The function of the excretory system of the teredinid bivalveNausitora fusticula is discussed on the basis of new informationon histology and ultrastructure. The wall of the auricles islined with podocytes that allow haemolymph ultrafiltration tothe pericardial cavity. These podocytes also show apical microvilliwith absorptive activity. The primary urine is drained fromthe pericardial cavity to the afferent ducts by a citiated bulb-likestructure. Theafferent and efferent ducts together form thekidney body. The afferent duct shows structures related to absorption,excretion and conduction of the urine. The efferent ducts, however,have structures concerned only with urine absorption and conduction. Abstract.
  • 1 The amino acid content of phloem exudates from leaves and of aphid honeydew were adopted as indices of the nutritional quality of phloem sap for aphids. Four plant species and associated leaf-dwelling aphids were investigated: the sycamore Acer pseudoplanatus and sycamore aphid Drepanosiphum platanoides; Prunus domestica (victoria plum) and the mealy plum aphid Hyalopterus pruni; and the spindle tree Euonymus europaeus and broad-bean Vicia faba, both hosts of the black bean aphid Aphis fabae.
  • 2 The concentration of amino acids in the phloem exudates varied with: (a) plant species (greater in the herb Vicia than in the tree species), (b) season (greater in the autumn than summer for Acer and Euonymus), and (c) position (greater in flush leaves than mature leaves of Prunus).
  • 3 For Acer and Prunus and their aphids, the concentration of amino acids in phloem exudates was significantly correlated with the amino acid content of the aphid honeydew.
  • 4 The amino acids in all exudates and honeydew were dominated by non-essential amino acids (glutamic acid, glutamine, asparagine or serine, varying with season and between plant species). The sole major discrepancy between the amino acid profiles of exudates and honeydew was the production of asparagine-rich honeydew by aphids feeding on leaves, whose exudates were dominated by glutamic acid; this applied to both H.pruni on mature Prunus leaves and Drepanosiphum platanoides on summer-leaves of Acer.
  • 5 It is suggested that EDTA-exudation may be a useful technique to study nutritional correlates of aphid life cycles, e.g. the time of migration between primary and secondary plant hosts.

European larch (Larix decidua Mill.) seedlings were inoculatedwith a number of Agrobacterium strains to screen susceptibilityto infection by agrobacteria. Nine of fourteen Agrobacteriumstrains tested were virulent on this conifer species. The attachmentof virulent Agrobacterium to larch seedling tissues was examinedusing scanning electron microscopy. Electron micrographs showedthe attachment specificity of virulent Agrobacterium to larchcells at wound sites in a manner similar to that described forcells of susceptible dicotyledonous angiosperms, indicatinga host-parasite relationship between oncogenic Agrobacteriumspp. and young seedlings of European larch. These unique electronmicrographs provide the first opportunity to document the compatibleinteractions of Agrobacterium and a conifer at the cellularlevel. Further, the evaluation of tumour formation frequencysuggested that the interactions between Agrobacterium and Europeanlarch were affected by wound site, position of inoculation,age of plant tissues, and time of co-culture. Successful infectionof Agrobacterium resulted in genetic transformation of hostcells. Agrobacterium-mediated DNA transfer and expression of bacterialgenes in larch tissues were confirmed by both Southern blotanalysis and opine assay with the transformed tissues. The rapid diversification of early Metazoa remains one of the most puzzling events in the fossil record. Several models have been proposed to explain a critical aspect of this event: the origin of Metazoan development. These include the origin of the eukaryotic cell, environmental triggers, key innovations or selection among cell lineages. Here, the first three hypotheses are evaulated within a phylogenetic framework using fossil, molecular and developmental evidence. Many elements of metazoan development are widely distributed among unicellular eukaryotes, yet only 3 of the 23 multicellular eukaryotic lineages evolved complex development. Molecular evidence indicates the lineage leading to the eukaryotic cell is nearly as old as the eubacterial and archaebacterial lineages, although the symbiotic events established that the eukaryotic cell probably occurred about 1.5 billion years ago. Yet Metazoa did not appear until 1000 to 600 million years ago (Myr), suggesting the origin of metazoan development must be linked to either an environmental trigger, perhaps an increase in atmospheric oxygen, or key innovations such as the development of collagen. Yet the first model fails to explain the unique appearance of complex development in Metazoa, while the latter fails to explain the simultaneous diversification of several ‘protist’ groups along with the Metazoa. A more complete model of the origin of metazoan development combines environmental triggering of a series of innovations, with successive innovations generating radiations of metazoan clades as lineages breached functional thresholds. The elaboration of new cell classes and the appearance of such developmental innovations as cell sheets may have been of particular importance. Evolutionary biologists often implicitly assume that evolution is a uniformitarian, time-homogeneous process without strong temporal asymmetries in evolutionary mechanisms, rate or context. Yet evolutionary patterns do exhibit such asymmetries, raising the possibility that such innovations as metazoan development impose non-uniformities of evolutionary process.  相似文献

SYNOPSIS. Lepidopteran juvenile hormone (JH) esterase appearsto have a functional role in the regulation of embryogenesis,larval growth and development, and adult reproduction. In preovipositionaland newly laid eggs of the tobacco hornworm, Manduca sexta,JH esterase activity was elevated presumably to metabolize maternalJHs, and then declined after blastoderm formation. Also, a singlepeak in hemolymph JH esterase activity was found prior to ecdysisin the second through the fourth instar of M. sexta, the functionof which is unclear. However, in the last instar, elevated hemolymphJH esterase activity was noted prior to wandering and againprior to ecdysis to scavenge the last traces of JH necessaryfor normal development. The hemolymph JH esterase is likelyof multiple tissue origin for the prewandering peak with thefat body excluded as a source for the prepupal peak; an inhibitoryfactor from the brain and JH regulate JH esterase biosynthesis.In adult cabbage loopers, Trichoplusia ni, elevated hemolymphJH esterase activity appeared to be important in reducing theJH titer and preventing egg maturation. Structure/activity datawith trifluoromethylketones were incorporated into the designof a novel, JH esterase inhibitor, the sulfone and hydrate ofoctylthio-1,1,1- trifluoropropan-2-one, with selective and persistent,in vivo inhibitory activity. The topical application of thiscompound to last instar larvae and virgin adults of T. ni producedjuvenizing effects (delayed pupation and induced egg maturation/oviposition,respectively) providing direct evidence of a functional rolefor JH esterase in lepidopteran development.  相似文献

Cultured human melanocytes derived from different skin types responded to frequent treatment with ultraviolet (UV) light with increased melanin synthesis, decreased proliferation, and morphologic signs of aging. These effects were augmented by increased frequency of irradiation with 15.5 mJ/cm2 UV light. Stimulation of melanogenesis by UV light involved an increase in tyrosinase activity, without any change in the amounts of either tyrosinase or tyrosinase-related protein (TRP)-1, and a decrease in the amount of TRP-2, as determined by Western blot analysis. These results are different from the mechanisms by which other melanogenic agents, such as cholera toxin and isobutyl methylxanthine, stimulated melanogenesis, whereby the amounts of tyrosinase, TRP-1 and TRP-2 were increased. The decrease in the amount of TRP-2 might be significant in that it might alter the properties of the newly synthesized melanin. The UV irradiation protocol that was followed blocked melanocytes in G2-M phase of the cell cycle without compromising cellular viability. Following three rounds of UV irradiation, melanocytes could recover from the growth arrest and resume proliferation. Treatment with 0.1 μM α-melanocyte stimulating hormone (α-MSH) postirradiation enhanced the melanogenic effect of UV light and stimulated the melanocytes to proliferate. The effects of α-MSH on the UV induced responses and their implications on photocarcinogenesis are being further investigated. Analyzing the mechanisms by which UV light exposure affects normal melanocytes might lead to a better understanding of how these cells undergo malignant transformation, and why individuals with different skin types differ in their susceptibility to skin cancers.  相似文献

Muscle spindles and tendon organs occur in most somatic musclesof the mammal and are particularly concentrated in muscles subservingfine movements, including postural muscles and small musclesof the distal extremities. In those mixed muscles in which thedifferent fiber and motor unit types are "compartmentalized,"the spindles, and perhaps tendon organs also, are virtuallylimited to those compartments predominated by "oxidative" musclefibers. These morphological observations based on a broad arrayof muscles in many species, complement electrophysiologicalstudies which have emphasized that (1) the "oxidative" motorunits have low reflex thresholds and (2) segmental proprioceptivereflexes may be primarily concerned with the control of finelygraded contractions. Consideration of the functional anatomyof the association between motor units and muscle receptorssuggests the need for detailed structural-functional analysesof those muscles with specializations in architecture, fiber-typecomposition and distribution, and in the number and distributionof their muscle spindles and tendon organs. An electrophysiologicalanalysis of the relationship between the spinal cord and suchmuscles might also reveal certain strategies and mechanismsunderlying segmental motor control which are either absent orobscured in the analysis of that select number of "homogenously-mixed"muscles conventionally used in the study of the mammalian segmentalmotor control system.  相似文献
