The endodermally differentiated mouse embryonal carcinoma cell line M1536-B3 produces the basement membrane-associated glycoproteins entactin and GP-2 in cell culture. Immunological techniques coupled with light and electron microscopy were used to study the intracellular distribution and fate of these molecules. The two proteins were localized in cisternae of rough endoplasmic reticulum and in discrete membrane-bound vesicles. There was no evidence for their presence in the Golgi apparatus. The membrane-bound vesicles appeared to fuse with the plasma membrane and thereby transfer their contents to discrete foci on the cell surface. These foci became apparent as cell-to-cell contact was made and were prominent at cell-cell contact sites. They eventually coalesced into continuous densely stained extracellular bands of amorphous material. These bands formed a honeycomb of spaces lined by cells. It appeared that production of the extracellular matrix was probably stimulated by cell-cell contact, and that cell density rather than age of the cell cultures dictated the pattern of extracellular distribution. These results support the hypothesis that these molecules are involved in cell adhesion and multicellular organization.  相似文献

Breath analysis, including measurement of nitric oxide (NO), is a noninvasive diagnostic tool that may help evaluate cetacean health. This is the first report on the effects of breath hold duration, feeding, and lung disease on NO in dolphin exhaled breath. Three healthy dolphins were trained to hold their breath for 30, 60, 90, and 120 s and then exhale into an underwater funnel. Exhaled NO values from 157 breath samples were compared among three healthy dolphins by breath hold time and after fasting and feeding. Exhaled NO values were also measured in two dolphins with pulmonary disease. NO in dolphin breath was higher compared to ambient air; healthy dolphins had higher NO concentrations in their breath after feeding compared to after overnight fasting; and there were no significant differences in exhaled NO levels by breath hold duration. A dolphin with Mycoplasma‐associated pneumonia and chronic gastrointestinal disease had higher postprandial exhaled NO levels compared to healthy controls. This study demonstrates, contrary to previous publications, that dolphins exhale NO. Given the high standard deviations present in exhaled breath NO values, future studies are needed to further standardize collection methods or identify more reliable samples (e.g., blood).  相似文献

Sorting and maintenance of the EGF receptor on the basolateral surface of renal epithelial cells is perturbed in polycystic kidney disease and apical expression of receptors contributes to severity of disease. The goal of these studies was to understand the molecular basis for EGF receptor missorting using a well-established mouse model for the autosomal recessive form of the disease. We have discovered that multiple basolateral pathways mediate EGF receptor sorting in renal epithelial cells. The polycystic kidney disease allele in this model, Bicc1, interferes with one specific EGF receptor pathway without affecting overall cell polarity. Furthermore one of the pathways is regulated by a latent basolateral sorting signal that restores EGF receptor polarity in cystic renal epithelial cells via passage through a Rab11-positive subapical compartment. These studies give new insights to possible therapies to reconstitute EGF receptor polarity and function in order to curb disease progression. They also indicate for the first time that the Bicc1 gene that is defective in the mouse model used in these studies regulates cargo-specific protein sorting mediated by the epithelial cell specific clathrin adaptor AP-1B.  相似文献

Mockingbirds normally secrete little or no testosterone during the period of autumnal territoriality. To determine the behavioral effects of exogenously administered testosterone, 20-mm lengths of Silastic tubing filled with crystalline testosterone were implanted into free-living resident mockingbirds during the autumn. Control residents were given sealed empty implants. Focal animal sampling showed that T-implanted males sang significantly more than controls. Perhaps as a consequence, a significantly greater percentage of the T-implanted males acquired mates. Though nest building does not naturally occur in autumn, T-implanted males also showed significantly more nest building than control males. However, T-implanted males only built if there was a female in the territory, suggesting a synergy between the presence of testosterone and social cues provided by the female. Examination of the effects of testosterone on territorial aggression showed that despite the high levels of territorial activity common in this species in autumn, territorial fights were unaffected by the presence of testosterone. One aggressive call, known to function in fall territorial defense, was significantly decreased in T-implanted versus control males. The presence of fall testosterone appears to stimulate a number of reproductive activities in mockingbirds, leaving autumnal aggressive interactions either unchanged or decreased. We discuss the application of these data to the effects of testosterone on the mockingbird's reproductive behavior during the breeding season.  相似文献

Currently much debate surrounds the significance of cross-cultural variation in mother-infant attachment. Is only one form of attachment “healthy,” or are different types of attachment adaptations to local socioecological conditions? Juvenile mortality rates have been promoted as important features of local environments that shape attachment, which in turn affects later reproductive strategies. To this we add fertility. Fertility changes the environment of a child by influencing the number of potential caregivers and competitors for care, and the cultural ethos regarding the rights of children. Different combinations of fertility and mortality will likely give rise to different attachment forms, and only under one regime (low fertility and mortality) do we expect exclusivity in attachment.  相似文献

CuZn superoxide dismutase is a highly stable dimer of identical subunits with a combined molecular mass of 32,000 daltons. Two human superoxide dismutase genes have been joined in the same translational reading frame, using spacers of different lengths, to encode single chain proteins consisting of two identical human superoxide dismutase subunits. The first construct encodes two directly linked subunits; the terminal glutamine codon of the first gene was changed to a methionine codon and followed immediately by the second gene. The second construct encodes two subunits linked by a 19-amino-acid human immunoglobulin IgA1 hinge sequence. Both constructs produce high levels of catalytically active superoxide dismutase when expressed in Escherichia coli. The protein containing the IgA1 hinge sequence forms polymers up to 750,000 in molecular weight, which are linked together noncovalently by the hydrophobic bonding of the dimer interface. The polymers are soluble, thermostable, and of near normal specific activity. Site-directed in vitro mutagenesis was used to inactivate one of the two human superoxide dismutase subunits. The resulting human superoxide dismutase polymers have approximately 50% activity, thus confirming that the products of both genes are catalytically active. Large amounts of individual polymeric forms have been purified from recombinant yeast and tested for serum stability in rats. The serum half-life is approximately 7 min for both the two-chain wild type human superoxide dismutase dimer (Mr 32,000) and the single chain molecule consisting of a human superoxide dismutase dimer covalently linked by the immunoglobulin hinge region (Mr 34,000), whereas the higher molecular weight polymers (Mr greater than or equal to 68,000) all have half-lives of approximately 145 min.  相似文献
