Abstract: Animal locations estimated by Global Positioning System (GPS) inherently contain errors. Screening procedures used to remove large positional errors often trade data accuracy for data loss. We developed a simple screening method that identifies locations arising from unrealistic movement patterns. When applied to a large data set of moose (Alces alces) locations, our method identified virtually all known errors with minimal loss of data. Thus, our method for screening GPS data improves the quality of data sets and increases the value of such data for research and management.  相似文献

Sensitization of adenylyl cyclase (AC) signaling has been implicated in a variety of neuropsychiatric and neurologic disorders including substance abuse and Parkinson''s disease. Acute activation of Gαi/o-linked receptors inhibits AC activity, whereas persistent activation of these receptors results in heterologous sensitization of AC and increased levels of intracellular cAMP. Previous studies have demonstrated that this enhancement of AC responsiveness is observed both in vitro and in vivo following the chronic activation of several types of Gαi/o-linked receptors including D2 dopamine and μ opioid receptors. Although heterologous sensitization of AC was first reported four decades ago, the mechanism(s) that underlie this phenomenon remain largely unknown. The lack of mechanistic data presumably reflects the complexity involved with this adaptive response, suggesting that nonbiased approaches could aid in identifying the molecular pathways involved in heterologous sensitization of AC. Previous studies have implicated kinase and Gbγ signaling as overlapping components that regulate the heterologous sensitization of AC. To identify unique and additional overlapping targets associated with sensitization of AC, the development and validation of a scalable cAMP sensitization assay is required for greater throughput. Previous approaches to study sensitization are generally cumbersome involving continuous cell culture maintenance as well as a complex methodology for measuring cAMP accumulation that involves multiple wash steps. Thus, the development of a robust cell-based assay that can be used for high throughput screening (HTS) in a 384 well format would facilitate future studies. Using two D2 dopamine receptor cellular models (i.e. CHO-D2L and HEK-AC6/D2L), we have converted our 48-well sensitization assay (>20 steps 4-5 days) to a five-step, single day assay in 384-well format. This new format is amenable to small molecule screening, and we demonstrate that this assay design can also be readily used for reverse transfection of siRNA in anticipation of targeted siRNA library screening.  相似文献

Saccharomyces cerevisiae strains carrying a mutation in the clathrin heavy chain gene (CHC1) are genetically unstable and give rise to heterogeneous populations of cells. Manifestations of the instability include increases in genome copy number as well as compensatory genetic changes that allow better growing clathrin-deficient cells to take over the population. Increases in genome copy number appear to result from changes in ploidy as well as alterations in normal nuclear number. Genetic background influences the frequency at which cells with increased genome content are observed in different Chc(-) strains. We cannot distinguish whether genetic background affects the rate at which aberrant nuclear division events occur or a growth advantage of cells with increased nuclear and/or genome content. However, survival of chc1-Δ cells does not require an increase in genome copy number. The clathrin heavy chain gene was mapped 1-2 cM distal to KEX1 on the left arm of chromosome VII by making use of integrated 2μ plasmid sequences to destabilize distal chromosome segments and allow ordering of the genes.  相似文献

1. Omnivorous predators can protect plants from herbivores, but may also consume plant material themselves. Omnivores and their purely herbivorous prey have previously been thought to respond similarly to host‐plant quality. However, different responses of omnivores and herbivores to their shared host plants may influence the fitness, trophic identity, and population dynamics of the omnivores. 2. The aim of the present study was to show that an omnivorous heteropteran (Anthocoris nemorum L.) and two strictly herbivorous prey species respond differently to different genotypes of their shared host plant, Salix. Some plant genotypes were sub‐optimal for the omnivore, although suitable for the herbivores, and vice versa. 3. The contrasting patterns of plant suitability for the omnivore and the herbivores highlight an interaction between plant genotype and omnivores' access to animal food. Plant genotypes that were sub‐optimal for the omnivore when herbivores were experimentally excluded became the best host plants when herbivores were present, as in the latter situation additional prey became available. By contrast, the quality of plant genotypes that were intrinsically suitable for omnivores, did not improve when herbivores were present as these plant genotypes were intrinsically sub‐optimal for herbivores, thus providing omnivores with almost no additional animal food. 4. The differential responses of omnivores and their prey to the same host‐plant genotypes should allow omnivores to colonise sub‐optimal host plants in their capacity as predators, and to colonise more suitable host plants in their capacity as herbivores. It may thus be difficult for Salix to escape herbivory entirely, as it will rarely be unsuitable for both omnivores and pure herbivores at the same time.  相似文献

The primary hypothesis of this study was that in the Galápagos Islands, fruit and seed set via nocturnal pollination would exceed that of diurnal pollination because of the greater insect activity at night typical of hot, arid regions. Clerodendrum molle, a widespread member of the Galápagos flora was submitted to pollination experiments, visitor observations, nectar sampling, pollen transfer studies, pollen?Covule ratio studies, and pollen measurements. Flowers set fruit and seed via open pollination, autonomous autogamy, facilitated autogamy, facilitated cross-pollination, diurnal pollination, and nocturnal pollination. Results indicate no significant difference in fruit set. Cross-pollinated flowers showed a significant increase in seed set over all treatments except facilitated autogamy. Nocturnal and diurnal fruit and seed set did not differ significantly. Nocturnal visitors included ants, spiders, hawk moths, and roaches whereas diurnal visitors included carpenter bees and ants. Galápagos members of C. molle exhibit incomplete protandry and set fruit via autonomous autogamy as a result of natural selection in an environment with few faithful pollinators. Overall low fruit and seed set are likely because of a combination of pollen and resource limitation, the latter exacerbated by nectar robbing. Studies of island angiosperms are crucial for complete understanding of the population dynamics of indigenous plants, which can assist conservation officials in protecting these species.  相似文献

Earlier field and laboratory studies have shown that Legionella species survive and multiply in the pH range 5.5 to 9.2. Additionally, the technical feasibility of operating cooling towers at elevated alkalinities and pH has previously been documented by published guidelines. The guidelines indicate that these conditions facilitate corrosion control and favor chlorine persistence which enhances the effectiveness of continuous chlorination in biofouling control. This information suggests that control of Legionella species in cooling towers can be accomplished by operating the towers under alkaline conditions. To test this possibility, we collected water samples over a period of months from a hospital cooling tower. The samples were analyzed for a variety of chemical parameters. Subsamples were pasteurized and inoculated with non-agar-passaged Legionella pneumophila which had been maintained in tap water. Correlation of subsequent Legionella growth with corresponding pH and alkalinity values revealed statistically significant inverse associations. These data support the hypothesis that operating cooling towers outside of the optimal conditions for Legionella growth (e.g., at elevated alkalinities and a pH greater than 9) may be a useful approach to controlling growth in this habitat.  相似文献
