Calcineurin is a calcium/calmodulin dependent protein phosphatase in eukaryotes that consists of a catalytic subunit A and a regulatory subunit B. Previous studies in the filamentous fungus Neurospora crassa had suggested that the catalytic subunit of calcineurin might be an essential protein. We generated N. crassa strains expressing the A (cna-1) and B (cnb-1) subunit genes under the regulation of Ptcu-1, a copper-responsive promoter. In these strains, addition of bathocuproinedisulfonic acid (BCS), a copper chelator, results in induction of cna-1 and cnb-1, while excess Cu2+ represses gene expression. Through analysis of these strains under repressing and inducing conditions, we found that the calcineurin is required for normal growth, asexual development and female fertility in N. crassa. Moreover, we isolated and analyzed cnb-1 mutant alleles generated by repeat-induced point mutation (RIP), with the results further supporting roles for calcineurin in growth and fertility in N. crassa. We demonstrated a direct interaction between the CNA-1 and CNB-1 proteins using an assay system developed to study protein-protein interactions in N. crassa.  相似文献

We have analyzed the allele frequency distribution at the highly polymorphic variable number of tandem repeat (VNTR) locus D1S80 (pMCT118) in seven ethnic populations (namely, New Guinea Highlanders of Papua New Guinea, Dogrib Indians of Canada, Pehuenche Indians of Chile, American and Western Samoans, Kacharis of Northeast India, and German Caucasians) using the polymerase chain reaction (PCR) technique. In the pooled sample of 443 unrelated individuals 20 segregating alleles were detected. A trimodal pattern of allelic distribution is present in the majority of populations and is indicative of the evolutionary antiquity of the polymorphism at this locus. In spite of the observed high degree of polymorphism (expected heterozygosity 56%–86%), with a single exception — the marginally significant P value (0.04) of the exact test in American Samoans — the genotype distributions in all populations conform to their respective Hardy-Weinberg expectations. Summary statistics indicate that, in general, the allele frequency distribution at this locus may be approximated by the infinite allele model. The data also demonstrate that alleles that are shared by all populations have the highest average frequency within populations. Furthermore, the kinship bioassay analysis demonstrates that the extensive variation observed at the D1S80 locus is at the interindividual within population level, which dwarfs any interpopulation allele frequency variation, consistent with the population dynamics of hypervariable polymorphisms. These characteristics of the D1S80 locus make it a very useful marker for population genetic research, genetic linkage studies, forensic identification of individuals, and for determination of biological relatedness of individuals.  相似文献

In this study, we designed a strategy for selecting the operating conditions of a mist reactor for the growth of hairy roots. This was accomplished by developing a mathematical model to optimize the ON/OFF mist duty cycle for the specified growth of hairy roots. The availability and rate of transport of nutrients to the roots are important parameters in the design and operation of the reactor. The thin liquid film over the root surface, which continuously builds up during the ON cycle, is a major factor that limits mass transfer. The same film also acts as a finite reservoir of nutrients in the absence of any replenishment during the OFF cycle. This reservoir gets depleted as growing roots continue to consume the nutrients. As a result of this depletion the duration of the OFF cycle must be limited such that the nutrient concentration does not go below the critical value required for the specified growth rate. The depleted reservoir is then replenished during the next ON cycle to an extent that depends on feed concentration and duration of the next ON cycle. It was shown that the use of increasing feed concentrations in the fed-batch mode of operation could maintain a specified growth rate in the reactor. Interestingly, this also resulted in the efficient operation of the reactor whereby the reactor operated at slightly above the required concentration and close to the point of minimum mass transfer resistance.  相似文献

A series of acylated chitin derivatives was prepared by reacting chitin in a solution of trifluoroacetic anhydride and each of the cyclopropyl, cyclobutyl, cyclopentyl, and cyclohexyl carboxylic acids. The degree of O-acyl substitution was in a range of 1.1-1.4 depending upon the nature of the cyclic acid added, as determined by FT-IR analysis. The solubility of the products in the organic solvents of DMF and THF increased with an increase in the cyclic chain length of the carboxylic acid. Thermal gravimetric analysis indicated that the products were stable up to 220 °C for chitin cyclopropanoate and cyclobutanoate, and 250 °C for chitin cyclopentanoate and cyclohexanoate. The surface morphology of the products by scanning electron microscopic analysis revealed porous and globular surface for chitin cyclobutanoate, cyclopentanoate, and cyclohexanoate, contrast to the dense and smooth organization for the cyclopropanoate.  相似文献
