Cytochalasin B and the kinetics of inhibition of biological transport: a case of asymmetric binding to the glucose carrier 总被引:7,自引:0,他引:7
Cytochalasin B inhibits glucose transport in human erythrocytes by competing with glucose for the carrier on the inner surface of the cell membrane, but there is no cytochalasin site associated with the outware-facing form of the carrier. Such asymmetry may be demonstrated by zero trans exit and entry experiments, whereas Sen-Widdas exit experiments are not easily interpretable. The orientation of the transport system appears to be reversed in certain other cell types: chich embryo fibroblasts, Novikoff hepatoma cells and HeLa cells. Electron microscopic evidence suggests that sperm can be spontaneously incorporated by cultured cells but cytogenetic and biochemical evidence indicate that sperm do not introduce new genes into such cells with detectable frequency. Sperm suspensions from mouse or Chinese hamster epididymis or human semen were added to cultures of RAG, a mouse cell line which dies in HAT medium because of HPRT deficiency. In EMs, sperm appeared to be readily phagocytized and degraded by the cells. When sperm-treated cultures were transferred to HAT medium resistant clones arose at a frequency of about 10−6, or at least 25× the reversion rate of RAG. Most HAT-resistant clones had HPRT activity which migrated electrophoretically like HPRT of the sperm donor species, though one was apparently a spontaneous RAG revertant. Most HAT-resistant clones had some chromosomes of the sperm donor species. In human sperm× RAG clones, the array of human chromosomes suggested that the human parent had been diploid rather than haploid; some cells contained both homologues of a polymorphic pair and some contained both X and Y. Furthermore, some sperm suspensions plated alone into flasks generated colonies, thus revealing the presence of low numbers of viable somatic cells. Presence of contaminating somatic cells in a sperm suspension was correlated with ability to induce HAT-resistant colonies when the suspension was added to RAG cells. Taken together, the data suggest that correction of the HPRT deficiency of RAG by sperm suspensions occurs at very low frequency and is probably due to efficient spontaneous fusion of low numbers of contaminating somatic cells with RAG cells. 相似文献

This study investigated the effects of dietary Bifidobacterium bifidum (BFD) and mannan-oligosaccharide (MOS), as a synbiotic, on the production performance, gut microbiology, serum biochemistry, antioxidant profile and health indices of broiler chicken. Six dietary treatments were T1 (negative control), T2 (positive control-20 mg antibiotic BMD kg−1 diet; BMD: bacitracin methylene disalicylate), T3 (0·1% MOS + 106 CFU BFD per g feed), T4 (0·1% MOS + 107 CFU BFD per g feed), T5 (0·2% MOS + 106 CFU BFD per g feed) and T6 (0·2% MOS + 107 CFU BFD per g feed). Significantly (P < 0·01) better growth performance and efficiency was observed in birds supplemented with 0·2% MOS along with 106 CFU BFD per g of feed compared to BMD and control birds. Supplementation with 0·2% MOS along with either 106 or 107 CFU BFD per g feed reduced (P < 0·01) the gut coliform, Escherichia coli, total plate count, and Clostridium perfringens count and increased the Lactobacillus and Bifidobacterium count. Significantly (P < 0·01) higher serum and liver antioxidant enzyme pool, serum HDL cholesterol and lower serum glucose, triglyceride, total cholesterol, cardiac risk ratio, atherogenic coefficient and atherogenic index of plasma were observed in birds supplemented with 0·2% MOS along with 106 CFU BFD per g of feed compared to control or BMD supplemented birds. Sudden elevations in external sodium chloride (NaCl) accelerate potassium (K+) efflux across the plasma membrane of plant root cells. It has been proposed that the extent of this acceleration can predict salt tolerance among contrasting cultivars. However, this proposal has not been considered in the context of plant nutritional history, nor has it been explored in rice (Oryza sativa L.), which stands among the world’s most important and salt-sensitive crop species. Using efflux analysis with 42K, coupled with growth and tissue K+ analyses, we examined the short- and long-term effects of NaCl exposure to plant performance within a nutritional matrix that significantly altered tissue-K+ set points in three rice cultivars that differ in salt tolerance: IR29 (sensitive), IR72 (moderate), and Pokkali (tolerant). We show that total short-term K+ release from roots in response to NaCl stress is small (no more than 26% over 45 min) in rice. Despite strong varietal differences, the extent of efflux is shown to be a poor predictor of plant performance on long-term NaCl stress. In fact, no measure of K+ status was found to correlate with plant performance among cultivars either in the presence or absence of NaCl stress. By contrast, shoot Na+ accumulation showed the strongest correlation (a negative one) with biomass, under long-term salinity. Pharmacological evidence suggests that NaCl-induced K+ efflux is a result of membrane disintegrity, possibly as result of osmotic shock, and not due to ion-channel mediation. Taken together, we conclude that, in rice, K+ status (including efflux) is a poor predictor of salt tolerance and overall plant performance and, instead, shoot Na+ accumulation is the key factor in performance decline on NaCl stress. 相似文献
