Control of nematode parasites with reduced reliance on the use of anthelmintics was studied in 16 ewes with suckling twin lambs on contaminated pasture in Denmark. Ewes and lambs were treated with albendazole at turn-out 3 May. Ewes were removed from the groups on 26 July, and lambs were slaughtered on 11 October. The animals were allocated to 4 groups of 8 lambs and their 4 ewes. Group TS was treated with albendazole at weeks 3, 6 and 8 after turnout and set-stocked; group TM was similarly treated but moved to clean pasture in conjunction with the last drenching; group US was untreated and set-stocked, and group UM was left untreated but moved to clean pasture week 8 after turn-out. Supplementary feed was offered in June and August due to scarcity of pasture. Strategic treatments of ewes and lambs weeks 3, 6 and 8 after turn-out, with or without a move to clean pasture, were highly effective in controlling nematode infections for most of the season. This was reflected in better weight gains and carcass characteristics in the treated compared to untreated lambs, resulting in an average increase in the value of the product by 36%. The effect of moving without treatment (UM) on faecal egg counts was limited but peak pasture infectivity was reduced to less than 10% compared to the set-stocked group and weight gains of lambs were significantly better despite poor feed availability in late season. The study showed that under set-stocked conditions repeated anthelmintic treatments of both ewes and lambs in early season may ensure sufficient nematode control whereas moving animals to clean pasture without dosing was less efficient. The latter may, however, still be a viable option in organic and other production systems where routine use of anthelmintics is banned, particularly if weaning and moving are combined or a second move is performed.  相似文献

Many viruses alter different stages of apoptosis of infected cells as a strategy for successful infection. Few studies have addressed mechanisms of equine herpesvirus 1 (EHV-1) strain-induced cell death. We investigated the effect of an abortigenic strain (AR8 strain) on heterologous Madin–Darby bovine kidney cells and homologous equine dermis (ED) cells cell lines. We compared morphologic and biochemical features of early and late apoptosis at different postinfection times. We investigated translocation of phosphatidylserine to the cell surface, nuclear fragmentation and changes in the cytoskeleton using flow cytometry and annexin V/propidium iodide staining, DNA laddering, terminal deoxynucleotidyl transferase UTP nick-end labeling assay and immunofluorescence staining of cytokeratin 18 cleavage. AR8 EVH-1 strain interfered with apoptosis in both cell lines, particularly during the middle stage of the replication cycle; this was more evident in ED cells. Although this antiapoptotic effect has been reported for other alpha herpesviruses, our findings may help elucidate how EHV-1 improves its infectivity during its cycle.  相似文献

近年来,可用于昆虫迁飞研究且可自动运行的垂直波束雷达(vertical-looking radar,VLR)的发展使得对迁飞性害虫的周年长期自动监测成为可能。本文提供了我们对能否将这种雷达应用于中国的褐飞虱和其他水稻害虫的监测与预测体系以改善其综合治理的可行性研究结果。以往的研究已经表明,这些害虫一般在300—2000m高度迁飞;而我们根据褐飞虱的雷达和射有效截面的计算结果表明,目前使用的3.2cm波长的VLR对褐飞虱个体目标的最大可检测高度仅约240m;虽然建造一部8.8mm波长的VLR即可覆盖褐飞虱迁飞高度的绝大部分,但其造价和维护费用均过于昂贵。为此,一个更可行的解决方案是,以3.2cm波长的VLR作为包括大多数水稻害虫在内的个体较大的迁飞性害虫的监测工具。  相似文献

Gizzard shad (Dorosoma cepedianum), a filter feeding omnivore, can consume phytoplankton, zooplankton and detritus and is a common prey fish in U.S. water bodies. Because of their feeding habits and abundance, shad have the potential to affect primary productivity (and hence water quality) directly through phytoplankton grazing and indirectly through zooplankton grazing and nutrient recycling. To test the ability of shad to influence primary productivity, we conducted a 16-day enclosure study (in 2.36-m3 mesocosms) and a 3-year whole-pond manipulation in 2–5 ha earthen ponds. In the mesocosm experiment, shad reduced zooplankton density and indirectly enhanced chlorophyll a concentration, primary productivity, and photosynthetic efficiency (assimilation number). While shad did not affect total phytoplankton density in the mesocosms, the density of large phytoplankton was directly reduced with shad. Results from the pond study were not consistent as predicted. There were few changes in the zooplankton and phytoplankton communities in ponds with versus ponds without gizzard shad. One apparent difference from systems in which previous work had been conducted was the presence of high densities of a potential competitor (i.e., larval bluegill) in our ponds. We suggest that the presence of these extremely high larval bluegill densities (20–350 larval bluegill m–3; 3–700 times higher density than that of larval gizzard shad) led to the lack of differences between ponds with versus ponds without gizzard shad. That is, the influence of gizzard shad on zooplankton or phytoplankton was less than the influence of abundant bluegill larvae. Differences in systems across regions must be incorporated into our understanding of factors affecting trophic interactions in aquatic systems if we are to be able to manage these systems for both water quality and fisheries.  相似文献

Myelin basic protein (MBP) and two peptides derived from MBP (MBP1–44 and MBP152–167) stimulated Schwann cell (SC) proliferation in a cAMP-mediated process. The two mitogenic regions of MBP did not compete with one another for binding to SC suggesting a distinctive SC receptor for each mitogenic peptide. Neutralizing antibodies to the fibroblast growth factor receptor blocked the mitogenic effect of the myelin-related SC mitogen found in the supernatant of myelin-fed macrophages. The binding of 125I-MBP to Schwann cells was specifically inhibited by basic fibroblast growth factor (bFGF) and conversely the binding of 125I-bFGF was competitively inhibited by MBP. These data suggested that the mitogenic effect of one MBP peptide was mediated by a bFGF receptor. The binding of MBP to ganglioside GM1 and the ability of MBP peptides containing homology to the B subunit of cholera toxin (which binds ganglioside GM1) to compete for the binding of a mitogenic peptide (MBP1–44) to SC, identified ganglioside GM1 as a second SC receptor. Based on these results, we conclude that MBP1–44 and MBP152–167 associate with ganglioside GM1 and the bFGF receptor respectively to stimulate SC mitosis.  相似文献

Proliferation of Schwann cells is one of the first events that occurs after contact with a growing axon. To further define the distribution and properties of this axonal mitogen, we have (a) cocultured cerebellar granule cells, which lack glial ensheathment in vivo with Schwann cells; and (b) exposed Schwann cell cultures to isolated granule cell membranes. Schwann cells cocultured with granule cells had a 30-fold increase in the labeling index over Schwann cells cultured alone, suggesting that the mitogen is located on the granule cell surface. Inhibition of granule cell proteoglycan synthesis caused a decrease in the granule cells' ability to stimulate Schwann cell proliferation. Membranes isolated from cerebellar granule cells when added to Schwann cell cultures caused a 45-fold stimulation in [3H]thymidine incorporation. The granule cell mitogenic signal was heat and trypsin sensitive and did not require lysosomal processing by Schwann cells to elicit its proliferative effect. The ability of granule cells and their isolated membranes to stimulate Schwann cell proliferation suggests that the mitogenic signal for Schwann cells is a ubiquitous factor present on all axons regardless of their ultimate state of glial ensheathment.  相似文献

Formation of microtubules at low temperature by tubulin from antarctic fish   总被引:1,自引:0,他引:1  
Tubulin was isolated from two species of antarctic fish, Pagothenia borchgrevinki and Dissostichus mawsoni, by cycles of temperature-dependent assembly, centrifugation, disassembly, and centrifugation. The preparations were found to consist almost entirely of tubulin and to contain negligibly small amounts of microtubule-associated proteins. This tubulin polymerized to make microtubules of ordinary dimensions. The formed microtubules appear to be in labile equilibrium with free tubulin dimer at all temperatures observed. In a buffer consisting of 0.1 M 1,4-piperazinediethanesulfonic acid, 2 mM dithioerythritol, 1 mM MgSO4, 2 mM ethylene glycol bis(beta-aminoethyl ether)-N,N,N',N'-tetraacetic acid, and 1 mM guanosine 5'-triphosphate, pH 6.9, the tubulin of P. borchgrevinki has a critical concentration for assembly of 0.046 (+/- 0.008) mg/mL at 35 degrees C and 0.74 (+/- 0.15) mg/mL at the habitat temperature of the fish, -1.8 degrees C. The critical concentration measured at the lower temperature is quite small relative to the critical concentration for formation of mammalian microtubules from pure tubulin at the same temperature, which must be at least 2 orders of magnitude larger. The antarctic fish microtubules may thus be called "cold stable" by comparison with mammalian microtubules. They do not fully dissociate at temperatures near 0 degree C because they are composed of tubulin that assembles more readily at these temperatures than does mammalian tubulin. There is no evidence for the presence of a cold-stabilizing factor in association with the tubulin. These findings suggest that alteration of tubulin may be a means by which some poikilotherms can adapt to a cold environment.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
