The site of feedback inhibition of the biosynthesis of pyrimidine nucleotides de novo was investigated in the isolated perfused rat liver. Hepatic uridine phosphate contents were specifically depleted by use of D-galactosamine. The effective activities of enzymes involved in the synthetic pathway were deduced from the rats of incorporation of labeled precursors into the acid-soluble uracil nucleotide pool and into some intermediates of the pathway. The labeling of hepatic urea was also monitored. When the uridine phosphate contents were less than 20% of controls, the incorporation of [14-C]-bicarbonate was stimulated about 20-fold. Label from [U-14C]oxaloacetate used as permeable precursor of intrace-lular aspartate was introduced into the uridylates to the same extent in normal and UTP-depleted livers. Similar results were obtained with labeled carbamoyl phosphate although the uptake of this compound by the liver was rather low. The lack of labeling of urea from exogenous carbamoyl phosphate does not indicate a free exchange of extra- and intramitochondrial carbamoyl phosphate. [ureido-14C]Ureidosuccinate produced in normal and D-galactosamine-treated livers almost identical labeling patterns of dihydroorotate, orotate and orotidine 5'-phosphate. Ten microsatellite DNA polymorphisms located on human chromosome 10 were regionally mapped using subchromosomal somatic cell hybrids and linkage analysis. The resulting order of the markers from pter-qter was [D10S89, D10S111], D10S107, D10S109, [D10S91, D10S110, D10S108, D10S88, D10S168], and D10S169. Order of the markers within brackets was uncertain, although the order given was most likely. Human dimensions research can help resource and wildlife managers make informed decisions, target information efforts, and gain a greater understanding of the factors that comprise attitudes toward wildlife management efforts. Despite these often-stated merits, studies addressing the human dimensions of resource and wildlife management efforts are rare in Europe. A proposed restoration of free-ranging European bison ( Bison bonasus ) in North Rhine-Westphalia, Germany, has presented an opportunity to help address this research gap. During May to July 2006, we used a randomly distributed, self-administered questionnaire ( n = 398) to assess local residents' attitudes, beliefs, and levels of support or opposition toward the proposed restoration. These factors were compared across two administrative regions spanned by the proposed restoration area. We found that respondents from the Siegen-Wittgenstein region held significantly more positive attitudes and significantly higher knowledge levels than respondents from the Hochsauerlandkreis region. Principal components analysis revealed that attitudes comprised a general attitude factor pertaining to issues such as the importance of conserving bison for future generations and a lifestyle impact factor, which included items pertaining to bison-caused damages to trees and crops. Logistic regression was used to show the influence of fear of bison on attitudes. We discuss the likely causes and management implications of our findings and provide suggestions to managers wishing to target information efforts and address the concerns of those affected by the proposed restoration. 相似文献

Combinations of monoclonal antibodies distinguish mesenchymal, myogenic, and chondrogenic precursors of the developing chick embryo 总被引:4,自引:0,他引:4
Monoclonal antibodies (MAbs) were used as probes for molecular differences in the surfaces of nonterminally differentiated cells of the developing chick limb. The specificity of the MAbs was determined by immunofluorescent localization performed on cultured breast muscle and limb bud cells and cryosections of a variety of embryonic (stages 15-37) and neonatal tissues. Subpopulations of MAb-positive and -negative cells were isolated by fluorescence-activated cell sorting and their developmental potential was assessed in vitro. Cells of the compacted somite, lateral plate mesoderm, and early limb bud were labeled with the CSAT MAb. Myogenic precursors of the dermatome and limb bud were labeled with the CSAT and L4 MAbs. Chondrogenic precursors of the sclerotome and limb bud were labeled with the CSAT, L4, and C5 MAbs. These precursors were distinguished from fibroblasts which were labeled with the CSAT and C1 MAbs. The differentiation and maturation of muscle and cartilage were accompanied by alterations in the labeling patterns of the MAbs. These results indicate that combinations of these MAbs can be used to distinguish mesenchymal, myogenic, and chondrogenic precursors, identify their site of origin during development, and isolate subpopulations of embryonic cells. 相似文献
