SYNOPSIS. A method is described for cultivation of large numbers of Spirostomum ambiguum , calcified intracellularly with hydroxyapatite (bone salt deposits). The structure and activity of living animals at various stages of calcification is described and also illustrated with unstained and silver impregnated ciliates. 1. The species composition and spatial distribution of small insects (Hemiptera, Coleoptera, Lepidoptera) and arachnids (Araneae, Opiliones, and Pseudoscorpiones) were investigated in three indigenous, upland grasslands identified as the National Vegetation Classification Festuca–Agrostis–Galium typical subcommunity (code U4a), Festuca–Agrostis–Galium, Vaccinium–Deschampsia subcommunity (code U4e), and Nardus stricta species-poor sub-community (code U5a), on which grazing management was manipulated experimentally. 2. Two hypotheses were tested that predicted arthropod diversity in upland grasslands. The habitat heterogeneity hypothesis predicts that the species number and abundance of arthropods will have an asymptotic relationship with increasing numbers of plant species and greater structural heterogeneity in the vegetation. The symbiosis between patches hypothesis states that the species number and abundance of arthropods will express a unimodal relationship with the grain size of sward patches created by grazing. The sward patches must be large enough to be apparent to, and support populations of, arthropods, but small enough that interspersed tussocks provide shelter from weather and a deterrent to disturbance by grazers. 3. The hypotheses were tested by sampling arthropods from the geometrical patterns represented by the individual tussocks and intermediate sward components of three indigenous grasslands produced by different grazing treatments. Paired samples of arthropods were taken by motorized suction sampler, the first of the pair from the grazed sward and the second, the accumulated samples from the surrounding triad of tussocks (U4a and U5a grasslands) or hummocks (U4e grassland). The paired samples were taken from six randomly-selected locations across both replicates of each of the grazing treatments. 4. Arthropod species composition and abundance were compared between the paired sward and tussock samples and in turn with measures of the vertical and horizontal components of vegetation structure, i.e. the variance in vegetation height per unit area and the area covered by tussock compared with sward. 5. There were consistently more species and a greater abundance of arthropods associated with tussocks than with swards and the average species number and abundance for the combined pair of samples declined with increased grazing pressure. The relationship between vertical and horizontal components of vegetation structure and the species number and abundance of selected arthropods was asymptotic as opposed to unimodal, supporting the habitat heterogeneity hypothesis, rather than the symbiosis between patches hypothesis. 6. Small and relatively sedentary insects and arachnids are more sensitive to grazing intensity and species of grazer in these upland, indigenous grasslands than are larger Coleoptera and Araneae, which respond less directly to varied grazing management. The overall linear reduction of small herbivorous and predatory arthropods with increased grazing intensity was buffered in grasslands with substantial tussock patches.  相似文献

Orchid seeds are minute and the first seedling stages of terrestrial species are underground, so there is little knowledge about the biology of the early life history and the size of seed/seedling populations relative to the number of plants that eventually emerge above ground. A recently developed field sowing technique makes it possible to explore a variety of topics such as the length of time that seeds spend as part of the soil seed bank, the phenology of seed germination, substrate requirements for germination, dependence of germination on fungal availability, substrate requirements and growth rate of seedlings, seedling phenology and development, and seedling-fungus specificity patterns in the soil. Several of these applications are relevant for conservation issues. Studies of Goodyera pubescens, Corallorhiza odontorhiza and Liparis lilifolia show considerable variation in germination strategies, patterns of seedling development and establishment, substrate requirements, and levels of specificity with the fungi.  相似文献

Mitochondrial and nuclear genes have different inheritance, thus studies of fungal populations should use both mitochondrial and nuclear markers. Using nuclear markers, the S238N strain of the ectomycorrhizal basidiomycete Laccaria bicolor ((Maire) Orton) has been previously shown to persist for at least 10 yr after outplanting in a plantation of Douglas fir ( Pseudotsuga menziesii (Mir.) Franco) inoculated with this strain. In the present study, we have sampled 539 sporophores of Laccaria spp. from this plantation, some of which had the S238N nuclear genotype, to study mitochondrial DNA polymorphism and persistence of the inoculated S238N mitochondrial genome. Length polymorphism in fragments of the large subunit of mitochondrial ribosomal DNA (LrDNA) allowed distinction of the haplotypes present in the plantation at the species level. In addition, heteroduplex analysis and sequencing revealed intraspecific polymorphism of LrDNA among the L. bicolor sporophores and enabled specific identification of S238N LrDNA. This haplotype was only retained in sporophores carrying the S238N nuclear genome, confirming the survival of this introduced strain in a natural population.  相似文献

The eddy covariance technique ascertains the exchange rate of CO2 across the interface between the atmosphere and a plant canopy by measuring the covariance between fluctuations in vertical wind velocity and CO2 mixing ratio. Two decades ago, the method was employed to study CO2 exchange of agricultural crops under ideal conditions during short field campaigns. During the past decade the eddy covariance method has emerged as an important tool for evaluating fluxes of carbon dioxide between terrestrial ecosystems and the atmosphere over the course of a year, and more. At present, the method is being applied in a nearly continuous mode to study carbon dioxide and water vapor exchange at over a hundred and eighty field sites, worldwide. The objective of this review is to assess the eddy covariance method as it is being applied by the global change community on increasingly longer time scales and over less than ideal surfaces. The eddy covariance method is most accurate when the atmospheric conditions (wind, temperature, humidity, CO2) are steady, the underlying vegetation is homogeneous and it is situated on flat terrain for an extended distance upwind. When the eddy covariance method is applied over natural and complex landscapes or during atmospheric conditions that vary with time, the quantification of CO2 exchange between the biosphere and atmosphere must include measurements of atmospheric storage, flux divergence and advection. Averaging CO2 flux measurements over long periods (days to year) reduces random sampling error to relatively small values. Unfortunately, data gaps are inevitable when constructing long data records. Data gaps are generally filled with values produced from statistical and empirical models to produce daily and annual sums of CO2 exchange. Filling data gaps with empirical estimates do not introduce significant bias errors because the empirical algorithms are derived from large statistical populations. On the other hand, flux measurement errors can be biased at night when winds are light and intermittent. Nighttime bias errors tend to produce an underestimate in the measurement of ecosystem respiration. Despite the sources of errors associated with long‐term eddy flux measurements, many investigators are producing defensible estimates of annual carbon exchange. When measurements come from nearly ideal sites the error bound on the net annual exchange of CO2 is less than ±50 g C m?2 yr?1. Additional confidence in long‐term measurements is growing because investigators are producing values of net ecosystem productivity that are converging with independent values produced by measuring changes in biomass and soil carbon, as long as the biomass inventory studies are conducted over multiple years.  相似文献

A model is described that explains the evolution of adult butterfly settling posture. The role of climate and the activity of butterflies is fundamental in determining the effectiveness of primary and secondary visual defence systems based on wing colour and pattern. Both dorsal (open winged) and lateral (closed wing) postures can be regarded as evolutionarily conservative; common postures are found within almost all sub-families. Lateral postures tend to be associated with cool temperate and arctic climates. Conditions that are marginal for activity place an emphasis on the development of underside primary defence, uncompromised by secondary visual defences. In turn, any primary defensive function of the upper surface is reduced but secondary defence may be emphasized. There is interaction between segregation of functions on different wing surfaces and the development of lateral posture. The use of predictable hostplant-habitat structures reduces the role of visual communication in mate-location and facilitates the adoption of a lateral resting posture and a cryptic underside wing pattern. When hostplant-habitat structures are unpredictable but a lateral posture is adopted, primary defence is more important than visual communication for the resting insect. Adjustments of wing morphology facilitate postural conservatism, allowing species to retain particular postures in new hostplant-habitats and climatic zones.  相似文献

A density-based load estimation method was applied to determine femoral load patterns. Two-dimensional finite element models were constructed using single energy quantitative computed tomography (QCT) data from two femora. Basic load cases included parabolic pressure joint loads and constant tractions on the greater trochanter. An optimization procedure adjusted magnitudes of the basic load cases, such that the applied mechanical stimulus approached the ideal stimulus throughout each model. Dominant estimated load directions were generally consistent with published experimental data for gait. Other estimated loads suggested that loads at extreme joint orientations may be important to maintenance of bone structure. Remodeling simulations with the estimated loads produced density distributions qualitatively similar to the QCT data sets. Average nodal density errors between QCT data and predictions were 0.24 g/cm(3) and 0.28 g/cm(3). The results indicate that density-based load estimation could improve understanding of loading patterns on bones.  相似文献

ABSTRACT. A soluble enzyme amylopectin synthase (UDP-glucose-α 1,4-glucan α-4-glucosyltransferase) which transfers glucose from uridine 5'-diphosphate glucose (UDP-glucose) to a primer to form α-I,4-glucosyl linkages has been identified in the extracts of unsporulated oocysts of Eimeria tenella . UDP-glucose and not ADP-glucose was the most active glucosyl donor. Corn amylopectin, rabbit liver glycogen, oyster glycogen and corn starch served as primers; the latter two were less efficient. The enzyme has an apparent pH optimum of 7.5 and exhibited typical Michaelis-Menten kinetics with dependence on both the primer and substrate concentrations. The Michaelis constants (Km). with respect to UDP-glucose, was 0.5 mM; and 0.25 mg/ml and 1.25 mg/ml with respect to amylopectin and rabbit liver glycogen. The product formed by the reaction was predominantly a glucan containing α-1,4 linkages. The specificity of the enzyme suggests that this enzyme is similar to glycogen synthase in eukaryotes and has been designated as amylopectin synthase (UDP-glucose-α-1,4-glucosetransferase EC 2.4.1.11).  相似文献
