Ernährungsbiologie einer rhipidoglossen Kiemenschnecke

Zusammenfassung Die Arbeitsweise und die Leistungen der Rhipidoglossenradula von Theodoxus fluviatilis werden durch die kräftigen 4. Zwischenzähne und die Randbürstenzähne bestimmt. Die 4. Zwischenzähne lockern im wesentlichen die dem Fre\grund anhaftenden oder aufliegenden ein- bis wenigzelligen Algen (Diatomeen, chlorococcale und konjugate Grünalgen), die Randzähne fegen das gelockerte Nahrungsgut quantitativ zusammen. Fädige Grünalgen (z. B. Cladophorales) und Gewebeteile höherer Pflanzen werden nicht abgebissen bzw. abgeschabt.Die Diatomeen werden nur verdaut, wenn die Kieselschalen bereits bei der Nahrungsaufnahme mechanisch zerkleinert werden. Diese Zerkleinerung erfolgt allein auf einem Substrat mit rauher Oberfläche; sie wird durch die während des Bisses zwischen 4. Zwischenzähnen und Substrat auftretenden Reibungskräfte erzielt. Theodoxus wurde bei refiner Diatomeenernährung über mehrere Generationen gezüchtet. Tierisches Eiwei\ ist als Zusatzkost nicht erforderlich. Mit besonderen Hilfsma\nahmen kann Theodoxus im Laboratorium auch mit Cyanophyceen oder besonders mit Flagellaten (Chlamydomonas), die den Schnecken an den im Freiland besiedelten Standorten nicht zur Verfügung stehen, ernährt werden.Sämtliche Grünalgen mit stärkerer Cellulosewandung (Chlorococcales, Conjugatae) sind unverdaulich. Die Unverdaulichlichkeit beruht vermutlich auf einem Fehlen von Cellulasen im Verdauungstraktus. Die Zellmembranen und extrazellularen Scheiden der Cyanophyceen, die aus Hemicellulosen und Pektinen aufgebaut sind, werden im Magen aufgelöst. Theodoxus ist ein reiner Diatomeenfresser. Die ökologische Einnischung in die litorale Steinregion ist vorwiegend ernährungs-physiologisch begründet und erklärt das Vorkommen in Flie\gewassern und an Brandungsufern stehender Sü\gewässer sowie der Ostsee.

Das Inselorgan des Crossopterygiers Latimeria chalumnae J. L. B. Smith

Zusammenfassung Das Pankreas und der Fettkörper von Latimeria chalumnae, dem einzigen rezenten Crossopterygier aus der Klasse der Coelacanthes, werden in ihrer Lagebeziehung zu Magen, Darm und Milz beschrieben. Das von Millot und Anthony (1958) als Pankreas geschilderte Organ wird histologisch als Fettkörper identifiziert. Der exokrine Pankreasanteil besteht, ähnlich wie bei anderen niederen Wirbeltieren, aus einem tubulösen System exkretorischer Zellen. Das Inselorgan ist mit dem Gangsystem verbunden. Mit der Aldehydfuchsin Ponceau de Xylidine-Färbung lassen sich A- und B-Zellen, mit der Versilberung nach Hellman und Hellerström (1960) Silberzellen darstellen. Geschwollene Zellen werden als Fixationsartefakt nach Formolfixierung betrachtet. B-Zellknospen an den Gängen werden als erste phylogenetische Ansätze zu einer Dissemination des Inselorgans in das exokrine Parenchym hinein gedeutet. Zellen zwischen den exokrinen Tubuli werden beschrieben. Die Befunde werden mit den Ergebnissen am Pankreas niederer Wirbeltiere und Säuger verglichen.

Summary The topographical anatomy of the pancreas and the fat-body of Latimeria chalumnae, the only living Crossopterygian of the suborder of Coelacanths, is described. The organ, which was considered by Millot and Anthony (1958) to be the pancreas, has been histologically identified as a fat-body. The exocrine part of the pancreas consists of a tubulous system of exocrine cells, which are similar to those of lower vertebrates. The islet-organ is connected to the ductsystem. The aldehydfuchsin Ponceau de Xylidine-staining makes it possible to demonstrate - and -cells, the argyrophil reaction according to Hellman and Hellerström (1960) silvercells. Swollen cells are thought to be fixation artefacts due to formalin-fixation. -cellbuds, which are attached to the ducts, are regarded as the first phylogenetic beginning of dissemination of the islet-organ into the exocrine tissue. Cells between the exocrine tubules are described. The findings are compared with the results of the investigations of the pancreas of lower vertebrates and mammals.

Spatial patterns of gene expression in Brassica napus seedlings: identification of a cortex-specific gene and localization of mRNAs encoding isocitrate lyase and a polypeptide homologous to proteinases.

We investigated the spatial expression of three genes that are expressed during seed germination and postgerminative development in Brassica napus L. using in situ hybridization procedures. Two of the mRNAs encode isocitrate lyase and a predicted polypeptide that is homologous to cysteine proteinases. We reported previously that the mRNAs are prevalent primarily in cotyledons of seedlings and accumulate with similar kinetics during postgerminative growth. Here, we show that the two mRNAs are detected in several seedling tissues, but they display different distribution patterns in both cotyledons and root-shoot axes. The third mRNA is abundant in seedling axes and accumulates specifically in the ground meristem and mature cortex of hypocotyls and roots. Distribution of the mRNA in root meristems suggests that the gene product participates in an early event in cortical cell differentiation. Our results provide insight into the physiological processes that characterize seedlings.     

Antheseabhängige UV-Muster in Blütenständen von Asteraceen

The pseudanthia ofHeliopsis scabra andRudbeckia vulgaris (Asteraceae) were examined during the anthesis for differences in their UV patterns. Distinct changes in the reflectance and absorbance properties could be observed. The results suggest a close correlation between different stages of floral development and pollinator attraction.

Mom1 Is a Semi-Dominant Modifier of Intestinal Adenoma Size and Multiplicity in Min/+ Mice

The intestinal tumor multiplicity in mice heterozygous for Apc(Min) is strongly modulated by genetic background. On the sensitive C57BL/6J (B6) background, mice develop large numbers of intestinal adenomas. The AKR/J (AKR) strain carries alleles that correlate with a strong reduction in tumor multiplicity. To study the effect of one of these modifiers, Mom1, we have generated a mouse line in which the AKR allele of Mom1 is carried on the sensitive B6 genetic background. This strain was produced by using a marker-assisted selection method to eliminate unlinked AKR alleles more rapidly. The application and efficiency of this method are discussed. We used this strain to determine that Mom1 affects both tumor multiplicity and tumor size in a semi-dominant fashion.     

Evaluation and routine application of the novel restricted-access precolumn packing material Alkyl-Diol Silica: coupled-column high-performance liquid chromatographic analysis of the photoreactive drug 8-methoxypsoralen in plasma

A fully automated coupled-column HPLC method for on-line sample processing and determination of the photoreactive drug 8-methoxypsoralen (8-MOP) in plasma has been developed. The method is based on the novel internal-surface reversed-phase precolumn packing materials Alkyl-Diol Silica (ADS). This new family of restricted-access materials has a hydrophilic, electroneutral outer particle surface and a hydrophobic internal pore surface. The supports tolerate the direct and repetitive injection of proteinaceous fluids such as plasma and allow a classical C₁₈-, C₈- or C₄-reversed-phase partitioning at the internal (pore) surface. The total protein load, i.e. the lifetime of the precolumn used in this study (C₈-Alkyl-Diol Silica, 25 μm, 25 × 4 mm I.D.), exceeds more than 100 ml of plasma. 8-MOP was detected by its native fluorescence (excitation 312 nm, emission 540 nm). Validation of the method revealed a quantitative and matrix-independent recovery (99.5–101.3% measured at five concentrations between 21.3 and 625.2 ng of 8-MOP per milliliter of plasma), linearity over a wide range of 8-MOP concentrations (1.2–3070 ng of 8-MOP/ml, r = 0.999), low limits of detection (0.39 ng of 8-MOP/ml) and quantitation (0.79 ng of 8-MOP/ml) and a high between-run (C.V. 1.47%, n = 10) and within-run (C.V. 1.33%, n = 10) reproductivity. This paper introduces coupled-column HPLC as a suitable method for on-site analysis of drug plasma profiles (bedside-monitoring).     

Effect of heterologous paralytic shellfish poisoning toxin-enzyme conjugates on the cross-reactivity of a saxitoxin enzyme immunoassay

A polyclonal antiserum against saxitoxin (STX) was used in a competitive enzyme immunoassay for the detection of paralytic shellfish poisoning toxins. The extent of cross-reactions was determined from the amounts of neoSTX, decarbamoylSTX and gonyautoxin 2/3 (GTX2/3) that gave 50% inhibition in the assay. Horseradish peroxidase (HRP) conjugates of the toxins and a bovine serum albumin conjugate of STX (STX-BSA) were used. When compared with STX-BSA and STX as standard, the extent varied to which heterologous conjugates affected the binding values of the other toxins to the antibodies. The antibodies did not bind GTX2/3-HRP. By use of neoSTX-HRP or decarbamoylSTX-HRP as the labelled antigen instead of STX-HRP, the detection limit for neoSTX was improved to 100 pg ml^-1.
