In this study, we aimed to analyse the spoilage potential of the isolated yeast, LAB and AAB species. Thus, 11 strains were inoculated at 0·3% (v/v) into a sterile filtered tchapalo and stored for 3 days at ambient temperature (27–30°C). All the tested strains grew well or remained stable except for Limosilactobacillus fermentum and Pediococcus acidilactici, which decreased throughout the storage time. A significant decrease of total soluble solids was observed only for Saccharomyces cerevisiae (from 7·8 to 5·8 °Brix) and Meyerozyma guilliermondii (from 7·8 to 5·5 °Brix). The tchapalo samples inoculated with the LAB strains Weissella paramesenteroides, P. acidilactici, L. fermentum and the yeast strain Candida tropicalis were judged similar to the control by the panellists. However, the strains of Lacticaseibacillus paracasei and Latilactobacillus curvatus (LAB), S. cerevisiae, M. guilliermondii and Kluyveromyces marxianus (yeasts) and Acetobacter pasteurianus and A. cerevisiae (AAB) induced the spoilage of the tchapalo appearance, smell and/or taste. In the spoiled tchapalo quantitative and qualitative modification of some volatile compounds (VOCs), such as lilac aldehyde, ethyl acetate, ethyl hexanoate, ethyl octanoate and phenethyl acetate, were observed. These results provide information about the microorganisms that need to be removed to extend the shelf life of tchapalo.  相似文献

Conventional aldehyde based fixatives produce good morphological preservation. However, owing to their cross-linking mechanism of action, epitope loss may occur during fixation compromising the tissue for subsequent immunohistochemical (IHC) analysis. IHC is an important tool for characterizing antigen, cytokine and cytomorphological markers. The increasing use of mouse models for study of pathogenesis has highlighted the need to investigate alternative fixatives. In the study reported here, tissue samples from RIII mice with immune mediated lesions, Mycobacterium bovis infected mice, and uninfected control mice were fixed in either zinc salt fixative or buffered formalin, then tested for IHC using a panel of antibodies (CD3, CD4, CD8, CD45, CD54, F4/80, Interferon-gamma and MIP2). Zinc salt fixation preserved processing-sensitive murine cell markers (CD4, CD8 and CD54) and improved the intensity of immunolabeling for CD45, F4/80 and CD3. Buffered formalin failed to preserve any of the processing-sensitive murine epitopes for demonstration by subsequent IHC.  相似文献

Most contemporary pig breeds have been constructed with a contribution of pigs of Asian origin (Volkopjalov et al., 1934; Tikhonov, 1967). At present, the closest relatives of the latter are the Vietnamese black and spotted pigs (Tikhonov, 1972a, 1972b). Immunogenetic characterization of Vietnamese pigs and hybrids between Vietnamese and European pigs may be of interst for the phylogenesis of the now existing pig breeds and the widening use of miniature laboratory pigs. Summary This work considers the continuous production of acetic acid by the homoacetogenic and thermophilic bacterium Acetogenium kivui. A mathematical model for the growth kinetics has been developed. The unstructured model for growth and product formation includes product and substrate inhibition as well as maintenance energy effects. The associated model parameters have been identified by non-linear optimization and evidenced experimentally in continuous culture as steady-state data. By using a mineral medium with glucose as the energy and carbon source for the bacteria proper carbon balances are available. The model permits good predictions of steady-state concentrations. This work aims to characterize the bioactive molecules produced by an antagonistic Bacillus sp. strain BCLRB2 isolated from healthy leaves of olive tree against Rhizoctonia solani and Sclerotinia sclerotiorum. The bacterial strain isolated showed a high and persistent antifungal activity against the two pathogens. The free-cell supernatant showed also a high antifungal activity against R. solani and at a lower extent against S. sclerotiorum. The partial purification of the antifungal substances with methanol gradient applied to C18 column binding the Bacillus BCLRB2 culture supernatant showed that the 20% and 60% methanol fractions had a high and specific activity against S. sclerotiorum and R. solani, respectively. The mass spectrometry identification of the compounds in the fraction specifically active against S. sclerotiorum revealed the presence of bacillomycin D C16 as a major lipopeptide. The fraction specifically active against R. solani contained bacillomycin D C15 and 2 unknown lipopeptides. The 80% methanol fraction had a moderate and a broad spectrum activity against the two pathogens and consisted from two iturin D (C13 and C14) as a major lipopeptides.  相似文献

In recent years, in urban areas of Novi Sad, unique ecological conditions, specific floristic and faunistic composition and poor habits of citizens in sense of public health, facilitate the development and maintenance of ticks. Regarding the importance of ticks as vectors of severe human and animal diseases, complex and detailed studies are conducted with an aim to find the most efficient methods for tick control. Two tick species, Ixodes ricinus and Dermacentor marginatus, were identified during a 3-year period on the territory of Municipality of Novi Sad. During 2006, the efficacy of the pyrethroid lambda-cyhalothrin in tick control varied from 60.7 to 100%. The highest efficacy recorded in 2007 was 92.3%. The efficacy of lambda-cyhalothrin in 2008 varied from 39.1 to 100%. Lambda-cyhalothrin showed high efficacy in tick control at localities which were improved before the application (mowed, litter removed, abundance control and euthanasia of abandoned cats and dogs). The results of this research indicate that lambda-cyhalothrin has a toxic effect on ticks and could be used as efficient acaricide for tick control, although its efficacy depends on formulation, terrain features and methods of application.  相似文献

Factor VIII (anti-hemophilia A factor) is isolated from human plasma. Purification is carried out by a combination of precipitation and chromatographic procedures. After precipitation, the first step in virus inactivation is achieved through the effect of a non-ionic detergent such as Tween 80, and a solvent, e.g. tri-n-butylphosphate (TnBP). By subsequent anion-exchange chromatography, a highly enriched product is isolated, consisting of a complex formed by factor VIII and von Willebrand factor (FVIII-vWF). This treatment also removes the virus-inactivating reagents to quantities in the low ppm range. The second step in virus inactivation is aimed specifically at the non-enveloped viruses and consists of pasteurization at temperatures higher than 60°C for 10 h. Through the addition of stabilizers, between 80% and 90% of the initial activity of FVIII is preserved during the modified pasteurisation. Along with the possibly denatured proteins the stabilizers, such as sugars, amino acids and bivalent cations, are subsequently removed by ion-exchange chromatography. The two-fold virus inactivation, by solvent/detergent treatment and subsequent pasteurisation, allows the destruction of both lipid-enveloped and non-enveloped viruses. During the procedure FVIII is stabilized through the high content of vWF. The complex consisting of FVIII and vWF can be dissociated by adding calcium ions. Subsequently both glycoproteins from this complex are separated from one another by further anion-exchange chromatography.  相似文献
