Comparisons among the primary sequences of five cloned eukaryotic esterases
reveal two distinct lineages, neither bearing any significant overall
sequence similarity to the functionally related serine protease multigene
family. We have not eliminated the possibility that the esterases may have
residual conformational similarities to the serine proteases. However, our
profile analysis and analyses of the predicted conformations of the
esterases reveal little similarity to the serine proteases. Four of the
esterase proteins share 27%-53% overall sequence similarity and evidence of
a catalytic mechanism involving the same Arg- Asp-Ser or His-Asp-Ser charge
relay. We propose that these four esterases, three of them cholinesterases,
form part of a multigene family essentially separate from the serine
proteases.

Protonemata of Funaria hygrometrica grown in artificial mediacontaining different lead concentrations grow more slowly thancontrols and show a disturbance of polar growth, changed arrangementof chloroplasts, alterations of nucleus and septa position.Morphological effects are dose-dependent. At the lowest leadconcentration (10-6 M), only a delay in development was observed,but no cellular alterations, At 10-5 M Pb nuclear migration,cellular shape, size and position of plastids, were alteredand a variety of aberrant forms were present. At 10-4 M, besidesthese alterations, a drastic reduction of the protonemal system,high vacuolation and the growth of protonemal filaments fromleaves were evident. The highest concentration, (10-3 M), causeddeath. Patterns of protonemal development and cellular arrangementin lead-treated samples showed similarities as well as differences,if compared to alterations induced by colchicine. Indirect immunofluorescencedemonstrated a correlation between lead concentration and alterationof cytoskeletal organization (alterations similar to those inducedby colchicine). We have isolated from calf thymus glands a new class of single-stranded DNA binding proteins (DBP) specifically endowed with the ability to stimulate DNA polymerase α activity . Such result was attained by using a partially new purification procedure and a functional assay, i.e. stimulation of DNA polymerase α on poly(dAT) throughout purification. We observed stimulations up to 30-fold of DNA polymerase α on poly(dAT) at a protein: DNA ratio of 1:1, that is much below the saturating level. Such stimulation is specific for DNA polymerase α. These results indicate a possible direct functional interaction between our DBP, polymerase α and the template. 相似文献
