The Kluyveromyces lactis SLN1 phosphorelay system includes the osmosensor histidine kinase Sln1, the phosphotransfer protein Ypd1 and the response regulator Ssk1. Here we show that K. lactis has a functional phosphorelay system. In vitro assays, using a heterologous histidine kinase, show that the phosphate group is accepted by KlYpd1 and transferred to KlSsk1. Upon hyperosmotic stress the phosphorelay is inactivated, KlYpd1 is dephosphorylated in a KlSln1 dependent manner, and only the version of KlSsk1 that lacks the phosphate group interacts with the MAPKKK KlSsk2. Interestingly, inactivation of the KlPtp2 phosphatase in a ΔKlsln1 mutant did not lead to KlHog1 constitutive phosphorylation. KlHog1 can replace ScHog1p and activate the hyperosmotic response in Saccharomyces cerevisiae, and when ScSln1 is inactivated, KlHog1 becomes phosphorylated and induces cell lethality. All these observations indicate that the phosphorelay negatively regulates KlHog1. Nevertheless, in the absence of KlSln1 or KlYpd1, no constitutive phosphorylation is detected and cells are viable, suggesting that a strong negative feedback that is independent of KlPtp2 operates in K. lactis. Compared with S. cerevisiae, K. lactis has only a moderate accumulation of glycerol and fails to produce trehalose under hyperosmotic stress, indicating that regulation of osmolyte production is different in K. lactis.  相似文献

A new Late Miocene bovid, Urmiatherium kassandriensis sp. nov., from Northern Greece is described. The material comes from the Fourka locality in the Kassandra Peninsula (Chalkidiki), and the included fauna is estimated to be of Vallesian age. The two preserved crania represent a medium-sized taxon with short, conical horn cores, a flat cranial roof (consisting of the posterior part of the frontals, parietal and occipital), thick and porous frontals and pneumatized short parietals, an extremely thick basioccipital with voluminous posterior tuberosities and accessory articular facets for the atlas. The specialized atlanto-occipital joint recalls Pleistocene and extant ovibovines, but the braincase structure as a whole and the horn core features closely match Late Miocene ovibovine-like taxa, especially Plesiaddax and even more Urmiatherium. Nevertheless, the Kassandra bovid differs from representatives of both genera in the simpler horn core morphology and external brain anatomy. Urmiatherium is known to appear first in China and Iran at about 7.8 Ma, whereas its westernmost appearance on Samos Island (Greece) is dated much later. The presence of Urmiatherium kassandriensis sp. nov. in N. Greece suggests a farther west and earlier (Vallesian at least) first appearance of the genus. This would justify a basic geographic and phylogenetic split of Urmiatherium into two main Turolian lineages: a central-eastern Asian one leading to the sister species U. polaki and U. intermedium and a western one leading to U. rugosifrons.  相似文献

Having arrived 1.8 Ma ago, bison prevailed in the bovid assemblages of the European subcontinent for more than 1.5 Ma. The current geometric morphometric study outlines a framework of ecomorphological differences among several Bison populations of the western Palaearctic, shown by inferences from the tibial and tarsal joint surfaces of their astragalus. Given the principal biomechanical role of this element in the locomotion mechanism, its anatomical features could be linked to diverse functional aspects. In terms of morphological affinity, it is possible to attribute the studied fossil Bison astragalar material to several morphological trends. Shape variation is not explained by size differences and is possibly associated with an open-close habitat gradient, as indicated by the presence of expanded or compressed astragali, respectively. This intragroup spatial and temporal phenotypic diversity among the examined populations could indicate a biogeographic segregation influenced by regional climatic and landscape heterogeneity in the European territory during Pleistocene. Furthermore, a relation to habitat-specific locomotor ecology could be supported, revealing forms with increased cursoriality, operating in open biomes and closed-country dwellers as well.  相似文献

In the Mediterranean, Paracentrotus lividus and Sphaerechinus granularis are important drivers of benthic ecosystems, often coexisting in sublittoral communities. However, the introduction of the invasive diadematoid Diadema setosum, which utilizes venomous spines, may affect these communities. To describe the mechanical properties of the test and spines of these three species, specimens were collected in winter of 2019 from the sublittoral zone of the Dodecanese island complex, southeastern Aegean Sea. This region serves as a gateway for invasive species to the Mediterranean Sea. Crushing test was conducted on live individuals, while 3‐point bending test was used to estimate spine stiffness. Porosity and mineralogy of the test and spine, thickness of the test, and breaking length of the spine were measured and compared, while the microstructural architecture was also determined. The test of S. granularis was the most robust (194.35 ± 59.59 N), while the spines of D. setosum (4.76 ± 2.13 GPa) exhibited highest flexibility. Increased porosity and thickness of the test were related to increased robustness, whereas increased flexibility of the spine was attributed to high porosity, indicating that porosity in the skeleton plays a key role in preventing fracture. The spines of S. granularis exhibited highest length after fracture % (71.54 ± 5.5%). D. setosum exhibited higher values of Mg concentration in the test (10%) compared with the spines (4%). For the first time, the mineralogy of an invasive species is compared with its native counterpart, while a comparison of the mechanical properties of different species of the same ecosystem also takes place. This study highlights different ways, in which sea urchins utilize their skeleton and showcases the ecological significance of these adaptations, one of which is the different ways of utilization of the skeleton for defensive purposes, while the other is the ability of D. setosum to decrease the Mg % of its skeleton degrading its mechanical properties, without compromising its defense, by depending on venomous bearing spines. This enables this species to occupy not only tropical habitats, where it is indigenous, but also temperate like the eastern Mediterranean, which it has recently invaded.  相似文献

This study investigates the diel vertical distribution and the diet of the most important chaetognath species found in the 0–50 m surface layer of a coastal area in the eastern Mediterranean during a 24-hour period in July 2004. Among the recorded chaetognaths, Sagitta enflata was the most abundant species (41.6%), followed by S. minima (32.5%) and S. serratodentata (20.8%). Those three species exhibited a “twilight migration” pattern, with only small differences among them. Vertical separation was found between S. enflata and S. minima. Both species preyed mainly on cladocerans, although copepods were the most abundant group in the zooplankton assemblage. The chaetognath species followed partially the diel vertical migration of their prey. S. enflata showed high feeding intensity at different times in both day and night, while S. minima fed more intensively at midday (12:00) and at dusk (20:00), and S. serratodentata in the morning (08:00). It seems that in order to coexist in an area of low productivity the chaetognath species follow the basic ecological rules of space, time and food-type separation, in order to reduce the inter- and intra-specific competition. The high preference of S. minima and especially of S. enflata for the cladocerans made them probably the most important predators of cladocerans during summer.  相似文献

Fetal heart rate (FHR) variations reflect the level of oxygenation and blood pressure of the fetus. Electronic Fetal Monitoring (EFM), the continuous monitoring of the FHR, was introduced into clinical practice in the late 1960s and since then it has been considered as an indispensable tool for fetal surveillance. However, EFM evaluation and its merit is still an open field of controversy, mainly because it is not consistently reproducible and effective. In this work, we present a novel method based on grammatical evolution to discriminate acidemic from normal fetuses, utilizing features extracted from the FHR signal during the minutes immediately preceding delivery. The proposed method identifies linear and nonlinear correlations among the originally extracted features and creates/constructs a set of new ones, which, in turn, feed a nonlinear classifier. The classifier, which also uses a hybrid method for training, along with the constructed features was tested using a set of real data achieving an overall performance of 90% (specificity = sensitivity = 90%).  相似文献

Considering its immediate costs of producing dispensable males, the maintenance of sexual reproduction is a major paradox in evolutionary biology. Asexual lineages that do not face such costs theoretically should replace sexuals over time. Nonetheless, several systems are known in which closely related sexual and asexual lineages stably coexist. In the present study, we studied a sexual/asexual mating complex of a sperm-dependent parthenogenetic fish (amazon molly, Poecilia formosa) and its sexual congeners, the sailfin molly P. latipinna and the Atlantic molly P. mexicana. We asked whether differences in feeding behavior could contribute to their stable coexistence. We conducted a laboratory experiment to compare feeding efficiencies and also measured the competitive abilities between the two reproductive forms. Additionally, we measured gut fullness of fishes caught in natural habitats. Contrary to our predictions, we could not find P. formosa to be less efficient in feeding. We argue that food competition in mollies plays a minor role in mediating coexistence between closely related asexual and sexual mollies.  相似文献

A respirophasic variation of the QRS complex was observed in a patient with bilateral pulmonary embolism and pulmonary hypertension. An analogous variation of the Sa systolic velocity of the tricuspid annular tissue Doppler imaging was also noticed. Both phaenomena persisted until the clinical and haemodynamic improvement of the patient. A mechanistic approach of the electric phaenomenon is proposed.  相似文献

We report a new type of non-deletional hereditary persistence of fetal hemoglobin that is due to a C→T transition at position –158, relative to the Cap site of the human Aγ-globin gene. This mutation was identified in three unrelated adult cases presenting slightly elevated levels of fetal hemoglobin (Hb F), i.e. 2.9–5.1%, and normal hematological indices. Our sequencing results, from both polymerase chain reaction-amplified and subcloned DNA fragments, indicate that the Aγ–158C→T mutation occurred by two independent gene conversion events in the three cases studied. In addition, hematological and molecular data, including restriction fragment length polymorphism haplotyping in the β-globin gene cluster, extended haplotype analysis inside the γ-globin gene region and routine analysis of three tandem repeat loci (D1S80, 3′-HVR/apoB and F8vWf), led us to conclude that the Aγ–158C→T mutation in one of the three cases occurred recently in the parental germ line (P=99.47%), representing the first example of a de novo gene conversion event identified in humans.
