPS (molecular weight: 1.0× 105 Da) was mainly composed of α-(1 → 4)-Glcp, α-Arap, α-Galp, α-Rhap, β-D-Glcp, α-(1 → 6)-linked Glcp and GalA. Immunological tests indicated that RPAPS could improve RAW264.7 phagocytic activity and LPS-induced splenocyte proliferation. For antioxidant activities, RPAPS showed reducing power and DPPH scavenging activity in dose dependent. Moreover, RPAPS could significantly protect the PC12 cells from H2O2 damage. These data implied polysaccharides RPAPS had the potential to be novel natural antioxidative and immunopotentiating agents for using in functional foods or medicine.  相似文献

Head and neck squamous cell carcinoma (HNSCC) is a common malignancy with high mortality and poor prognosis due to a lack of predictive markers. Increasing evidence has demonstrated small nucleolar RNAs (snoRNAs) play an important role in tumorigenesis. The aim of this study was to identify a prognostic snoRNA signature of HNSCC. Survival-related snoRNAs were screened by Cox regression analysis (univariate, least absolute shrinkage and selection operator, and multivariate). The predictive value was validated in different subgroups. The biological functions were explored by coexpression analysis and gene set enrichment analysis (GSEA). One hundred and thirteen survival-related snoRNAs were identified, and a five-snoRNA signature predicted prognosis with high sensitivity and specificity. Furthermore, the signature was applicable to patients of different sexes, ages, stages, grades, and anatomic subdivisions. Coexpression analysis and GSEA revealed the five-snoRNA are involved in regulating malignant phenotype and DNA/RNA editing. This five-snoRNA signature is not only a promising predictor of prognosis and survival but also a potential biomarker for patient stratification management.  相似文献

Aims

To assess whether the yew roots, which are able to provide a very constant environment due to their long life-span, can maintain the original arbuscular mycorrhizal (AM) fungal community during yew population decline.

Methods

The diversity of AM fungi (AMF) colonizing the roots of yew was analyzed by selecting the small subunit ribosomal RNA genes to construct a database of the overall community of AMF in the experimental area. A terminal restriction fragment length polymorphism (TRFLP) approach was used to identify the AMF communities present in yew roots. Physiological and environmental variables related to topology and soil and plant characteristics were determined as markers of habitat degradation.

Results

The AMF communities within yew roots were found to be dependent on soil, plant and topological variables indicative of habitat degradation surrounding the yew. The phylogenetic diversity of AMF associated to the yews was lower in habitats more exposed to degradation than in those better conserved.

Conclusions

The target yews can be grouped into two degradation levels. AMF communities were also affected by the degradation processes affecting their hosts. This finding rules out the role of these trees as refugia for their original AMF community, a fact that should be considered in plant reintroduction programs using AMF as bioenhancers.  相似文献

Plants in their natural environment frequently face various abiotic stresses, such as drought, high salinity, and chilling. Plant mitochondria contain an alternative oxidase (AOX), which is encoded by a small family of nuclear genes. AOX genes have been shown to be highly responsive to abiotic stresses. Using transgenic plants with varying levels of AOX expression, it has been confirmed that AOX genes are im- portant for abiotic stress tolerance. Although the roles of AOX under abiotic stresses have been extensively studied and there are several excellent reviews on this topic, the differential expression patterns of the AOX gene family members and the signal regulation of AOX gene(s) under abiotic stresses have not been extensively summarized. Here, we review and discuss the current progress of these two important issues.  相似文献

目的:探讨280例胃食管反流病(GERD)的分布特点及危险因素。方法:对临床诊断和胃镜确诊的280例GERD患者进行临床和风险因子相关性分析。结果:不论汉族还是维族,男性患者比例均明显高于女性;汉族患者高发年龄段早于维族患者(z=-2.939,P=0.003,);汉族和维族患者占反流性食管炎和Barrett食管比例分别为42.4%、81_3%及56.5%、18.8%,其中汉族患者Barrett食管比例较高(X2=14.358,P=0.000);肥胖、习惯性便秘、重体力活动者、饮食习惯不良在维族患者中的比例较高(P〈0.001)。结论:GERD与性别、年龄密切相关,男性多于女性,汉族患者发病年龄高峰旱于维族患者;汉族患者Barrett食管发生比例高于维族患者;肥胖、习惯性便秘、重体力活动、饮食习惯不良可能是GERD尤其是维族人群GERD的危险因素。  相似文献
