Abstract. Embryonic development from coeloblastula to fully developed larva was investigated in 8 Mediterranean homoscleromorph species: Oscarella lobularis, O. tuberculata, O. microlobata, O. imperialis, Plakina trilopha, P. jani, Corticium candelabrum , and Pseudocorticium jarrei. Morphogenesis of the larva is similar in all these species; however, cell proliferation is more active in species of Oscarella than in Plakina and C. candelabrum. The result of cell division is a wrinkled, flagellated larva, called a cinctoblastula. It is composed of a columnar epithelium of polarized, monoflagellated cells among which are scattered a few non-flagellated ovoid cells. The central cavity always contains symbiotic bacteria. Maternal cells are also present in O. lobularis, O. imperialis , and P. jarrei. In the fully developed larva, cell shape and dimensions are constant for each species. The cells of the anterior pole have large vacuoles with heterogeneous material; those of the postero-lateral zone have an intranuclear paracrystalline inclusion; and the flagellated cells of the posterior pole have large osmiophilic inclusions. Intercellular junctions join the apical parts of the cells, beneath which are other specialized cell junctions. A basement membrane underlying the flagellated cells lines the larval cavity. This is the first observation of a basement membrane in a poriferan larva. The basal apparatus of flagellated cells is characterized by an accessory centriole located exactly beneath the basal body. The single basal rootlet is cross striated. The presence of a basement membrane and a true epithelium in the larva of Homoscleromorpha—unique among poriferan clades and shared with Eumetazoa—suggests that Demospongiae could be paraphyletic.  相似文献

Summary The Belgian Frasnian carbonate mounds occur in three stratigraphic levels in an overall backstepping succession. Petit-Mont and Arche Members form the famous red and grey “marble” exploited for ornamental stone since Roman times. The evolution and distribution of the facies in the mounds is thought to be associated with ecologic evolution and relative sea-level fluctuations. Iron oxides exist in five forms in the Frasnian mounds; four are undoubtedly endobiotic organized structures: (1) microstromatolites and associated forms (blisters, veils...), possibly organized in “endostromatolites”; (2) hematitic coccoids and (3) non dichotomic filaments. The filaments resemble iron bacteria of theSphaerotilus-Leptothrix “group”; (4) networks of dichotomic filaments ascribable to fungi; (5) a red ferruginous pigment dispersed in the calcareous matrix whose distribution is related to the mound facies type. The endobiotic forms developed during the edification of the mounds, before cementation by fibrous calcite. The microbial precipitation of iron took place as long as the developing mounds were bathed by water impoverished in oxygen.  相似文献

The fatal character of the infection caused by inhalation of Bacillus anthracis spores results from a complex pathogenic cycle involving the synthesis of toxins by the bacterium. We have shown using immunofluorescent staining, confocal scanning laser microscopy and image cytometry analysis that the alveolar macrophage was the primary site of B. anthracis germination in a murine inhalation infection model. Bacillus anthracis germinated inside murine macrophage-like RAW264.7 cells and murine alveolar macrophages. Germination occurred in vesicles derived from the phagosomal compartment. We have also demonstrated that the toxin genes and their trans -activator, AtxA, were expressed within the macrophages after germination.  相似文献

The composition of the phytoplanktonic communities in the surface waters of the La Reunion-Kerguelen transect (from 38°36S to 46°33S) has been investigated under spring conditions (Antarès 3 cruise, France-JGOFS, 28 September–8 November, 1995). The study, conducted at six stations in the subtropical frontal zone, involved size fractionations (threshold: 2 μm). The large variations in the overall biomass and autotrophic carbon fixation, calculated via Rubisco activity measurements and expressed respectively in terms of μg chlorophyll (a + b + c) per liter and nmol fixed carbon dioxide per liter and per hour, were attributable only to phytoplanktonic cells of >2 μm, with a peak observed in the frontal zone. The picophytoplankton (<2 μm) biomass remained constant throughout the transect, but the evolution of the species composition of the picophytoplanktonic population, as calculated from flow cytometry measurements through this frontal zone, changed. This study provides evidence, for the first time in this area, of the disappearance of prochlorophytes from the south of the frontal zone (42–47°S). Picoeukaryotes (<2 μm) and cyanobacteria populations, resolved by flow cytometry, were present all along the transect. However, their abundance decreased southward up to the quasi-disappearance of cyanobacteria at the southernmost station (52°S) that is characteristic of antarctic waters. The presence of prochlorophytes that is exclusive to the subtropical surface waters, and the low carbon fixation activity associated with these waters, may be linked to the specific hydrological features encountered. In contrast, the marked reduction in the cyanobacteria and the abundance of picoeukaryotes along the north-south transect is more likely to be a result of the reduction in temperature through the frontal zone. A hallmark of advanced maternal age is a significant increase in meiotic chromosome segregation errors, resulting in early miscarriages and congenital disorders. These errors most frequently occur during meiosis I (MI). The spindle assembly checkpoint (SAC) prevents chromosome segregation errors by arresting the cell cycle until proper chromosome alignment is achieved. Unlike in mitosis, the SAC in oocytes is desensitized, allowing chromosome segregation in the presence of improperly aligned chromosomes. Whether SAC integrity further deteriorates with advancing maternal age, and if this decline contributes to increased segregation errors remains a fundamental question. In somatic cells, activation of the SAC depends upon Aurora kinase B (AURKB), which functions to monitor kinetochore–microtubule attachments and recruit SAC regulator proteins. In mice, oocyte‐specific deletion of AURKB (Aurkb cKO) results in an increased production of aneuploid metaphase II‐arrested eggs and premature age‐related infertility. Here, we aimed to understand the cause of the short reproductive lifespan and hypothesized that SAC integrity was compromised. In comparing oocytes from young and sexually mature Aurkb cKO females, we found that SAC integrity becomes compromised rapidly with maternal age. We show that the increased desensitization of the SAC is driven by reduced expression of MAD2, ZW10 and Securin proteins, key contributors to the SAC response pathway. The reduced expression of these proteins is the result of altered protein homeostasis, likely caused by the accumulation of reactive oxygen species. Taken together, our results demonstrate a novel function for AURKB in preserving the female reproductive lifespan possibly by protecting oocytes from oxidative stress.  相似文献

Bacillus anthracis lethal toxin (PALF) stimulated the proliferation of human peripheral blood T-cells in vitro. Activation of T-lymphocytes by PALF required the presence of monocytes and did not result from a collaborative effect between T-cells and B-cells. PALF acted directly on monocytes and independently of T-cells. The monocytes contributed to the proliferation of T-cells by secretion of mediator(s). The mitogenic activity of the lethal toxin was dependent on its metalloprotease activity.  相似文献

A number of 2,3-epoxy alcohol derivatives (1–16), obtained either as racemates or through the Sharpless asymmetric epoxidation reaction, were studied on a Chiralcel OD column. Nearly all compounds exhibit good enantioselective resolution on this chiral support. The order of elution of enantiomers is reversed between nerol and geraniol compounds. For 2,3-epoxy alcohols bearing a remote alkoxy (or silyloxy) group, the order of the enantiomeric elution alternates with the number n (n = 1–3) of methylenic groups present between the epoxide ring and the terminal OR (R = p − BrBn or OSitBuPh2) functionality. In the case of trans 2,3-epoxy alcohols for the same number n, the order of elution is reversed when changing the terminal group −OSi to −OR. Background Polycystic kidney disease (PKD) is an autosomal recessive disorder resulting from mutations in the PKHD1 gene on chromosome 6 (6p12), a large gene spanning 470 kb of genomic DNA.ObjectiveThe aim of the present study was to report newly identified mutations in the PKHD1 gene in two Iranian families with PKD.Materials and Methods Genetic alterations of a 3-month-old boy and a 27-year-old girl with PKD were evaluated using whole-exome sequencing. The PCR direct sequencing was performed to analyse the co-segregation of the variants with the disease in the family. Finally, the molecular function of the identified novel mutations was evaluated by in silico study. ResultsIn the 3 month-old boy, a novel homozygous frameshift mutation was detected in the PKHD1 gene, which can cause PKD. Moreover, we identified three novel heterozygous missense mutations in ATIC, VPS13B, and TP53RK genes. In the 27-year-old woman, with two recurrent abortions history and two infant mortalities at early weeks due to metabolic and/or renal disease, we detected a novel missense mutation on PKHD1 gene and a novel mutation in ETFDH gene.Conclusion In general, we have identified two novel mutations in the PKHD1 gene. These molecular findings can help accurately correlate genotype and phenotype in families with such disease in order to reduce patient births through preoperative genetic diagnosis or better management of disorders.  相似文献
