The biosynthesis of ethylene was examined in suspension-cultured cells of parsley (Petroselinum hortense) treated with an elicitor from cell walls of Phytophthora megasperma. Untreated cells contained 50 nmol g-1 of the ethylene precursor, 1-aminocyclopropane-1-carboxylic acid (ACC), and produced ethylene at a rate of about 0.5 nmol g-1 h-1. Within 2 h after addition of elicitor to the culture medium, the cells started to produce more ethylene and accumulated more ACC. Exogenously added ACC did not increase the rate of ethylene production in control or elicitor-treated cells, indicating that the enzyme converting ACC to ethylene was limiting in both cases. The first enzyme in ethylene biosynthesis, ACC synthase, was very rapidly and transiently induced by the elicitor treatment. Its activity increased more than tenfold within 60 min. Density labelling with 2H2O showed that this increase was caused by the denovo synthesis of the enzyme protein. Cordycepin and actinomycin D did not affect the induction of ACC synthase, indicating that the synthesis of new mRNA was not required. The peak of ACC-synthase activity preceded the maximal phenylalanine ammonia-lyase (PAL) activity by several hours. Exogenously supplied ethylene or ACC did not induce PAL. However, aminoethoxyvinylglycine, an inhibitor of ACC synthase, suppressed the rise in ethylene production in elicitor-treated cells and partially inhibited the induction of PAL. Exogenously supplied ACC reversed this inhibition. It is concluded that induction of the ethylene biosynthetic pathway is a very early symptom of elicitor action. The intracellular response of the ocellar nerve dendrite, the second order neuron in the retina of the dragonfly ocellus, has been modified by application of various drugs and a model developed to explain certain features of that response. Curare blocked the response completely. Both picrotoxin and bicuculline eliminated the "off" overshoot. Bicuculline also decreased the size of response and the sensitivity. gamma-Aminobutyric acid (GABA), however, increased the size of response. The evidence indicates the possibility that the receptor transmitter is acetylcholine and is inhibitory to the ocellar nerve dendrite whereas the feedback transmitter from the ocellar nerve dendrite may be GABA and is facilitory to receptor transmitter release. The model of synaptic feedback interaction developed to be consistent with these results has certain important features. It suggests that the feedback transmitter is released in the dark to increase input sensitivity from receptors in response to dim light. This implies that the dark potential of the ocellar nerve dendrite may be determined by a dynamic equilibrium established by synaptic interaction between it and the receptor terminals. Such a system is also well suited to signalling phasic information about changes in level of illumination over a wide range of intensities, a characteristic which appears to be a significant feature of the dragonfly median ocellar response.  相似文献

The similarity transformation approach is used to analyze the structural identifiability of the parameters of a nonlinear model of microbial growth in a batch reactor in which only the concentration of microorganisms is measured. It is found that some of the model parameters are unidentifiable from this experiment, thus providing the first example of a real-life nonlinear model that turns out not to be globally identifiable. If it is possible to measure the initial concentration of growth-limiting substrate as well, all model parameters are globally identifiable.  相似文献

Squalene synthetase, an integral membrane protein and the first committed enzyme for sterol biosynthesis, was solubilized and partially purified from tobacco (Nicotiana tabacum) cell suspension cultures. Tobacco microsomes were prepared and the enzyme was solubilized from the lipid bilayer using a two-step procedure. Microsomes were initially treated with concentrations of octyl-β-d-thioglucopyranoside and glycodeoxycholate below their critical micelle concentration, 4.5 and 1.1 millimolar, respectively, to remove loosely associated proteins. Complete solubilization of the squalene synthetase enzyme activity was achieved after a second treatment at detergent concentrations above or at their critical micelle concentration, 18 and 2.2 millimolar, respectively. The detergent-solubilized enzyme was further purified by a combination of ultrafiltration, gel permeation, and Fast Protein Liquid Chromatography anion exchange. A 60-fold purification and 20% recovery of the enzyme activity was achieved. The partially purified squalene synthetase protein was used to generate polyclonal antibodies from mice that efficiently inhibited synthetase activity in an in vitro assay. The apparent molecular mass of the squalene synthetase protein as determined by immunoblot analysis of the partially purified squalene synthetase protein separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis was 47 kilodaltons. The partially purified squalene synthetase activity was optimal at pH 6.0, exhibited a Km for farnesyl diphosphate of 9.5 micromolar, and preferred NADPH as a reductant rather than NADH.  相似文献

A general effect by dissolved electrolytes to destabilize the curvature of bilayer tubules prepared from the diacetylenic phospholipid, 1,2-bis(10,12-tricosadiynoyl)-sn-glycero-3-phosphocholine is not found. This observation discounts the role of an electrostatic interaction between polarization charges on the edges of a ferroelectric bilayer as a means by which the cylindrical curvature may be stabilized in these structures (de Gennes, P. G. 1987. C. R. Acad. Sci. Paris. 304:259-263). The solution-mediated ionic interactions of electrolytes with this phospholipid appear not to influence significantly the relative stability of the crystalline state of the tubule, but at high levels of a few salts, may affect the nucleation and growth of the crystalline bilayer. Curvature of the bilayer in these tubular structures apparently derives from an interaction that is not very sensitive to the presence of electrolytes. Cylindrical curvature may alternatively arise from a bending force within the bilayer that is intrinsic to the anisotropic packing of the lipid molecules (Helfrich, W., and J. Prost. 1988. Phys. Rev. A38:3065-3068; Chappell, J. S., and P. Yager. 1991. Chem. Phys. Lipids. Summary Body temperatures (T b) and daily activity patterns of free-living arctic ground squirrells (Spermophilus undulatus) were determined via telemetry at a field site in northern Alaska. Simultaneous measurements were made of ambient temperature (T a), wind speed (V), and incident solar radiation. The operative environmental temperature (T e) for ground squirrels was obtained from fur-covered, thin metal taxidermic models of the animals. Standard operative temperature (T es), a comparative index of heat flow, was calculated from T e, V, and laboratory measurements of thermal conductivity.During the period of the study (August), S. undulatus were active for about 14 h per day (06.00 to 20.00 h). T b was high throughout the daily cycle, averaging 38–39°C. Circadian variations in T b were slight; average T b values dropped <1°C at night. Daytime T b fluctuations were not closely correlated to activity or to changes in environmental conditions. Air temperatures during the study were low, usually between 10 and 15°C during the day. However, T es in exposed areas was normally higher, even though skies were generally overcast. During periods of sunshine, T es may be as high as 34°C. The absence of nocturnal activity may result from increased costs of thermoregulation at night, which sharply reduces foraging efficiency. The high and stable body temperatures of S. undulatus probably result from thermoneutral daytime T es, low activity levels, and the use of well-insulated nests.  相似文献

Immature Schistosoma mansoni in mice are less susceptible to antimony therapy than adult worms. KSb tartrate inhibited phosphofructokinase (PFK) (EC 2.7.1.11) to a greater extent in extracts of 3-week-old worms than adults, and inhibited production of lactate in both immature and adult worms in vitro. In vivo, KSb tartrate was accumulated similarly by 3-week-old worms and by adults: measurements of hexosephosphate following drug treatment suggested similar inhibition of PFK in the two worm stages. If antimony acts by inhibition of PFK it is not clear why the young worms are more resistant to chemotherapy than adults.  相似文献

Spectral sensitivities were recorded intracellulary in median ocelli of Anax junius, Aeschnatuberculifera, and Libellulapulcella. All cells had peak sensitivities at 360 and 500 nm while UV-blue+green cells found only in Anax had a third peak sensitivity at 440 nm. Ratios of UV-to-green sensitivities varied from cell to cell in each ocellus, but no UV-only or green-only cells were recorded. Half of the cells tested had a reverse Purkinje shift: They were more sensitive in the green at low illuminations but more sensitive in the UV at high illuminations; their intensity-response curves at 370 and 520 nm crossed but became parallel for large responses. Wave-lengths 420 nm and shorter elicited a family of low intensity-response curves with one slope; wavelengths 440 nm and longer elicities a family of curves with another slope. Orange-adapting lights selectively adapted sensitivity in the green, but UV-adapting lights had little selective effect. Amounts of log-selective adaptation were proportional to log orange-adapting intensity. It is concluded that two spectral mechanisms can be recorded from each cell, possibly by coupling of UV and green cells or possibly because each cell contains two visual pigments. Selective chromatic adaptations may provide the ocellus with a kind of "authomatic color control," while the reverse Purkinje shift could extend the ocellus' sensitivity to prevailing skylight.  相似文献
