Purpose

The purpose of this study was to quantify the spatial and technological variability in life cycle greenhouse gas (GHG) emissions, also called the carbon footprint, of durum wheat production in Iran.

Methods

The calculations were based on information gathered from 90 farms, each with an area ranging from 1 to 150 ha (average 16 ha). The carbon footprint of durum wheat was calculated by quantifying the biogenic GHG emissions of carbon loss from soil and biomass, as well as the GHG emissions from fertilizer application and machinery use, irrigation, transportation, and production of inputs (e.g., fertilizers, seeds, and pesticides). We used Spearman’s rank correlation to quantify the relative influence of technological variability (in crop yields, fossil GHG emissions, and N2O emissions from fertilizer application) and spatial variability (in biogenic GHG emissions) on the variation of the carbon footprint of durum wheat.

Results and discussion

The average carbon footprint of 1 kg of durum wheat produced was 1.6 kg CO2-equivalents with a minimum of 0.8 kg and a maximum of 3.0 kg CO2-equivalents. The correlation analysis showed that variation in crop yield and fertilizer application, representing technological variability, accounted for the majority of the variation in the carbon footprint, respectively 76 and 21%. Spatial variation in biogenic GHG emissions, mainly resulting from differences in natural soil carbon stocks, accounted for 3% of the variation in the carbon footprint. We also observed a non-linear relationship between the carbon footprint and the yield of durum wheat that featured a scaling factor of ?2/3. This indicates that the carbon footprint of durum wheat production (in kg CO2-eq kg?1) typically decreases by 67% with a 100% increase in yield (in kg ha?1 year?1).

Conclusions

Various sources of variability, including variation between locations and technologies, can influence the results of life cycle assessments. We demonstrated that technological variability exerts a relatively large influence on the carbon footprint of durum wheat produced in Iran with respect to spatial variability. To increase the durum wheat yield at farms with relatively large carbon footprints, technologies such as site-specific nutrient application, combined tillage, and mechanized irrigation techniques should be promoted.

The human papillomavirus (HPV) is the main cause of cervical cancer in developing countries. Rapid diagnosis and initiation of treatment of the HPV infection are critical. Various methods have been employed to reduce the immunogenicity of antibodies targeting HPV serotypes. Nanobodies are the smallest fragments of naturally occurring single-domain antibodies with their antigenbinding site compromised into a single domain. Nanobodies have remarkable properties such as high stability, solubility, and high homology to the human VH3 domain. In this study, a phagemid library was employed to enrich for nanobodies against the L1 protein of the human papilloma virus. Binding reactivity of the selected clones was evaluated using phage enzyme-linked immunosorbent assay (phage-ELISA). Finally, two nanobodies (sm5 and sm8) with the best reactivity against the Gardasil vaccine and the purified HPV-16 L1 protein were expressed and purified using a Ni(+)-NTA column. The accuracy of expression and purification of the nanobodies was confirmed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblotting assays. In vitro studies demonstrated that neutralization was achieved by the selected nanobodies. The ease of generation and unique features of these molecules make nanobodies promising molecules for the new generation of HPV diagnosis and therapy.  相似文献

Objective

Obesity is a risk factor for the development of insulin resistance and is one of the most important contributors to the pathogenesis of type2 diabetes, which acts mainly through the secretion of adipokines such as TNF-α that may influence insulin sensitivity. TNF-α affects many aspects of adipocyte function, such as adipocyte development and lipid metabolism.

Material and Methods

We demonstrated that there is a correlation between the expressions of TNF-α in retroperitoneal WAT and insulin-resistance in 8 genetically obese fa/fa rats. Treatment of animals with CL 316,243, a β3-adrenergic agonist, showed an improvement of insulin-resistance that was linked with the suppression of TNF-α mRNA expression in WAT.

Results

These results confirm the association between TNF-α expression and the insulin-resistant condition in rats. Our finding indicates that the hyperglycaemia and hyperinsulinemia induced by insulin-resistance correlated positively with the expression of TNF-α mRNA in an abdominal WAT depot.

Conclusion

We conclude that CL 316,243 possesses both anti-diabetic effects and anti-obesity effects in rodents.  相似文献

Evaluation of metabolic pathways is one of the challenging areas in biological and pharmaceutical sciences. Phenanthridine oxidation to phenanthridinone is used commonly to study aldehyde oxidase activity. This reaction could pass through phenanthridine N-oxide intermediate. In the present study, the application of multivariate curve resolution, optimized by alternating least squares (MCR-ALS) to investigate this metabolic pathway has been described. The results obtained from MCR-ALS analysis along with those obtained from the use of potassium ferrocyanide method indicated that phenanthridine is directly oxidized to phenanthridinone by rat liver aldehyde oxidase without passing through phenanthridine N-oxide intermediate. It was also found that the later compound is not metabolized by this enzyme.  相似文献
