It has been proposed that the distribution of Robertsonian chromosome fusions and the Chromosome 11 Nucleolar Organizer Region (NOR) in the Danish hybrid zone between M. m. musculus and M. m. domesticus stems from centromeric incompatibilities between the two subspecies. To test this hypothesis, we identified and characterized a diagnostic subspecific marker closely linked to the centromere on mouse Chromosome 11. Using an allele-specific PCR assay, we investigated the introgression pattern of this centromere in a large sample of mice from a North–South transect of the hybrid zone in Jutland. Domesticus alleles were found to introgress far away from the center of the zone on the musculus side. These results suggest there is no incompatibility between the domesticus centromere of Chromosome 11 in the musculus genomic background.Two first authors contributed equally to this work. 相似文献

A simple equipment was developed to cultivate young cereal plants under enhanced CO2 concentration. Cultivation system permits growing of about 40 barley seedlings for about 2 to 3 weeks. The system consists

Kin cohesiveness and possible inbreeding in the mouthbrooding tilapia Sarotherodon melanotheron (Pisces Cichlidae) 总被引:1,自引:0,他引:1
Four microsatellite markers were used to study genetic variation among individuals of the mouthbrooding tilapia Sarotherodon melanotheron (Rüppel 1852) caught in separate but adjacent shoals. A comparison was also made with fish from six other localities. Populations originating from riverine environments appear to be panmictic, while samples from open waters such as lagoons showed highly significant heterozygote deficiencies. For instance, at the 33-allele locus SMEL4, 32 homozygous individuals were observed among the 82 individuals from the same lagoon location instead of only five homozygotes expected if random mating occurred. A further assessment of the genetic similarity of individuals within each shoal, validated by robust permutation techniques requiring no precise knowledge of gene frequencies, showed that related individuals tend to aggregate, and suggested that mating occurs preferentially within small groups of kin. Cichlids are often presented as a group of fish where microallopatric speciation takes place. The possible link between kin aggregation, inbreeding and shoaling behaviour we propose here may have important consequences for our understanding of the mechanisms involved in this fast speciation process. 相似文献

Lipoxygenases contain a nonheme iron that undergoes oxidation and reduction during the catalytic cycle. The conversion from the Fe3+ enzyme form to the Fe2+ form can be achieved using reducing inhibitors, a reaction that can be reversed with lipid hydroperoxides. The present study describes the properties of N-(4-chlorophenyl)-N-hydroxy-N'-(3-chlorophenyl)urea (CPHU), which functions as a reducing agent for various lipoxygenases and stimulates the degradation of lipid hydroperoxide catalyzed by these enzymes (pseudoperoxidase activity). CPHU was a substrate for the pseudoperoxidase reaction of purified soybean lipoxygenase-1 with apparent Km values for CPHU and 13-hydroperoxy-9,11-octadecadienoic acid (13-HpODE) of 14 and 15 microM, respectively. CPHU was converted during the pseudoperoxidase reaction to a mixture of products that can be resolved by reverse-phase high pressure liquid chromatography. By comparison with the chemical reaction of CPHU and potassium nitrosodisulfonate, the major enzymatic reaction product was tentatively identified as a one-electron oxidation product of CPHU. At low concentrations (50 microM), dithiothreitol completely protected against the degradation of hydroxyurea without inhibiting the pseudoperoxidase reaction. Under these conditions, the rate of the pseudoperoxidase reaction with CPHU as a substrate can be quantitated by the change in absorbance at 234 nm owing to the consumption of 13-HpODE. In addition to soybean lipoxygenase-1, CPHU was found to be a substrate for the pseudoperoxidase activities of purified recombinant human 5-lipoxygenase and porcine leukocyte 12-lipoxygenase. Some Daphnia species are difficult to culture due to a tendencyto become trapped in the surface film. A low level of cetylalcohol (1.26 0.12 mg) was shown to be highly effective inreducing the surface film entrapment of a Daphnia species typicallysusceptible to such a problem (D.rosea). In a species that doesnot exhibit surface film problems (D.pulicaria), a low levelof cetyl alcohol had no effect, indicating that this substanceis not toxic to Daphnia in low quantities. However, a high level(10.6 1.0 mg) of cetyl alcohol was shown to decrease the survivalof both species and reproduction of the susceptible species. 相似文献
