It was observed that the viral structural protein p15 from avian myeloblastosis virus emerges from ion-exchange column chromatography along with a proteolytic activity. p15 is apparently pure, as judged by sodium dodecyl sulfate-polyacryl-amide gel electrophoresis and isoelectric focusing. Increase and decrease in proteolytic activity coincided exactly with increasing and decreasing amounts of p15 during ion-exchange chromatography and during size fractionation by gell filtration. The proteolytic activity cleaved various substrates such as bovine serum albumin, ovalbumin, concanavalin A, and casein after denaturation by sodium dodecyl sulfate and heat. Highest enzyme activity was observed around pH 5.7. As judged from its cleavage pattern and its response to proteolytic inhibitors, the proteolytic activity appears papain-like, and the protease responsible for it may be classified as a thiol protease. If added to immunoprecipitated viral polyprotein precursor Pr76, p15 resulted in cleavage of Pr76,which could be inhibited by antibodies against p15. 相似文献

Chronic pontile cats (without hypothalamo-hypophysis) were kept during 4 days at central T (TC) between 37.5 and 30.8 degrees C at stable ambiant T (TA) between 28.5 and 23 degrees C. The vasomotor index of the forepaw was chosen for studying change in vasomotricity. Small and slow variations of TA (+1.5 degrees C) around 27 degrees C were followed by thermoregulatory response since a progressive decrease of TA under 27 degrees C led to vasoconstriction and increase of TC while progressive increase of TA above 27 degrees C led to vasodilatation and decrease of TC. However rapid and large decrease of TA under 27 degrees C (24-23 degrees C) led to the expected hypothermia with decrease of TC but without vasoconstriction. Paradoxical sleep (PS) amounts were strongly correlated with TC. At TC above 35.5 degrees C PS was almost totally suppressed while it increased significantly under 35 degrees C (Q10 = 0.10). Under 35 degrees C at stable TC and TA, PS occurred with an endogenous circahoral rhythm which did not vary significantly between 35 and 32 degrees C. These results strongly suggest that in pontile cats, PS is both gated and regulated by TC, while TC is regulated by pontobulbar vasomotor systems in response to TA. The putative role of the ventro-lateral medulla, in controlling both vasomotricity, TC and the excitability of the locus coeruleus is discussed in relation with PS. 相似文献

The highly enriched anaerobic bacterium that couples the reductive dechlorination of tetrachloroethene to growth, previously
referred to as PER-K23, was obtained in pure culture and characterized. The bacterium, which does not form spores, is a small,
gram-negative rod with one lateral flagellum. It utilized only H2 as an electron donor and tetrachloroethene and trichloroethene as electron acceptors in an anaerobic respiration process;
it could not grow fermentatively. Acetate served as a carbon source in a defined medium containing iron as the sole trace
element, the two vitamins thiamine and cyanocobalamin, and the three amino acids arginine, histidine, and threonine. The cells
contained menaquinones and b-type cytochromes. The G+C content of the DNA was 45.3 ± 0.3 mol%. The cell wall consisted of type-A3γ peptidoglycan with
ll-diaminopimelic acid and one glycine as an interpeptide bridge. The cells are surrounded by an S-layer; an outer membrane
was absent. Comparative sequence analysis of the 16S rRNA sequence showed that PER-K23 is related to gram-positive bacteria
with a low G+C content of the DNA. Based on the cytological, physiological, and phylogenetic characterization, it is proposed
to affiliate the isolate to a new genus, Dehalobacter, with PER-K23 as the type strain of the new species Dehalobacter restrictus.
