Crystallization of the DpnM methylase from the DpnII restriction system of Streptococcus pneumoniae 总被引:1,自引:0,他引:1
Three proteins, two DNA methylases and an endonuclease, from the DpnII restriction system of Streptococcus pneumoniae recognize the DNA sequence 5' GATC 3' but have very different amino acid sequences, which make them interesting subjects for structural determination. A purification procedure was developed that conveniently yields milligram amounts of the DpnM methylase. The DpnM protein tends to precipitate at reduced ionic strength, and this property was exploited to yield well-formed bipyramidal crystals. By X-ray diffraction, the crystals of DpnM were found to be orthorhombic, with cell dimensions a = 56.9 A, b = 68.2 A, c = 84.5 A; systematic absences identify the space group as P2(1)2(1)2(1). Diffraction extends beyond 3 A, so the crystals may allow structural determination at atomic resolution. 相似文献

Oreochromis aureus was imported from Israel into South Africa in 1959 but data on its current status in South Africa are lacking. Genomic DNA was extracted and the COI gene amplified at the South African Institute for Aquatic Biodiversity. The identity of the sequences and specimens was determined using the Barcode of Life Data Systems and GenBank. Morphological and genetic assessment demonstrated that 11 specimens collected from two farm dams in the Eerste River System, Western Cape province, were Oreochromis aureus. A MaxEnt model compiled using global distribution, rainfall and temperature data predicted that large areas of southern Africa were climatically suitable for this species, indicating considerable invasion debt in southern Africa. As a result, surveys to assess for the extent of the invasion in South Africa and eradication of existing populations, if feasible, are recommended management actions. 相似文献

Salinity intrusion in the coastal freshwater rivers due to climate change and construction of the dam in the upstream rivers are alarming in aquaculture. Hence, an experiment was conducted to know the effects of salinity on growth performance, hemato‐biochemical parameters and erythrocytes structure in a commercially cultivable catfish species, striped catfish (Pangasianodon hypophthalmus). Firstly, median lethal concentration (LC50) of salinity for striped catfish was determined and then the fish were exposed to three salinity conditions (4, 8 and 12‰) and a control (0‰). Fish were sacrificed at day 7, 14, 28 and 56 after the start of salinity exposure. The 96 hr LC50 value was found to be 14.87‰. Salinity levels from freshwater to 8‰ showed optimal conditions with high survival rate and good growth performances of fish in terms of weight gain and specific growth rate (SGR). Interestingly, the lowest food conversion ratio (FCR) was found in 4‰ group. The hemoglobin (Hb) level and number of red blood cells (RBCs) were found to be decreased significantly in 8 and 12‰ compared to 0 and 4‰ at the initial days of exposure, while number of white blood cells (WBCs) and glucose level showed opposite scenario. Frequencies of ENA (erythrocytic nuclear abnormalities) and ECA (erythrocytic cellular abnormalities) were significantly increased with increasing salinities in the initial days of exposure. Overall, findings of the present study revealed that striped catfish might be suitable fish species for culture in the brackish water containing salinity up to 10‰. 相似文献

Identifying an organism's migratory strategies and routes has important implications for conservation. For most species of European ducks, information on the general course of migration, revealed by ringing recoveries, is available, whereas tracking data on migratory movements are limited to the largest species. In the present paper, we report the results of a tracking study on 29 Eurasian Teals, the smallest European duck, captured during the wintering period at three Italian sites. The departure date of spring migration was determined for 21 individuals, and for 15 the entire spring migratory route was reconstructed. Most ducks departed from wintering grounds between mid‐February and March following straight and direct routes along the Black Sea‐Mediterranean flyway. The breeding sites, usually reached by May, were spread from central to north‐Eastern Europe to east of the Urals. The migratory speed was slow (approximately 36 km/day on average) because most birds stopped for several weeks at stopover sites, mainly in south‐eastern Europe, especially at the very beginning of migration. The active flight migration segments were covered at much higher speeds, up to 872 km/day. Stopover duration tended to be shorter when birds were closer to their breeding site. These results, based on the largest satellite tracking effort for this species, revealed for the first time the main features of the migratory strategies of individual Teals wintering in Europe, such as the migration timing and speed and stopover localization and duration. 相似文献

The F-spondin genes are a family of extracellular matrix molecules united
by two conserved domains, FS1 and FS2, at the amino terminus plus a
variable number of thrombospondin repeats at the carboxy terminus.
Currently, characterized members include a single gene in Drosophila and
multiple genes in vertebrates. The vertebrate genes are expressed in the
midline of the developing embryo, primarily in the floor plate of the
neural tube. To investigate the evolution of chordate F-spondin genes, I
have used the basal position in chordate phylogeny of the acraniate
amphioxus. A single F-spondin-related gene, named AmphiF-spondin, was
isolated from amphioxus. Based on molecular phylogenetics, AmphiF-spondin
is closely related to a particular subgroup of vertebrate F-spondin genes
that encode six thrombospondin repeats. However, unlike these genes,
expression of AmphiF-spondin is not confined to the midline but is found
through most of the central nervous system. Additionally, AmphiF-spondin
has lost three thrombospondin repeats and gained two fibronectin type III
repeats, one of which has strong identity to a fibronectin type III repeat
from Deleted in Colorectal Cancer (DCC). Taken together, these results
suggest a complex evolutionary history for chordate F-spondin genes that
includes (1) domain loss, (2) domain gain by tandem duplication and
divergence of existing domains, and (3) gain of heterologous domains by
exon shuffling.

Cloning, Expression, and Mapping of Ribonucleases H of Human and Mouse Related to Bacterial RNase HI
We identified two human sequences and one mouse sequence in the database of expressed sequence tags that are highly homologous to the N-terminal sequence of eukaryotic RNases H1. The cDNAs for humanRNASEH1and mouseRnaseh1were obtained, their nucleotide sequences determined, and the proteins expressed inEscherichia coliand partially purified. Both proteins have RNase H activityin vitroand they bind to dsRNA and RNA–DNA hybrids through the N-terminal conserved motif present in eukaryotic RNases H1. TheRNASEH1gene is expressed in all human tissues at similar levels, indicating that RNase H1 may be a housekeeping protein. The humanRNASEH1and mouseRnaseh1cDNAs were used to isolate BAC genomic clones that were used as probes for fluorescencein situhybridization. The human gene was localized to chromosome 17p11.2 and the mouse gene to a nonsyntenic region on chromosome 12A3. The chromosomal location and possible disease association of the humanRNASEH1gene are discussed. 相似文献
