Phylogenetic studies suggest that mobile element families are unstable components of the Drosophila genome. Two examples of immobilization of a transposable element family are presented here: as judged by their constant genomic organization among unrelated strains, the F and I element families have been respectively immobilized for a long time in D. simulans and in the reactive D. melanogaster strains (these are the laboratory strains which escaped the recent I invasion of D. melanogaster natural populations). All the elements of these defective families are located in the heterochromatic portion of the genome. Moreover, most if not all of the heterochromatic sequences into which the defective I elements are embedded are themselves non-mobile members of various nomadic families such as mdg 4, 297, 1731, F and Doc. These results are discussed with special emphasis on the possible nomadic origin of heterochromatin components and on the mechanisms of evolutionary turnover of the transposable element families.  相似文献

Bacteria indicating faecal contamination, cell-culturable enteroviruses and hepatitis A virus (HAV) were investigated in sea-water and in mussels exposed in an unpolluted marine environment, over a 7-month period with two samplings per month. Of the 16 mussel samples examined, none contained cell-culturable enteroviruses, four showed a low-level contamination by HAV and two did not conform to the current bacteriological norms. No connection was observed between the viral and bacterial contamination. No viral contamination was detected in the sea-water samples, but two gave bacterial counts above current norms.  相似文献

A new fluorescent acridine orange derivative, 3-amino-6-methoxy-9-(2-hydroxyethylamino) acridine (AMHA), has been applied to Hela cells in order to set up appropriate conditions for the detection of mycoplasma contaminations. Since AMHA staining reveals intensely fluorescent nuclei and slight fluorescent cytoplasm, we can visualize and localize mycoplasma contamination on each cell. In combination with a shortened Chen's staining method (1977), AMHA should allow a better detection of mycoplasma in animal cell cultures than the well established Hoechst dye.  相似文献

Summary The effect of substrate (buckwheat seeds) pretreatment on the growth and the sporulation behaviour of Penicillium roqueforti is presented. When a saccharifying enzyme (-amylase) is added to a medium which exhibits a low water content (0.46 g water/g initial dry matter, IDM), a more rapid internal colonization of the seeds occurs, but the final spore production does not increase and remains close to 8.109 spores/g dry matter (DM) at 500 h. No carbon source limitation is then observed. The addition of casein hydrolysate to this medium gives rise to a great increase of the sporulation, since 14.5 109 spores/g DM are obtained after 600 h. This result is attained by a better spore yield from the mycelium, the substrate colonization being unchanged. High water content (0.60 g water/g IDM) of buckwheat seeds induces a shorter cultivation time along with a higher biomass production. However, the spore content of the medium remains close to the low water content one, but 60% total spores are external against 30% to 35% in the other media.  相似文献

Using the synthetic peptide substrate Kemptide and cytosolic extracts of mouse fibroblasts transfected with a human insulin receptor cDNA construct, we have studied an insulin-sensitive serine kinase activity. This activity is rapidly stimulated by insulin (maximum within 5 min) and also by orthovanadate. During cell extract preparation, paranitrophenylphosphate and phosphotyrosine are able to preserve the enzyme activity, while phosphothreonine and phosphoserine fail to do so. Using antiphosphotyrosine antibodies, specific immunoprecipitation of this insulin- and orthovanadate-sensitive serine kinase was obtained. We then analysed by gel filtration chromatography eluates containing tyrosine-phosphorylated proteins obtained from unstimulated, insulin- and vanadate-treated cells. We found that several activities, with molecular weights estimated to be 30 kDa and smaller, are stimulated by both, insulin and orthovanadate. Thirty-two strains of opine-utilizing rhizobacteria were evaluated for physiological traits which have been related to plant growth-promoting activity. Tests included antibiosis against two bacterial and eight fungal pathogens of potato (Solanum tuberosum L.), production of hydrogen cyanide and fluorescent pigment production. On average, 71 and 12% of the bacteria inhibited the growth of Erwinia carotovora subsp. carotovora and Agrobacterium tumefaciens, respectively. The growth of Botrytis sp. was inhibited by 62% of the bacteria, and half of these produced an inhibition zone of more than 7 mm in diameter. Fusarium solani, Colletotrichum coccodes, Phoma exigua, Verticillium dahliae, F. oxysporum, V. albo-atrum and F. sambucinum were antagonized by 43, 34, 31, 25, 19, 18, and 12% of the bacteria, respectively. Only four strains produce hydrogen cyanide. The inhibition of a plant pathogen was not correlated to the production of fluorescent pigment. No strain produced a hypersensitive reaction whereas only three strains induced soft-rot and two produced polygalacturonase. Some opine-utilizing rhizobacteria were strong inhibitors of all plant pathogens, while most were active against specific plant pathogens.  相似文献

Transgenic tomato plants expressing antisense RNA to a ripening-related cDNA clone (pTOM5) had yellow ripening fruit and pale coloured flowers. Carotenoid levels in fruit of these plants were reduced by up to 97%. In order to determine the step of carotenoid biosynthesis which was blocked, a cell-free system active in the synthesis of carotenoid intermediates was prepared. Incubations with radiolabelled carotenoid precursors led to the identification of the block between GGDP and phytoene. Analysis of carotenoids in different tissues of transgenic and control plants indicated that although ripe fruit and flower carotenoid levels were reduced in the modified plants, leaf carotenoid levels were not decreased. This implies that the pTOM5 gene product is not involved in carotenoid synthesis in the leaf.  相似文献

5-Hydroxytryptamine (serotonin) uptake by male and female Schistosoma mansoni was studied in vitro. Initial uptake was determined during the first 2 min of incubation with the indoleamine. There were two types of uptake: a saturable component that is apparent at low concentrations of serotonin, and a passive component that predominates at high concentrations. Female S. mansoni take up more 5-hydroxytryptamine per milligram protein than do the males. Studies on the distribution of 5-hydroxytryptamine taken up by the parasites show that a large proportion of the indoleamine is in the tegument. The female tegument had a greater proportion of the 5-hydroxytryptamine taken up than did the male tegument. The evidence indicated that 5-hydroxytryptamine associated with tegument represented the passive uptake while uptake in the main parasite body was both active and passive. Compounds that were shown to inhibit 5-hydroxytryptamine uptake include methylclonazepam, metergoline, imipramine, and ouabain. Maximal inhibition by these compounds ranged from 50 to 60%. Inhibition appeared to be confined to the saturable uptake component of the body fraction.  相似文献

A 12 month survey was carried out of the spider population in a commercial citrus grove at Kibbutz Afeq in Northern Israel. The spiders collected from grapefruit foliage and ground cover were identified; young stages were reared to maturity and then identified.Chiracanthium mildei L. Koch represented 52% of all spiders captured during the year andTheridion sp. accounted for 34%. In the undercover,Gnaphosidae spiders represented 43% of the total spiders captured in pitfall traps andLycosidae 35%. A field experiment was carried out to evaluate the effectivness of spiders in biological control of the scaleCeroplastes floridensis Mask. in the citrus ecosystem. Wherever spiders were undisturbed on tree branches, populations ofC. floridensis were not able to develop to a level sufficient to cause economic damage. The increase in the number of scales was minimal: from 47 initially to 56. There was no damage to leaves nor was honeydew or sooty mold observed. During the same time period, on the 3 branches from which spiders had been eliminated, the number of scales increased seven-fold from 44 to 309. Leaves were heavily infested with sooty mold fungi that developed on the honeydew produced by the scales.
