Delta3(E)-unsaturated fatty acids are characteristic components of glycosylceramides from some fungi, including also human- and plant-pathogenic species. The function and genetic basis for this unsaturation is unknown. For Fusarium graminearum, which is pathogenic to grasses and cereals, we could show that the level of Delta3-unsaturation of glucosylceramide (GlcCer) was highest at low temperatures and decreased when the fungus was grown above 28 degrees C. With a bioinformatics approach, we identified a new family of polypeptides carrying the histidine box motifs characteristic for membrane-bound desaturases. One of the corresponding genes was functionally characterized as a sphingolipid-Delta3(E)-desaturase. Deletion of the candidate gene in F. graminearum resulted in loss of the Delta3(E)-double bond in the fatty acyl moiety of GlcCer. Heterologous expression of the corresponding cDNA from F. graminearum in the yeast Pichia pastoris led to the formation of Delta3(E)-unsaturated GlcCer. 相似文献

The aim of this numerical work is to analyze the effect of the position of the swimmer's head on the hydrodynamic performances in swimming. In this initial study, the problem was modeled as 2D and in steady hydrodynamic state. The geometry is generated by the CAD software CATIA and the numerical simulation is carried out by the use of the CFD Fluent code. The standard k-epsilon turbulence model is used with a specific wall law. Three positions of the head were studied, for a range of Reynolds numbers about 10(6). The obtained numerical results revealed that the position of the head had a noticeable effect on the hydrodynamic performances, strongly modifying the wake around the swimmer. The analysis of these results made it possible to propose an optimal position of the head of a swimmer in underwater swimming. 相似文献

Taguchi’s methods were used for the design of an experimental strategy aimed at optimizing cell density and monoclonal antibody
(mAb) production from a spinner flask hybridoma culture. 23G11 is an antibody to the human leukocyte adhesion molecule, CR3
or β 2 integrin (CD11b/CD18). It recognizes specifically the A-domain of the α subunit CD11b. Anti β 2 integrin monoclonal
antibodies hold a great potential for preventing inflammation mediated tissue injuries. An L8 orthogonal experimental design
was used to investigate four different culture components: stirring speed, nature of serum, concentration of serum and nature
of media (RPMI 1640 or RPMI 1640 supplemented with glucose and glutamine). The experiments were conducted using two levels
for each factor studied and a direct ELISA test was used to estimate the level of antibody production. Statistical analysis
of the collected data pointed to the stirring speed and serum concentration, and the interaction between these parameters,
as the components that affected cell growth. Antibody production was affected by these factors and by the nature of medium
but also by the following two interactions: stirring speed/nature of serum and stirring speed/concentration of serum. This
study emphasizes the value of using Taguchi’s methods as a basis for optimization of mAb production from a hybridoma culture,
in cost effective and significantly less labor intensive ways.

The optimum conditions for using the method of radioimmunoprecipitation (RIP) for the detection of human immunodeficiency virus (HIV) in serum samples have been established. Out of several available cell lines persistently infected with HIV, specially selected line 17 has been chosen. The characteristic feature of this is the high and stable (under the conditions of prolonged cultivation) accumulation of virus-specific proteins in infected cells. The optimum conditions for making the test and its evaluation have also been established. The data of literature on the advantages of the method of RIP over such traditional methods as the enzyme immunoassay and immunoblotting have been confirmed. Thus, the presence of specific antibodies in several serum samples registered as false negative has been established. The intertypical reactivity of two serotypes of the virus, HIV-1 and HIV-2, has been studied. Cross reactivity of antibodies with respect to the HIV gene gag, but not with respect to viral glycoproteids, has been established. Ideas on the expediency and prospects of using RIP for the serological control of HIV infection are presented. 相似文献

Toxin production and the intensity of toxin formation in 265 C. diphtheriae strains circulating in different areas of the USSR have been studied by the method of the enzyme-linked immunosorbent assay (ELISA). This study has been carried out with the use of the assay system consisting of monoclonal antibodies to the COOH-area of the B-fragment of the toxin molecule adsorbed onto the surface of polystyrene plates, affinity-purified polyclonal antidiphtheria antibodies labeled with horse-radish peroxidase and substrate indicator mixture (5-aminosalicylic acid and hydrogen peroxide). Some specific features of using ELISA for the detection of C. diphtheriae toxin directly in liquid culture medium are presented. High sensitivity, specificity and good reproducibility of this method permitting the detection of C. diphtheriae toxin and the determination of the intensity of toxin formation in the C. diphtheriae strains under study are shown. The dynamics of the formation of postvaccinal immunity after immunization with preparations obtained with the use of hydroxylamine (HA) preparations from Proteus strains of different O serogroups, Salmonella minnesota Re-mutant and the common antimicrobial antigen isolated from Escherichia coli 14 has been studied on mice with Proteus peritonitis-bacteremia used as a model. The study has revealed that intraperitoneal immunization with Proteus HA preparations stimulates the phagocytic activity of peritoneal mononuclear cells in mice and induces an increase in the titers of specific O antibodies. Proteus antigens ensure the formation of anti-Proteus immunity, preventing the death of the animals from peritonitis-bacteremia. The protection of mice from such infection resulting from the injection of the common antigens of gram-negative bacteria is considerably less. These data are indicative of the possibility of using Proteus peritonitis-bacteremia as a model for the study of the protective potency of Proteus vaccines. 相似文献

Studied has been the fine structure of the kidney's glomerulus endotheliocyte of man (age 2-6 years) and fish (year-old Atlantic salmon Salmo salar L.), as organisms occupying extreme place in development of kidney in phylogenetic line of the vertebrates. Revealed has been the uniformity of geometric reconstruction of vascular endothelium of kidney's glomerulus of investigated objects under heterogeneous factors' influence (compensatory hyperfunction of man and environment influence of fish). Formation of star-shaped structures with large open fenestrae in its central part (diameter 100-150 nm) both in the local parts of endothelium and in space between them (diameter 250-350 nm) has been observed. Such construction peculiarity sharply increases permeability of capillaries of kidney's glomerulus and thus appears as a characteristic sign of strenuous functional activity of the filtration apparatus of metanephros of man and mesonephros of fish. Revealed reaction of endothelium is considered as universal and nonspecific. 相似文献
